Abstract
Staphylococcus cohnii is an opportunistic coagulase-negative Staphylococcus that has been identified in human, animal, and environmental sources, with reports of antimicrobial resistance and biofilm formation. In this study, the bacteriophage pSS8, isolated from swine wastewater, was investigated for its biological properties, antibiofilm activity, interaction with antibiotics, and genomic features. The phage was isolated using S. cohnii SS8 as the host strain and characterized through assays of lytic spectrum, adsorption, physicochemical stability, antibiofilm activity, synergism with tetracycline and erythromycin, electron microscopy, and comparative genomic analysis. pSS8 formed clear lytic plaques, showed morphology consistent with tailed bacteriophages, rapidly adsorbed to the host cell, and infected 17 of 42 strains evaluated (40.5%), including eight different species. The phage maintained infectivity under different conditions of pH, temperature, UV radiation, and chloroform exposure, reduced both developing and pre-established biofilms, and showed synergistic activity with tetracycline and erythromycin. Its genome is 145,736 bp in length and contains 258 predicted CDSs, with no genes associated with antimicrobial resistance, bacterial virulence, or canonical markers of lysogeny. Comparative analyses placed pSS8 within the family Herelleviridae, close to phages of the genus Baoshanvirus, but with sufficient genomic differences to classify it as a novel species. These findings indicate that pSS8 has relevant attributes for future studies on the control of S. cohnii and other members of Staphylococcaceae. Keywords: Staphylococcus cohnii; Staphylococcaceae; biofilm; antimicrobial resistance; phage therapy
