Permeabilização da parede celular de basidiósporos de Pisolithus microcarpus por agentes químicos e enzimáticos

Resumo
Pisolithus é gênero fúngico com espécies capazes de formar associações simbióticas
ectomicorrízicas com grupos florestais de alto valor econômico, a exemplo dos pinhos
e eucaliptos. A germinação in vitro de basidiósporos de Pisolithus é baixa,
inviabilizando o uso desse material como inoculantes comerciais e dificultando a
obtenção de isolados monocarióticos, importantes para estudos genéticos. A
impermeabilidade da parede celular, observada em esporos maduros, pode ser
responsável pela baixa porcentagem de germinação in vitro. O objetivo do presente
trabalho foi desenvolver método de permeabilização da parede celular de
basidiósporos de P. microcarpus com agentes químicos e enzimas visando à
manutenção da viabilidade desses propágulos. Foram testados como agentes
permeabilizadores Triton X-100, ácido metanoico, hidróxido de sódio e os coquetéis
enzimáticos comerciais Lysing Enzymes e Driselase, em diferentes concentrações e
tempos de incubação. NaOH nas concentrações de 100, 250, 500 e 1000 mmol L -1
mostrou-se eficaz na remoção de pigmentos e proteínas, resultando na
permeabilização dos basidiósporos sem perda de viabilidade. O melhor tratamento foi
com NaOH a 1000 mmol L -1 por um minuto, permeabilizando 97 % dos basidiósporos.
A permeabilização com NaOH mostrou-se reprodutível quando aplicada a
basidiósporos oriundos de 9 áreas de coleta distintas. As enzimas líticas não foram
capazes de permeabilizar os esporos quando aplicadas isoladamente. No entanto,
após o tratamento prévio dos basidiósporos com NaOH, Lysing Enzymes aumentou
significativamente as porcentagens de permeabilização quando aplicadas nas
concentrações de 30 e 60 mg mL -1 . O método desenvolvido tem potencial para
melhorar as porcentagens de germinação dos basidiósporos de P. microcarpus,
facilitando a obtenção de estirpes monocarióticas e o desenvolvimento de inoculantes
ectomicorrízicos.

In vitro germination rates of Pisolithus basidiospores are low, making the use of this material as commercial inoculants unfeasible and making it difficult to obtain monokaryotic isolates, which are important for genetic studies. The impermeability of the cell wall, observed in mature badidiospores, may be responsible for the low in vitro germination percentages. The objective of the present work was to develop a method for permeabilizing the cell wall of P. microcarpus basidiospores with chemical agents and enzymes maintaining the viability of these propagules. Triton X-100, methanoic acid, sodium hydroxide and the commercial enzymatic cocktails Lysing Enzymes and Driselase were tested as permeabilizing agents at different concentrations and incubation times. NaOH at concentrations of 100, 250, 500 and 1000 mmol L -1 proved to be effective in removing pigments and proteins, resulting in basidiospore permeabilization without loss of viability. The method developed herein has the potential to improve the germination percentages of P. microcarpus basidiospores, facilitating the obtention of monokaryotic strains and the development of ectomycorrhizal inoculants.
