Item Acetogenin as tool to control Aedes aegypti: a perspective of toxicity and gene regulation(Universidade Federal de Viçosa, 2016-08-12) Costa, Marilza da Silva; Serrão, José Eduardo; http://lattes.cnpq.br/9321013764019273Dengue, Chikungunya and Zika are arboviruses transmitted mainly by the mosquito Aedes aegypti. Although the control of this insect vector has been accomplished by eliminating rearing sites, use of biological control, genetic methods and the use of insecticides, it is important the use alternative control strategies for the integrated management of this pest. Thus it is expected decrease in the emergence of resistant mosquito strains and in the environmental risks and toxicology. This work evaluated a secondary metabolite of Annonaceae plant with insecticidal properties in the control of A. aegypti larvae. In the first step an acetogenin of Annona mucosa seeds was isolated and tested against A. aegypti larvae (target insect), its predators Culex bigotti and Toxorhynchites theobaldi (non-target insects) and on human leukocytes. The second step evaluated the morphology of the midgut cell in larvae exposed to acetogenin and the effect of this molecule in the expression of genes for autophagy (Atg1 and Atg8), V-H+-ATPase (membrane carrier protein) and aquaporin -4 (Aqp4) (water channel protein). The third step tested the effect of acetogenin on anal papilla of A. aegypti larvae, on the morphology expression of V-H+ -ATPase and Aqp4. The isolated molecule was squamocin that showed high toxicity to the A. aegypti larvae, but without toxicity to non-target insects and human leukocytes, indicating the selectivity of this molecule. In the midgut, squamocin showed cytotoxic effects in the digestive cells, increased expression of genes for autophagy (Atg1 and Atg8), decrease of V-ATPase and Aqp4 expression in LC20 as well as their inhibition in LC50, showing multiple modes of action in this organ. In anal papillae, squamocin caused morphological damages in the epithelium, decreased transcription levels of AaAQP4 and inhibited the AaV-H + -ATPase transcription, suggesting effect in the osmoregulation process of this organ. The modes of action of squamocin at morpho- physiological and molecular levels are described for the first time. Our results indicated that squamocin can be a prototype of natural products with potential to control A. aegypti larvae, contributing to the establishment of alternative strategies for the control of insect vectors with high efficiency and low environmental impact.Item Agroecosystem management strategies and trophic interactions among herbivores, pathogens and predators(Universidade Federal de Viçosa, 2016-05-28) Perez, André Lage; Venzon, Madelaine; http://lattes.cnpq.br/2630831726662116Reducing the environmental complexity and increasing biomass of cultivated plants offer favorable conditions for pest species in conventional systems. Among biological control strategies, the restoration of vegetation diversity in agroecosystems and the adoption of management strategies aiming to reduce impacts on natural enemy populations are practices that can be employed in different types of farming systems. Much of the conservative biological control efforts are focused on maintaining populations of parasitoids and predators. However, pathogens represent key mortality factors in the regulating pest populations in crops. The maintenance of indigenous plants aiming to increment populations of predators and parasitoids, may also promote the preservation of propagules of entomopathogenic fungi in the soil. In addition, the reduction of soil disturbance may reduce impacts on the entomopathogenic community in agroecosystems soil. Studiyng the ecological interactions between predators, pathogens and herbivores is a key factor in understanding the role of biological control of pests in agroecosystems. As a model of crop management practices and conservative biological control strategies we used chili pepper crops (Capsicum frutescens) and melon (Cucumis melo var cantalupensis) represented by small fields and high susceptibility to pests. Interactions between natural enemies were studied in microcosms in greenhouse. Therefore, the goal of this thesis was to evaluate the role of agro-ecosystems management practices as biological control strategies of pests in annual crops. Aim is also to study in detail the ecological interactions between natural enemies, herbivores and plants. From this framework we addressed the following hypothesis: a) Non-crop plants promote the modification of biotic and abiotic factors affecting virulence, occurrence and abundance of entomopathogenic fungi in the soil; b) the use no-tillage in annual crops performs the preservation of entomopathogens in the soil resulting in higher virulence and occurrence of microorganisms in agroecosystems; c) herbivores exposed to spores of entomopathogenic fungi reduce their herbivory on the host plant; d) the exposure of herbivore to pathogen spores affect predation; e) herbivores reduce and modulate feeding behavior in the presence of a predator; f) joint action of pathogen and predator results in a further reduction in the damage caused by herbivores to plants. As a result we found that the maintenance of non-crop plants in chili pepper crops is not an effective as a short-term strategy for preserving entomopathogenic fungi. The fungal virulence in two experimental fields with non- crop plants was higher than in chili pepper monoculture, but in one field we observed an inverse result for fungi virulence. The tillage regime in melon crops did not affect the virulence of soil-borne entomopathogens. The occurrence and virulence of entomopathogens was affected by sampling time during the growing season, showing a time-scale pattern of entomopathogenic dynamics in agroecosystems. In laboratory assay and in the greenhouse we observed that spiders do not show preferential predation for prey exposed to the pathogen. The exposure to spores of pathogen did not reduce the herbivory D. undecimpunctata on melon plants, however the presence of predator reduced the damage caused by the herbivore, both due predation and reduce time allocated by beetle on the plant. Also, we observed that herbivores exposed to pathogen and in the presence of predator damaged the plants in a similar rate compared to beetles in the absence of any natural enemy (predators or pathogen). In conclusion, from the results obtained in this thesis, it is concluded that the use of habitat management strategies should pay attention not only to the landscape composition and reduced disturbances but especially the time factor restore ecosystem services performed by entomopathogens. We also concluded that pathogens, predators, herbivores and plants have trophic interactions that differentially affect the dynamics of natural enemies and their ecosystem services.Item Alterações histopatológicas e citotóxicas no intestino médio de Apis mellifera (Hymenoptera: Apidae) causadas por acaricida e fungicida(Universidade Federal de Viçosa, 2020-02-17) Serra, Raissa Santana; Serrão, José Eduardo; http://lattes.cnpq.br/9748751800760712Polinizadores, incluindo abelhas melíferas, são responsáveis pela reprodução de mais de 87% das espécies de plantas angiospermas, sendo vitais para a saúde dos ecossistemas e dos serviços agrícolas ao redor do mundo. A abelha Apis mellifera tem seu desaparecimento ligado a utilização de pesticidas, que além da mortalidade direta causam efeitos subletais, tais como alteração na função de órgãos. O epitélio do intestino médio é responsável pela digestão e absorção de nutrientes, além da detoxificação de substâncias ingeridas, como inseticidas. O objetivo deste estudo foi avaliar as mudanças histológicas e citológicas do intestino médio de operárias de A. mellifera causadas pelo acaricida espiromesifeno e pelo fungicida azoxistrobina. O teste-limite foi conduzido de forma que se assegurasse DL50 acima de 100 μg/abelha para cada pesticida. Confirmada a não toxicidade dos compostos, a dose recomendada para campo foi administrada per os, dissecando-se o intestino após 24h e 48h de exposição para análises. O intestino médio do grupo controle apresentou uma única camada de células digestivas, com núcleos esféricos, ninhos de células regenerativas e lúmen revestido de matriz peritrófica. As abelhas tratadas com ambos os pesticidas apresentaram mudanças histológicas, como desestruturação do epitélio, vacuolização e fragmentos celulares liberados para o lúmen, e mudanças citológicas, como aumento no número e alteração de mitocôndrias, alteração do labirinto basal e desestruturação do retículo endoplasmático rugoso. A ocorrência de danos ao intestino médio em A. mellifera é uma indicação de estes pesticidas devem ser investigados para garantir que sua utilização seja segura para as abelhas. Toxicologia.Item Amostragem de Liriomyza trifolii (Burgess) (Diptera: Agromyzidae) no tomateiro(Universidade Federal de Viçosa, 2004-01-29) Gusmão, Marcos Rafael; Picanço, Marcelo Coutinho; http://lattes.cnpq.br/4032769085084783O manejo de mosca minadora, Liriomyza trifolii (Burgess) (Diptera: Agromyzidae), é uma medida importante como forma de evitar a mudança de status desta praga na cultura do tomateiro. Este trabalho objetivou determinar a unidade amostral que melhor representa a variabilidade de minas com larvas na planta e propor planos de amostragem convencionais e seqüenciais de contagem e de presença-ausência para L. trifolii no tomateiro. A unidade amostral ideal foi determinada com o registro do tempo gasto para a contagem de minas com larvas nas unidades amostrais compostas por uma, duas e três folhas consecutivas ao longo do dossel da planta. Foram calculadas as variâncias relativas das unidades amostrais e realizadas análises de correlação e de regressão linear simples entre o as densidades absolutas (no de minas/terço do dossel e n de minas/planta) e densidades relativas (no de minas/unidade amostral e no de minas/terço do dossel). A unidade amostral com uma folha inserida entre os terços mediano e basal do dossel do tomateiro foi a que melhor representou a variabilidade e o número total de minas de L. trifolii na planta de tomate. Os números de minas de L. trifolii/folha e de folhas infestadas com densidades acima de zero, uma, duas e três minas/folha foram utilizados para se determinar os planos de amostragem. Os dados amostrais foram ajustados ao modelo de distribuição binomial negativo e obtido parâmetro de dispersão comum (Kcomum ). As proporções de unidades amostrais infestadas com densidades acima de zero, uma, duas e três minas/folha foram determinadas com o modelo de probabilidade de distribuição binomial negativa. Os planos de amostragem foram obtidos com o valor de Kcomum igual a 0,835, nível de controle igual a 3,0 minas/folha e os valores das proporções de unidades amostrais infestadas. Os planos convencionais de contagem e de presença-ausência foram determinados aos níveis de erro de 5, 10, 15, 20 e 25%. Os planos de amostragem seqüencial de contagem e de presença-ausência foram determinados através do teste da razão de probabilidade seqüencial de Wald. Foram requeridas 24 e 96 amostras/talhão para a amostragem convencional ao nível de erro de 25% pela contagem do número de minas e avaliação da presença- ausência, respectivamente. Os planos de amostragem seqüencial, através da contagem de minas e da avaliação de presença-ausência foram consistentes, precisos e requereram o máximo de 15 e 27 amostras para tomada de decisão correta, respectivamente. O plano seqüencial de contagem proporcionou uma economia de 37,5% no tempo de amostragem em relação ao plano convencional. Verificaram- se economias de 93, 84, 75 e 66% no tempo de amostragem com os planos seqüenciais de presença-ausência em relação aos planos convencionais com densidades de infestação acima de zero, uma, duas e três minas/folha, respectivamente. Em lavouras com baixas infestações, recomenda-se os planos seqüenciais de presença-ausência com densidades de infestação da amostra acima de zero mina/folha.Item Amostragem e distribuição espaço-temporal de ácaros praga em pinhão manso(Universidade Federal de Viçosa, 2013-02-28) Rosado, Jander Fagundes; Picanço, Marcelo Coutinho; http://lattes.cnpq.br/5000438962823323Este trabalho teve como objetivos determinar planos de amostragens e a distribuição espaço-sazonal do ataque dos ácaros praga Polyphagotarsonemus latus e Tetranychus bastosi em pinhão manso. Para tanto, estes ácaros foram monitoradas em lavouras de pinhão manso em Caseara, Formoso do Araguaia e Palmas, TO de 2010 a 2012. O plano de amostragem determinado para T. bastosi é constituído da avaliação do número de ácaros. cm-2 em 102 amostras com lupa manual de 20 aumentos. O local ideal para amostragem deste ácaro foi à face abaxial da 4a folha mais apical do ramo localizado no terço mediano do dossel das plantas. Na face abaxial T. bastosi deve ser avaliado nas partes laterais da porção mediana da folha próximo à sua borda. Já o plano de amostragem para P. latus, a avaliação do número de ácaros. cm-2 será em 135 amostras com lupa manual de 20 aumentos. O local ideal para amostragem deste ácaro foi à face abaxial da 4a folha mais apical do ramo localizado no terço apical do dossel das plantas. Na face abaxial P. latus deve ser avaliado nas partes laterais à inserção do pecíolo da folha. Os maiores ataques de P. latus e T. bastosi ocorreram no outono de 2012 e primavera de 2011, respectivamente. As maiores densidades de P. latus ocorreram em períodos chuvosos e de menor fotoperíodo e quando foram maiores as populações de aranhas predadoras. Já as maiores densidades de T. bastosi ocorreram em períodos secos, quentes, de maior fotoperíodo, de menor ocorrência de ventos e quando foram maiores as populações do predador Stethorus sp. O ataque de P. latus e T. bastosi foi agregado no centro das plantações e a dispersão dos ácaros ocorreu de forma rápida em todas as direções das lavouras. O padrão de distribuição espacial do ataque de P. latus e T. bastosi numa lavoura teve pouca variação de um ano para outro. As densidades dos ácaros estiveram correlacionadas (alcance) a uma distância de 7,37 a 45m.Item Análise cladística e biogeográfica de Herdoniini (Heteroptera: Miridae)(Universidade Federal de Viçosa, 2000-05-29) Paula, Alexandre Silva de; Ferreira, Paulo Sérgio Fiuza; http://lattes.cnpq.br/2193708427094253A análise cladística e os dados de distribuição geográfica deste trabalho foram baseados no exame da informação presente na literatura e nos espécimes adultos da tribo Herdoniini pertencentes a 38 gêneros previamente considerados. A análise cladística dos gêneros foi efetuada com a utilização conjunta dos programas Phylogenetic Analysis Using Parsimony (PAUP) e MacClade. Os resultados desta pesquisa demonstram que Herdoniini forma um grupo polifilético. A reclassificação da tribo foi proposta a partir de sinapomorfias encontradas na análise, assim como a diagnose da tribo e dos gêneros americanos. Os caracteres da genitália dos machos revelaram sinapomorfias que suportam o monofiletismo de 18 gêneros americanos. O cladograma de taxon–área, derivado da análise filogenética, propõe o monofiletismo destes gêneros com origem na América do Sul e demonstra eventos biogeográficos relacionados com padrões de vicariância e dispersão nas américas. Os dados biológicos de literatura continham pouca informação para inferir as características biológicas e associações com espécies de formigas para a explicação do habito mirmecomórfico comum aos gêneros de Herdoniini.Item Aspectos neuroendocrinos e comportamentais do desenvolvimento da formiga Camponotus (Myrmothryx) rufipes Fabricius, 1775 (Hymenoptera, Formicidae, Formicinae)(Universidade Federal de Viçosa, 2004-07-31) Soares, Paula Andréa Oliveira; Serrão, José Eduardo; http://lattes.cnpq.br/1439917455162632O desenvolvimento pós-embrionário dos insetos apresenta uma reorganização profunda no sistema nervoso, como uma adequação à futura vida adulta. Porém, a plasticidade do sistema nervoso também ocorre durante a vida adulta devido a plasticidade comportamental, idade, sexo e do status endócrino. Neste estudo, foram avaliadas as mudanças estruturais sofridas no cérebro em representantes das castas reprodutiva e operária de Camponotus rufipes, com ênfase nas operárias polimórficas, e confrontadas com a titulação de hormônios morfogenéticos e seu repertório comportamental, visando possíveis relações entre ambos. Os resultados confirmaram a ocorrência de plasticidade neural pós- embrionária, bem como adulta. No entanto, a influência hormonal e comportamental neste fenômeno apresentou perfis distintos principalmente nas subcastas de operárias, sobretudo nas operárias minor. Operárias minor com três dias de vida adulta apresentaram altos títulos de hormônio juvenil e volume total do cérebro, mas operárias com idades a partir dos sete dias de vida, apresentaram valores decrescentes do volume do cérebro embora com repertório comportamental significante. No entanto, os altos títulos observados nas operárias major de 45 dias e a ausência de certos atos comportamentais não apresentaram qualquer relação com o volume do cérebro, trazendo dúvidas sobre a influência destes na plasticidade cerebral. É possível que as adaptações hormonais e comportamentais sejam cíclicas, visto que o volume total do cérebro das operárias menores apresentou platôs em momentos distintos da vida adulta, possivelmente em resposta a fatores que promovam aprendizagem e memória. O volume da neurópila das subcastas de operárias parece ser correlacionado à idade, sugerindo o aumento de ramificações neuronais e possível atividade sináptica, em resposta à adição de informações processadas ao longo do tempo.Item Associative learning in immatures and adults of generealist predators(Universidade Federal de Viçosa, 2016-09-20) Oliveira, Cleber Macedo de; Pallini Filho, Angelo; http://lattes.cnpq.br/5062021577770689Plants produce organic volatiles continuously, however this blend of volatiles changes when they are infested by herbivores. These odours can be used by beneficial arthropods to locate prey. However, the blend of volatiles emitted by plants can vary with changing biotic and abiotic conditions. Due to variability of odours, it is expected that predators learn the association of odours and availability of food. Learning occurs when the response to an odour change due to an experience. Learning was demonstrated in many species of arthropods, however those studies assessed learning in adults. Those studies evaluated learning in the laboratory conditions and did not study the effect of the conditioned stimulus (i.e. odours) in the learning. Here, we aimed to study learning in immatures of the generalist predator Ceraeochrysa cubana and adults of Orius insidiosus. We assessed the effect of odours in the learning in two generalist predators and evaluated the effect of learning in the foraging behaviour of immature predators. We assessed the response of the predators to odours in the olfactometer and in a release-recapture experiment. The odours studied were methyl salicylate, rosemary oil liquid (Rosmarinus officinnalis) and mint oil. Immatures of lacewings learned the association of methyl salicylate with availability and unavailability of food. This was the first study that assessed the learning in immatures predators. Learning changed the foraging behaviour of immature predators. Immatures of lacewings were able to learn two association with the same odour, association of volatile with food and after association of volatile without food, during the immature phase. Additionally, our results showed that the learning ability of O. insidiosus is dependent of the conditioned stimulus (odours).Item Atividade formicida de IB 151239 em Atta sexdens rubropilosa(Universidade Federal de Viçosa, 2011-11-04) Ribeiro, Myriam Marques Ramos; Della Lucia, Terezinha Maria Castro; http://lattes.cnpq.br/9067587442716563Dentre as pragas de importância no Brasil, as formigas cortadeiras se destacam por causarem danos consideráveis nos eucaliptais e em diversos cultivos. Atualmente, o seu controle em larga escala utiliza ingredientes tóxicos como o fipronil, a sulfluramida e o clorpirifós, que eventualmente deverão ser proibidos frente à pressão exercida pelas certificadoras estrangeiras dos produtos florestais brasileiros. Assim, os objetivos deste estudo foram: avaliar o efeito de folhas de IB 151239 em colônias da formiga cortadeira Atta sexdens rubropilosa, bem como seu efeito subletal nas operárias; avaliar o efeito fungicida dos extratos etanólico e hexânico de folhas de IB 151239 sobre o fungo simbionte das cortadeiras, Leucoagaricus gongylophorus, e identificar as substâncias causadoras desse efeito presentes no extrato. Para avaliar a ação de folhas de IB 151239 nas colônias, os estudos foram realizados em formigueiros de laboratório, no Insetário do Departamento de Biologia Animal da Universidade Federal de Viçosa em salas sob a temperatura de 25 ± 5 oC, 75 ± 5 % U.R. e 12:12 (L:E). Folhas desse vegetal eram oferecidas diariamente às colônias. Os efeitos subletais eram avaliados, retirando-se operárias para medição da taxa de encapsulação, contagem do número de hemócitos totais presentes na hemolinfa e medição da taxa respiratória. Além disso, eram verificados o consumo foliar e o volume do jardim de fungo dessas colônias. Para avaliar a atividade fungicida de IB 151239, extratos hexânico e etanólico foram acrescidos ao meio de cultura do L. gongylophorus. Após a detecção do extrato mais ativo foi feito o fracionamento do mesmo e posterior identificação das possíveis substâncias causadoras do efeito fungicida. De acordo com os resultados encontrados, folhas de IB 151239 inibem o sistema imune de operárias de A. sexdens rubropilosa, uma vez que, em colônias tratadas com esse vegetal, as operárias apresentaram menor taxa de encapsulação e diminuição na quantidade total de hemócitos. Além disso, após oferecimento desse vegetal às colônias, houve diminuição do consumo foliar e do volume do jardim de fungo. Os extratos etanólico e hexânico de IB 151239 provocaram efeito fungicida no simbionte mutualista das cortadeiras, sendo que a fração clorofórmica do extrato etanólico demonstrou a maior atividade. O sub-fracionamento indicou possíveis substâncias fungicidas; dentre elas, o liral/intermedeol e avocadinofurano/(E)-nerolidil isobutirato/difenil-disulfide que necessitam ser confirmadas e testadas biologicamente.Item Bees increase tomato crop production and are impacted by both landscape context and farm management(Universidade Federal de Viçosa, 2018-07-25) Barbosa, Fernando Mendes; Campos, Lucio Antônio de Oliveira; http://lattes.cnpq.br/3086699951766083Most agricultural crops depend on animal-mediated pollination to some degree. Among insects, bees are the main pollinators, increasing yield and improving the quality of the agricultural production. On the other hand, the changes resulting from the intensification of the agriculture are pointed out as the main causes of the loss of diversity in pollinator communities, which may compromise the stability of food production. Given this, understanding how bees affect agricultural production and how changes resulting from intensification of agriculture impact the community of these pollinators is important to outline strategies that promote the increase and stability of the pollination services in agroecosystems. Thus, initially, I identified bees visiting the tomato flowers and the main characteristics of the identified species; I verified the effect of climatic variables and the period of day in which bees’ activity took place in the tomato flower; and I evaluated fruit production under different types of pollination. Subsequently, I evaluated how the landscape configuration (landscape scale) and farm management (local scale) affect the diversity of bees in small tomato crops in the region of Zona Mata, Minas Gerais state, Brazil. Finally, I assessed the effect of the irrigation method on pollination and pollinators in a tomato crop. Overall, bees increase tomato production and respond negatively to loss of natural habitat and increased use of agrochemicals. In this study, an important step was taken in understanding how bees affect tomato production and how the community of these pollinators responds to changes resulting from the intensification of agriculture. Considering the current discussion involving expansion of agricultural activity and biodiversity conservation, the results of this study are particularly relevant for providing important data that can guide pollinator management plans.Item Behavioral manipulation of spiders by parasitoid wasps: natural history and mechanism of action(Universidade Federal de Viçosa, 2015-12-02) Kloss, hiago Gechel; Sperber, Carlos Frankl; http://lattes.cnpq.br/6213884442511218Host-parasite and host-parasitoid relationships are common in nature and present interesting systems for gaining understanding of evolutionary mechanisms. One of the central topics in the ecology of parasitic organisms is transmission between hosts. Parasites often have specific adaptations that increase the likelihood of transmission. One of the most interesting strategies is behavioral manipulation, in which the parasite or parasitoid increases the probability of transmission by changing host behavior. One well-studied example of behavioral manipulation is that of parasitoid wasps in the Polysphinctine clade (Insecta: Hymenoptera), which manipulate spider behavior. The wasp stings a spider causing temporary paralysis, then deposit a single egg on abdomen. After a short period of time the egg then hatches into a larva that feeds on spider hemolymph. During the final stage of parasitoid larval development, the spiders modify web architecture, then the larva kills the spider and pupates in the center of the modified web. The origin and evolution of these interactions are not well understood. This thesis work investigates the natural history of two parasitoid wasp and two spider species, focusing on the adaptive value of behavioral modifications in terms of parasitoid survival and the mechanisms used by parasitoid wasps to induce spider behavioral changes. I used as model systems Cyclosa fililineata and Cyclosa morretes (Araneae: Araneidae), which are parasitized by Polysphincta sp. nr. purcelli and Polysphincta janzeni (Hymenoptera: Ichneumonidae), respectively. In the first chapter, I describe the attack behavior and life cycle of Polysphincta sp. nr. purcelli and Polysphincta janzeni. I assert that this behavior is facilitated by biological characteristics of the host, as has been observed for other species. In the second chapter, I use a manipulative experiment to show that web modification by C. fililineata and C. morretes enhances parasitoid survival, indicating adaptive value of this behavior for Polysphinctine wasps. I also present results from a second manipulative experiment evaluating whether changes in web architecture are caused by reduced nutritional status of spiders (i.e., due to parasite exploitation of host resources). I demonstrated that behavioral modification of parasitized spiders is not the result of food restriction, which I interpret as evidence of a chemical mechanism for spider behavioral changes. In the third chapter, I investigated possible chemical mechanisms and found high levels of ecdysone in parasitized spiders with altered behavior, but not in either parasitized spiders with unaltered behavior or in nonparasitized spiders. Thus, behavioral modification in spiders may occur through activation of mechanisms controlling ecdysis, resulting in construction of modified webs and increased parasitoid survival.Item Benefícios da alimentação em plantas de Eucalyptus cloeziana e Psidium guajava em campo para o predador Brontocoris tabidus (Heteroptera: Pentatomidae)(Universidade Federal de Viçosa, 2005-11-30) Medeiros, Rômulo Sátiro de; Zanuncio, José Cola; http://lattes.cnpq.br/7261796176825148O objetivo desse trabalho foi estudar o desempenho reprodutivo de Brontocoris tabidus (Signoret) (Heteroptera: Pentatomidae) com e sem plantas de Eucalyptus cloeziana ou Psidium guajava, tendo como presa pupas de Tenebrio molitor Linnaeus (Coleoptera: Tenebrionidae) em campo, a 21,16 ± 1,52°C, umidade relativa de 77,88 ± 0,98% e fotofase de 11,61 ± 0,26 horas, e avaliar o efeito da duração da fase ninfal e do peso na fecundidade e longevidade desse predador. A adição de material vegetal à dieta de B. tabidus aumentou a fecundidade, que foi 3,5 e 2,9 vezes maior, nos tratamentos com plantas de E. cloeziana e P. guajava, respectivamente, que naquele sem planta. A longevidade de B. tabidus em plantas de E. cloeziana e P. guajava foi 1,9 e 1,5 vezes maior, respectivamente, que no tratamento sem planta. O número de ovos e a longevidade de B. tabidus foram, respectivamente, 2,2 e 1,5 vezes maiores para fêmeas cuja fase ninfal foi curta e alimentadas com plantas de E. cloeziana. No entanto, a duração da fase ninfal não afetou as variáveis reprodutivas de B. tabidus quando alimentado, apenas, com pupas de T. molitor. A taxa líquida (Ro) de reprodução de B. tabidus variou de 9,7 (sem planta, fase ninfal curta) a 70,4 (com E. cloeziana, fase ninfal curta) fêmeas/fêmea. A duração de uma geração (DG) variou de 41,4 (sem planta, fase ninfal longa) a 62,1 dias (com E. cloeziana, fase ninfal curta) indicando que B. tabidus pode ter 8,8 e 5,9 gerações por ano, respectivamente. O tempo necessário para esse predador dobrar sua população variou de 10,1 (com E. cloeziana, fase ninfal curta) a 13,3 (sem planta, fase ninfal curta) dias. A taxa intrínseca de crescimento populacional (r m) mostrou potencial biótico de B. tabidus de 0,052 (sem planta, fase ninfal curta) e 0,069 (com E. cloeziana, fase ninfal curta). A rm de B. tabidus, proveniente de indivíduos de fase ninfal curta e alimentado em plantas de E. cloeziana foi 1,1 vezes maior que aquela proveniente de indivíduos de fase ninfal longa, e 1,3 vezes maior quando os indivíduos apresentaram fase ninfal curta e alimentados, apenas, com pupas de T. molitor. Fêmeas de B. tabidus mais pesadas e alimentadas com E. cloeziana depositaram 2,3 vezes mais ovos que aquelas leves submetidas à mesma dieta, e 3,7 e 8,4 vezes mais que fêmeas pesadas e leves alimentadas, apenas, com pupas de T. molitor, respectivamente. A longevidade de fêmeas mais pesadas de B. tabidus alimentadas com E. cloeziana foi 1,6 vezes maior que fêmeas leves do mesmo tratamento e 2,6 vezes maior que fêmeas leves e alimentadas, apenas, com pupas de T. molitor. As variáveis reprodutivas, a taxa líquida de reprodução e a taxa intrínseca de crescimento populacional de B. tabidus indicam que o desempenho reprodutivo desse predador é melhor para fêmeas pesadas, provenientes de fase ninfal curta e alimentadas com E. cloeziana. B. tabidus apresenta potencial para ser criado em condições naturais em plantas de E. cloeziana e P. guajava com pupas de T. molitor. Esta metodologia permite obter indivíduos com melhor desempenho reprodutivo. O controle químico é a principal estratégia de controle desta praga, podendo afetar os inimigos naturais, o controle biológico é uma alternativa, reduzindo os impactos negativos nos inimigos naturais. Podisus nigrispinus (Dallas) (Heteroptera: Pentatomidae), inimigo natural é utilizado em programas de controle biológico. Os óleos essenciais podem substituir os produtos químicos causando menor impacto ambiental e, facilitando a transição da agricultura convencional para a orgânica. O objetivo foi avaliar a atividade inseticida do óleo essencial de capim-limão (Cymbopogon citratus) (Poaceae) e seus constituintes contra lagartas de A. gemmatalis e o predador P. nigrispinus. Foi realizado cromatografia para análise dos constituintes do óleo essencial de capim-limão. Seis concentrações de óleo essencial de capim- limão e de seus constituintes foram aplicados no tórax das lagartas de A. gemmatalis e ninfas de P. nigrispinus. Avaliou se o comportamento locomotor das lagartas e ninfas com sistema de vídeo rastreado e a taxa respiratória, com os constituintes do óleo essencial nas doses DL 50 e DL 90 , com respirometro. A toxicidade do óleo essencial foi crescente entre os instares de lagartas de A. gemmatalis e ninfas de P. nigrispinus. Os principais constituintes desse óleo incluem neral (31,5%), citral (26,1%) e acetato de geranila (2,27%). Os ensaios de dose resposta, com os constituintes mostraram ser o acetato de geranila mais tóxico que o citral em A. gemmatalis e o inverso para P. nigrispinus. As taxas de respiração (μL de CO 2 h -1 /inseto) de A. gemmatalis e P. nigrispinus com o acetato de geranila e citral, com doses estimadas para os valores de DL 50 e DL 90 , diferiram. Lagartas e ninfas reduziram a mobilidade após a exposição aos constituintes do óleo em superfícies tratadas apresentando repelência e/ou hiperatividade. Óleos essenciais e seus constituintes são eficientes no controle de pragas, porém os consórcios com a utilização de inimigos naturais devem ser realizados com cautela.Item Bioecologia, dano e controle de Planococcus citri (Risso) (Hemiptera: Pseudococcidae) em Coffea canephora Pierre ex Froehner (Rubiaceae)(Universidade Federal de Viçosa, 2016-02-24) Fornazier, Maurício José; Zanuncio, José Cola; http://lattes.cnpq.br/1696597030678586Café é uma das principais commodities agrícolas brasileiras exportadas, possui forte aspecto social, distribui renda e possibilita qualidade de vida no meio rural. Café Arábica (Coffea arabica L.) é o mais cultivado no mundo e o café Robusta (Coffea canephora Pierre ex Froehner) representa cerca de 45% da produção mundial e 30% da produção brasileira. Insetos, incluindo Planococcus citri (Risso) (Hemiptera: Pseudococcidae) têm sido relatados reduzindo a produtividade do café no Brasil. Essa cochonilha é praga polífaga, causa dano a diversas culturas e em café Robusta tem mostrado rápido crescimento populacional. O objetivo dessa tese foi contribuir para o conhecimento da dinâmica populacional de P. citri, seus inimigos naturais, dispersão no cafeeiro, disseminação na maior região produtora brasileira desse café, danos à produtividade e possibilidade de seu controle em condições de campo. Os resultados foram organizados em três artigos. O primeiro artigo constatou que P. citri é espécie de cochonilha dominante em café Robusta, com baixa população de inimigos naturais, disseminada de 14-714 m snm e associada à altitude, temperatura e precipitação. Flores e frutos do café permitiram seu desenvolvimento na primavera-verão e plantas associadas ao cultivo do café podem ser refúgio. O segundo artigo relata danos diretos à produtividade entre 25,2% a 46,4% e seu controle via foliar, no tempo correto, proporcionou acréscimo médio de 1.360,82 kg.ha^-1 .ano^-1. Rosetas infestadas se correlacionaram positivamente com cochonilhas vivas e a produtividade negativamente com rosetas infestadas e cochonilhas vivas. O terceiro artigo mostrou três níveis de infestação natural e dano superior a 60% na produtividade. Neonicotinoides reduziram a população de P. citri em dose única aplicados de julho a setembro, sem reaplicação e seu uso tardio permitiu maior dispersão de P. citri nas plantas de café Robusta. Esses produtos podem ser usados em programas de manejo integrado de pragas nesse café.Item Bioeconomia para manejo racional de Spodoptera frugiperda em milho: relação injúria-produtividade e níveis de dano econômico em diversos cenários(Universidade Federal de Viçosa, 2023-05-12) Souza, Thadeu Carlos de; Pereira, Eliseu José Guedes; http://lattes.cnpq.br/5861213318935614A lagarta do cartucho do milho, Spodoptera frugiperda (Lepidoptera: Noctuidae) é considerada muito destrutiva em cultivos de milho, porém se desconhece a relação numérica da intensidade da sua injúria foliar com a produtividade de milho em condições representativas dos cultivos comerciais. Neste trabalho, determinamos curvas de dano e níveis de dano econômico (NDE) para S. frugiperda como desfolhadora em cultivos de milho grão e milho silagem. O trabalho foi conduzido nas safras de 2020/21 (1a safra, outubro–fevereiro) e 2021/22 (2a safra, março–julho), em quatro municípios de Minas Gerais, Brasil. Nos cultivos de milho, rotularam-se plantas com diferentes intensidades de injúria foliar pela lagarta do cartucho baseando-se na escala Davis. No ponto adequado para colheita, contabilizou-se a produção de cada planta rotulada, em silagem ou grãos. Com esses valores, estimou-se a porcentagem de produtividade da planta em relação à das plantas sem injúria foliar da lagarta do cartucho. Na análise de regressão para cada safra e finalidade da produção, equações do modelo logístico de três parâmetros se ajustaram adequadamente aos dados de produtividade em função da intensidade de injúria foliar. A máxima perda de produtividade foi 20-30% na maior intensidade de injúria (valor 9 na escala Davis, plantas com o cartucho destruído). As plantas tiveram nula redução na produtividade quando submetidas a baixas intensidades da desfolha pela lagarta do cartucho (valores 1–5 na escala Davis, i.e., até várias lesões com mais de 2,5 cm de comprimento nas folhas do cartucho). Esse padrão de tolerância à desfolha ocorreu nas plantas das duas épocas de cultivo. Na 1a safra, a curva de dano de grãos foi mais inclinada (7,3%) do que a de silagem (4,5%), mas na 2a safra declividade das curvas para grãos e silagem com semelhante (6,5 vs. 6,9%). O custo de controle do inseto em milho grão foi em média 4,3 e 2,0% do valor da produção em médio e alto grau de investimento, respectivamente. Para silagem, o custo de controle foi de 3,6 e 1,7% para na 1a safra. Na 2a safra, esse custo representou 3,6 e 1,7% (grão) e 1,4 e 1,0% (silagem) para médio e alto investimento, respectivamente. O menor valor de NDE em intensidade de injúria na escala Davis para milho grão na 1a safra foi de 8,0 e 7,1 para médio e alto grau de investimento. Para milho silagem esses valores foram de 5,9 e 5,4. Na 2a safra, para milho grão com alto ou médio investimentos, os valores de NDE foram de 6,1 e 5,5 e, para silagem, de 5,8 e 5,5, respectivamente. Esses menores valores foram obtidos no cenário de duas aplicações manuais por hectare. Valores fixo de NC podem ser tomados como 75% desses valores de NDE. Esta pesquisa é pioneira em fornecer informações realísticas sobre NDE para milho grão e silagem e elas podem ser utilizadas por produtores de milho e outros agentes em diversas regiões no mundo. Coletivamente, os resultados mostram que a situação de menor nível dano econômico da lagarta do cartucho em milho ocorre quando 100% das plantas apresentam injúria de nota 4 na escala Davis (i.e., NDE = 4). O atual valor de NC recomendado (20% de plantas com injúria foliar Davis nota 3) é, no mínimo, cinco vezes menor que o valor de NDE aqui obtido, sendo assim bastante conservativo como nível de ação para controle de S. frugiperda em cultivos de milho. The subfamily has been recently target of evolutionary and ecological studies once they are the only crickets known to use high-frequency calling songs. Two groups of Eneopterinae are showing this innovation in the communication system: Lebinthini distributed in the Pacific region and Eneopterini from the Neotropics. Specifically, this thesis aims to give answers about the biogeography and evolution of acoustic communication of Eneopterinae crickets in South America. We also explore trough different approaches the complex taxonomy of this group. In the first chapter we used a dated phylogeny calibrated with fossils and biogeographical analyses to estimate the origin, how many times Eneopterini colonized South America and the routes toward and from this continent. In the second chapter, we studied the bioacoustics of the Neotropical genera of Eneopterinae crickets. In the third chapter we used DNA and morphology-based approaches to explore species diversity in the cryptic Eneopterini genus Ligypterus. And finally, in the fourth chapter we described a new genus of Eneopterinae, Gnominthus, focusing on general morphology, bioacoustical analyses of the calling song and the description of the mating behavior. This study advances the understanding of the global biogeographical processes that shaped current distribution patterns. Our dating results show that the subfamily is a Gondwanan group, far older than expected and that its diversification dates back to Late Creataceous (ca. 76 Ma). In this context the colonization of the Neoptropics would have occurred twice independently, very early from an Antarctic origin, resulting from the break-up of a Gondwanan fauna, and later by a northern recolonization coming from South-east Asia, likely related with a Holarctic Boreotropical distribution of the species during the Eocene. Our results indicate a boreotropical migration for Eneopterinae, with interesting patterns of recolonization of South America through the Northern hemisphere. Boreotropical migration, is well-studied in plants, for which it is an important explanation for the biogeography, but is not very explored in animals. In the second chapter, we clarified the evolution of high-frequency songs in Neotropical Eneopterinae crickets, showing a scenario of two independent origins for this feature. In the third chapter, the species discovery methods indicated a considerably higher diversity in the cryptic genus Ligypterus, with probably twice more species than we currently know. And finaly in the fourth chapter we described an endemic Eneopterinae genus from Papua New Guinea with a distinct reproductive behavior and high-frequency calling songs. We believe that the perspectives now open by discussion on this thesis have the potential to offer base for further studies regarding this subfamily and crickets in general. The exploration of the use high-frequency communication in other cricket groups is urgent, given the importance of this feature in the evolution and ecological processes in Eneopterinae. Foi analisado os efeitos da maturação sexual para a as populações resistentes e suscetíveis da espécie E. connexa. Também se realizou uma descrição detalhada da anatomia e histologia do sistema reprodutor masculino e feminino de E. connexa. Finalmente, se realizou uma descrição anatómica do sistema reprodutor da espécie nativa Coleomegilla maculata e a invasora Harmonia axyridis, com o fim de estabelecer semelhanças e diferenças entre e obter informação que contribui a explicar a sucesso adaptativo de H. axyridis na América. Os resultados sugerem que as populações resistentes de E. connexa possuem maturação sexual precoce e potencialmente maior sucesso reprodutivo. Para esta espécie também foi descrita a anatomia do sistema reprodutor masculino o qual apresenta glândulas assessórias como ocorre nos besouros em geral. Ainda, a média de 15 ovaríolos por ovário, bem maior do que foi descrita para esta espécie. O maior número de folículos e ovariolos de H. axyridis, e a presença de glândulas no final do oviducto comum, indicaria uma vantagem na capacidade de colonizar ambientes e seu alto desempenho reprodutivo. However, insect and mite pests have significantly constrained such agricultural production as these arthropods indirectly reduce yields or quality of the production. The control of such pests still is heavily dependent of synthetic compounds, which has threatened the environment and human health. Two relevant agricultural systems in the Neotropical region is the beans (e.g., Phaseolus vulgaris e Vigna unguiculata) and the coconut production, Cocus nucifera L., (and storage for the case of bean grains). Thus, in the present dissertation are described the investigations conducted aiming to provide novel biorational approaches for controlling insect pests that attack stored bean grains (i.e., the bruchid beetles Acanthoscelides obtectus (Say), Zabrotes subfasciatus (Boheman) and Callosobruchus maculatus (Fabricius)) as well as the red palm mite (i.e., Raoiella indica (Hirst)) that has caused severe losses in the coconut production in the Neotropical region. Firstly, the present dissertation dealt with the competitive abilities of bean weevils in different hosts. The next investigations were conducted aiming to evaluate the potential of essential oil extracted from clove (Syzygium aromaticum) and cinnamon (Cinnamomun zeylanicum) for controlling these bean weevils (including potential side-effects associated with sublethal exposures), and were compared with a commercial product Deltamethrin (Decis- 25 EC; 25 g a.i./L). Finally, experiments were conducted aiming to evaluate the potential of the lacewing Ceraeochrysa caligata (Banks) as biological agent for controlling the red palm mite R. indica. The results for the competition bioassays revealed that C. maculatus exhibits the best competitive abilities on cowpea beans, independently of its interspecific competitors (i.e., A. obtectus or Z. subfasciatus); although the weevil Z. subfasciatus also show hormetic responses in presence of moderate densities of the competitor C. maculatus on this host. On common bean grains the weevil C. maculatus can’t produce offspring. The toxicological bioassays with essential oils revealed that A. obtectus exhibited equally susceptibility to clove (LD50 = 43.6 (38.9-49.3) µL of essential oil/kg bean) and cinnamon (LD50 = 46.8 (42.3-51.4) µL of essential oil/kg bean) essential oils. Furthermore, there was a trade-off between reproductive fitness and susceptibility to insecticides accordingly to the host where A. obtectus was being reared. Clove and cinnamon essential oils also exhibited insecticidal activities against C. maculatus similar to the synthetic insecticide Deltamethrin (Clove: LD50 = 78.2 (71.6-84.8)µL of essential oil/kg bean; cinnamon: LD50 = 131 (124.0- 137.0)µL of essential oil/kg bean; deltamethrin: LD50 = 83.7 (80.6-86.6)µL of a.i/ kg−1 bean). Females of C. maculatus exposed to sublethal doses of clove (LD20 = 48.6µL−1 ) and cinnamon (LD20 = 106.2µL−1 ) significantly reduced oviposition. In the last part of this dissertation, the results revealed than the predatory behavior of C. caligata feeding upon R. indica is dependent of the larval development of the predator and prey size. It is clearly demonstrated that C. caligata (especially at the second-instar stage) has potential as a tool for the ecological management of the red palm mite. The results reveal that essential oils of clove and cinnamon, and the natural enemy C. caligata are alternatives for the integrated management of stored grain pests and the red palm mite respectively.Item Bt toxin in the soybean looper: susceptibility, fitness & selection(Universidade Federal de Viçosa, 2015-07-28) Silva, Nilson Rodrigues da; Pereira, Eliseu José Guedes; http://lattes.cnpq.br/8506992351582777The soybean looper, Chrysodeixis includens, and the cotton bollworm, Heliothis virescens, (Lepidoptera: Noctuidae), cause significant losses in soybean and cotton in Brazil and are targeted by transgenic Bacillus thuringiensis (Bt) cultivars producing Cry1A and Cry2A toxins. In this study, three experiments were conducted to determine: i) the susceptibility of Brazilian populations of C. includens to Cry1Ac and Cry2Aa as well as the toxicity of these toxins to H. virescens, ii) the lethality and fitness effects on C. includens caused by Bt soybean or cotton producing one or the two toxins, and iii) the response to selection for Cry1Ac resistance to assess the risk of evolution of resistance and cross-resistance to Cry toxins in C. includens. Cotton and soybean cultivars used included two non-Bt isolines, two single-gene Bt cultivars (Cry1Ac cotton and soybean), and two pyramided Bt cottons: (Cry1Ac + Cry2Ab2 and Cry1Ac + Cry1Fa). Diet-surface bioassays showed that susceptibility of C. includens to Cry1Ac and Cry2Aa was variable among the populations tested. LC50 values ranged from 16 to 241 ng/cm2 for Cry1Ac and from 3 to 24 ng/cm2 for Cry2Aa, respectively. Leaf-tissue bioassays with Cry1Ac soybean and pyramided Bt cotton caused 100% mortality of C. includens larvae after two days of exposure. Cry1Ac cotton caused low mortality but negatively affected life-history traits and strongly reduced the net reproductive rate (R0) as well as the intrinsic rate of increase (rm) compared with controls. Selection for Cry1Ac resistance in C. includens using continuous exposure to Cry1Ac cotton during larval development or purified Cry1Ac protein for seven days resulted in 31- and 126- fold resistance with realized heritability of 0.78 and 0.41, respectively, after 12 generations of selection. Importantly, this level of resistance in the Cry1Ac-selected populations was insufficient to allow for survival of larvae in foliage of the Bt soybean producing Cry1Ac. Additionally, soybean loopers of the two laboratory-selected populations showed resistance to Cry1Ab too, but not the Cry2Aa or Cry1Fa in relation to the control, non-selected susceptible population. Taken together, the results of this investigation show that there is natural variation in susceptibility to Cry1Ac and Cry2Aa in the C. includens populations, but the single-toxin Bt soybean and the two- toxin Bt cottons efficiently killed their larvae, indicating that cultivars of these Bt crops should be effective for managing populations of this pest species in field settings. In addition, this study shows that C. includens have potential to evolve relevant resistance levels to Cry1Ac, yet the increase of about 130 fold in the resistance does not allow for survival of neonate soybean loopers in the Bt Cry1Ac soybean. Furthermore, selection for resistance to Cry1Ac did not lead to cross-resistance to Cry2A and Cry1F, suggesting that these two Bt toxins are compatible for resistance management of C. includens.Item Bt toxins and insecticides against noctuid pests: susceptibility, parental effects, and growth potential(Universidade Federal de Viçosa, 2020-04-30) Rabelo, Marcelo Mendes; Pereira, Eliseu José Guedes; http://lattes.cnpq.br/1122797337931616Lepidopteran has around 120 thousand species described, and all of them are phytophagous. Several species, mostly Noctuidae damage row crops cultivated across the south and north Americas. Historically, lepidopteran pests have been managed with the use of synthetic insecticides, and plants expressing Bacillus thuringiensis toxins. This thesis has reported the effect of Bt toxins and insecticides against noctuid pests from Brazil and the United States. In Brazil, the susceptibility Spodoptera cosmioides (Walker), to three Cry toxins and its life- history traits feeding on dual-gene Bt cotton and an artificial diet containing sublethal concentrations of Cry1Ac was documented. In concentration-response bioassays, S. cosmioides larvae showed low susceptibility to Cry toxins. The decreasing rank of toxicity was Cry1F, Cry2Aa, and Cry1Ac. Foliage of dual-toxin Bt cotton varieties (Cry1Ac+Cry1F and Cry1Ab+Cry2Ae) caused higher larval mortality than Cry1Ac-expressing cotton. Spodoptera cosmioides showed reduced larval weight when growing on the Cry1Ac-treated diet, yet they reached adulthood and produced offspring. Interestingly, these offspring when grown on the control diet showed reduced weight gain associated with the toxin exposure of the previous generation, indicating a parental effect of the exposure to Cy1Ac. The reduced larval weight was recovered in later instars, and there was no significant change in the population fitness of the parental S. cosmioides or their offspring. In the United States, Helicoverpa zea (Boddie) life-history traits were determined for individuals of three field populations from a region where this pest likely overwinter. Triple-gene Bt cotton cultivars expressing Cry and Vip3Aa toxins killed 100% of the larvae in all populations tested. In contrast, dual-gene Bt cotton expressing Cry1Ac+Cry1F and Cry1Ac+Cry2Ab2 allowed population growth with the intrinsic rate of population growth (r m ) 38% lower than on non- Bt cotton. The insects feeding on Bt cotton plants expressing Cry1Ac+Cry2Ab2, Cry1Ac+Cry1F, and Cry1Ab+Cry2Ae exhibited reduced larval weight, survival rate, and increased development time. Additionally, fitness parameters varied significantly among the insect populations, even on non-Bt cotton plants, likely because of their different genetic background and/or previous Bt-toxin exposure. The susceptibility of eight noctuid speciesfrom the Florida Panhandle to bifenthrin (pyrethroid) and chlorantraniliprole (diamide) was also determined. Larvae from field and laboratory populations were exposed to commercial insecticide formulations using the leaf-dip method in concentration-mortality bioassays. The field populations of H. zea, S. frugiperda (Smith), S. eridania (Stoll), S. exigua (Hubner), and Chloridea virescens (Fabricius) had reduced susceptibility to bifenthrin compared with the laboratory populations, while there was no reduced susceptibility in Agrotis ipsilon (Hufnagel). The susceptibility to chlorantraniliprole was similar between the field and laboratory populations studied, except for S. exigua that exhibited 630-fold resistance to the diamide. The probit regression equations indicated that the larval mortality of S. exigua and S. frugiperda populations was less than 80% with bifenthrin at the concentration equivalent to the label rate. Likewise, the estimated mortality of S. exigua larvae with chlorantraniliprole at the label rate concentration was less than 80%. This study advances our understanding of potential responses of target and nontarget pest species exposed to Bt toxins and insecticides, which help to design pest management programs for lepidopteran species in the Americas.
