In this context, leaf structures such as bundle sheath extensions (BSEs) can modulate the response to water deficit by influencing the leaf’s hydraulic architecture. The objective of this study was to investigate the role of BSEs in anatomical, physiological, and metabolic responses to water stress in tomato (Solanum lycopersicum L.), as well as to analyze the interaction between the OBSCURAVENOSA (OBV) and AUXIN RESPONSE FACTOR 4 (ARF4) genes in leaf development. To this end, the Micro-Tom (MT) genotype and the obv mutant were used, grown under controlled conditions, water deficit, and rehydration in a factorial design. Morphological, anatomical, physiological, metabolic, and gene expression parameters were evaluated, in addition to the characterization of segregating populations. The results demonstrated that the obv mutant exhibited leaf morphoanatomical changes, such as increased mesophyll thickness and modifications in vascular and stomatal density, associated with distinct water-use strategies under water stress. Physiological and metabolic changes indicative of greater resource-use efficiency and higher drought tolerance were also observed compared to the MT genotype. The investigation of the interaction between the OBV and ARF4 genes focused on standardizing genotyping strategies for identifying homozygous double mutants, with the evaluation of their effects on BSE remaining a future prospect. Thus, the results indicate that the absence of BSEs may confer adaptive advantages under water stress, contributing to the understanding of the mechanisms involved in drought tolerance in tomato plants.
