Proteínas de baru: extração e potencial como carreadoras de curcumina

Abstract

A crescente busca por ingredientes alimentares sustentáveis e funcionais tem estimulado pesquisas sobre proteínas vegetais extraídas de espécies nativas do Brasil. Esta tese investigou as proteínas da amêndoa de baru (Dipteryx alata Vogel), com foco em métodos de extração, propriedades tecno-funcionais e seu potencial como carreadores de curcumina (CUR), um composto bioativo com reconhecida atividade antioxidante, mas com aplicação limitada em função da sua baixa solubilidade em água e instabilidade química frente à luz. O trabalho foi composto por dois estudos complementares. O primeiro artigo, de natureza revisional, abordou os métodos de extração convencionais para obter as proteínas de baru e informações sobre as propriedades físico-químicas e tecno-funcionais das proteínas do baru que poderão contribuir com conhecimentos científicos básicos estimulando a aplicação dessa fonte proteica em sistemas alimentícios. Dentre essas informações, pode-se citar a elevada solubilidade em pH alcalino (83%), boa capacidade emulsificante (54,8%), estabilidade de espuma (até 96%), e propriedades satisfatórias de retenção de água (118,3%) e óleo (155,5%). A principal lacuna identificada foi a escassez de estudos sobre a capacidade de gelificação das proteínas, que seria essencial para aplicações em produtos vegetais estruturados. O segundo artigo investigou as interações intermoleculares entre a CUR e os isolados proteicos de baru (IPB) e do soro de leite (IPS), visando à formação de complexos com maior estabilidade fotoquímica. A espectroscopia de fluorescência indicou supressão estática, sugerindo a formação de complexos proteína–CUR. A análise termodinâmica mostrou que, a 25°C, os valores de G° foram –18,08kJ·mol¹ (IPB- CUR) e –22,76kJ·mol¹ (IPS-CUR), indicando que a formação dos complexos é termodinamicamente favorecida em relação à existência dos componentes na forma livre. Os valores negativos de TS° (–237,82kJ·mol¹ para IPB e –63,81kJ·mol¹ para IPS) refletem redução da entropia no processo. A formação dos complexos foi predominantemente guiada por interações entalpicamente favoráveis, com H° = –255,90kJ·mol¹ para IPB-CUR e H° = –86,57kJ·mol¹ para IPS-CUR. A interação com os isolados proteicos aumentou a fotoestabilidade da CUR, reduzindo sua constante de fotodegradação e ampliando o tempo de meia-vida. A complexação da CUR com isolados proteicos não aumentou significativamente sua atividade antioxidante, mas os sistemas mantiveram sua atividade antioxidante. Esses resultados indicam que os isolados proteicos estabilizam a CUR, destacando o IPB como uma alternativa promissora ao IPS para formulações vegetais funcionais, especialmente para consumidores veganos ou intolerantes a lácteos. A tese contribui para a valorização de biomas nativos e o desenvolvimento de sistemas alimentares mais saudáveis e inovadores. The growing demand for sustainable and functional food ingredients has encouraged the research of plant proteins extracted from native species of Brazil. This thesis investigated proteins from baru almonds (Dipteryx alata Vogel), focusing on extraction methods, techno-functional properties, and their potential as carriers for curcumin (CUR), a bioactive compound known for its antioxidant activity but limited by poor water solubility and chemical photoinstability. The work comprised two complementary studies. The first article, a review, discussed conventional extraction methods (such as alkaline extraction with isoelectric precipitation) and emerging techniques (including ultrasound, deep eutectic solvents, pulsed electric fields, and enzyme-assisted extraction), to produce baru almond proteins. Furthermore, the review presents important information on the physicochemical and techno-functional properties of baru proteins, which may contribute to basic scientific knowledge, stimulating the application of this protein source in food systems. Among this information, one can mention the baru protein isolates exhibited high solubility under alkaline pH (~83%), good emulsifying capacity (54.8%), high foam stability (up to 96%), and satisfactory water- (118.3%) and oil-holding capacities (155.5%). The main knowledge gap identified was the lack of studies on gelation behavior, which is essential for applications in structured plant-based products. The second article investigated the intermolecular interactions between CUR and protein isolates from baru (IPB) and whey (IPS), aiming to form complexes with enhanced photochemical stability. Fluorescence spectroscopy indicated static quenching, suggesting the formation of CUR–protein complexes. Thermodynamic analysis showed that at 25°C, the Gibbs free energy values were –18.08kJ·mol¹ (IPB–CUR) and –22.76kJ·mol¹ (IPS–CUR), indicating that complex formation is thermodynamically favored compared to the free components. The negative TS° values (–237.82kJ·mol¹ for IPB and –63.81kJ·mol¹ for IPS) reflect entropy reduction during the process. Complex formation was predominantly driven by favorable enthalpic interactions, with H° = –255.90kJ·mol¹ for IPB–CUR and –86.57kJ·mol¹ for IPS–CUR. The interaction with protein isolates enhanced CUR photostability by reducing its photodegradation constant and increasing its half-life. Although complexation did not significantly increase CUR antioxidant activity, the systems maintained their antioxidant activity. These findings indicate that protein isolates stabilize CUR effectively, with IPB emerging as a promising alternative to IPS for plant-based functional formulations, especially suitable for vegan consumers or individuals with dairy intolerance. This thesis contributes to the valorization of brazilian native biomes and the development of healthier and more innovative food systems.
