Item Metais pesados em pequenos mamíferos: efeitos em modelos murinos e em morcegos frugívoros(Universidade Federal de Viçosa, 2023-08-24) Destro, Ana Luiza Fonseca; Freitas, Mariella Bontempo Duca; http://lattes.cnpq.br/5840448577950611Os metais liberados no ambiente durante a mineração são persistentes no ambiente e contaminam cadeias alimentares. Dentre os animais que são afetados por poluentes, os morcegos estão susceptíveis a contaminação oral ou por contato. Devido ao hábito alimentar variado e a capacidade de voo, esses mamíferos prestam serviços ecológicos e econômicos importantes como dispersão de sementes, polinização, contribuição na fertilidade do solo e indicador ecológico de qualidade ambiental, portanto, a contaminação de populações de morcegos é prejudicial à conservação ambiental. Apesar dos efeitos de diversos metais em modelos murinos ser bem descrito, as formas de prevenção, os efeitos danosos da contaminação em animais selvagens e a avaliação dos danos da liberação de metais pela mineração ainda não estão claros. Portanto, essa tese foi dividida em três capítulos no formato de artigos científicos. No primeiro capítulo foi apresentado uma revisão sistemática dos efeitos de extratos vegetais na toxicidade hepática induzida por chumbo (Pb) em modelos murinos. Esta revisão sistemática teve como objetivo trazer uma atualização e novas perspectivas da ação dos extratos vegetais no metabolismo hepático como prevenção da toxicidade de Pb. No segundo capítulo, foi realizado uma análise experimental e comparativa da toxicidade de baixas doses de quatro metais: Niquel (Ni), Cadmio (Cd), Cromo (Cr) e Chumbo (Pb) em morcegos fugívoros (Artibeus lituratus) em cativeiro. O terceiro capítulo foi realizado um estudo in situ com o objetivo de avaliar possíveis efeitos da mineração de Ferro (Fe) e Alumínio (Al) em morcegos frugívoros morcegos. uma avaliação dos efeitos fisiológicos da. No trabalho experimental, observou-se que o fígado mostrou maior sensibilidade quando os animais foram tratados com Ni ou Pb. Cr, Cd e Ni foram mais nocivos para os rins. Nos testículos, o Ni dobrou os níveis de dano em comparação aos outros metais. Já os músculos mostraram ser mais sensíveis ao Pb e Cd. O cérebro demonstrou ser mais suscetível a Pb e Ni. Nossos resultados demonstram que doses agudas, mesmo em baixas doses (1,5mg/kg), causaram efeitos prejudiciais em vários órgãos de morcegos frugívoros, o que é uma preocupação para o ecossistema. Assim, baseados em nossos achados, propomos a seguinte ordem de toxicidade dos metais em morcegos frugívoros: Ni> Pb> Cd> Cr. Nós observamos que morcegos frugívoros (A. lituratus) coletados em área de mineração de Fe naMata Atlântica possuem mais estresse oxidativo no fígado, rins, cérebro e músculo, além de histopatologias no fígado e nos rins em comparação à A. lituratus de fragmento de Mata Atlântica sem mineração. Já os morcegos frugívoros (Sturnira lilium) coletados em área de mineração de Al, apresentaram alterações em alguns marcadores de balanço redox, indicando estresse oxidativo, quando comparados aos S. lilium de fragmento de Mata Atlântica sem mineração. Os morcegos da área de mineração de Al apresentaram histopatologias nos rins e fígado, bem como fibroses no fígado. Os resultados do terceiro capítulo demonstram que morcegos da mineração de Fe na Mata Atlântica estão mais afetados do que os da mineração de Al. Porém ambas as áreas demonstram que os morcegos frugívoros estão sofrendo com os efeitos da mineração. Todos os resultados dos três capítulos da tese, demonstram que metais pesados são um problema para a sobrevivência de pequenos mamíferos e a legislação deve ser atualizada e seguida para diminuir a exposição dos animais selvagens aos metais pesados vindos da ação antrópica. Histologia.Item Phytase and carbohydrases in plant-based diets for tambaqui juveniles (Colossoma macropomum)(Universidade Federal de Viçosa, 2024-03-18) Carneiro, Cristiana Leonor da Silva; Salaro, Ana Lúcia; http://lattes.cnpq.br/7081260633740252With this study, we aimed to investigate the effects of dietary supplementation with phytase and exogenous carbohydrates on juvenile tambaqui (Colossoma macropomum) by evaluating growth performance, body composition, plasma biochemistry, activity of digestive enzymes, intestinal histomorphometry, intestinal microbiota, and gene expression analysis of liver tissue (RNA-seq). An extruded basal diet (307.8 g kg−1 of crude protein and 17.08 MJ kg−1 of gross energy), composed of vegetable ingredients, was used in the preparation of four experimental diets: CON = control diet without enzyme supplementation, PHY = diet supplemented with phytase, XB = diet supplemented with xylanase and β-glucanase, and PHY+XB = diet supplemented with phytase, xylanase, and β-glucanase. To achieve the proposed objectives, an 8-week experimental trial was conducted with juveniles weighing 1.01 ± 0.15 g. Enzyme supplementation did not significantly affect fish survival, final weight, weight gain, feed conversion rate, or visceral indices. However, fish fed PHY+XB showed a higher protein efficiency rate and lower visceral fat index compared to those fed CON. This suggests better protein utilization and reduced fat deposition. Furthermore, fish fed PHY+XB had higher body protein and Fe3+ content. Plasma biochemistry analysis revealed higher levels of HDL and lower levels of plasma protein in fish fed PHY+XB, along with reduced ALP levels, indicating potential metabolic benefits. Enzyme supplementation did not affect the activities of total proteases, lipase, and amylase in the fish intestine, nor did it alter the intestinal epithelial structure. High-throughput sequencing analysis revealed that the majority of reads derived from tambaqui digesta belong to the Firmicutes and Proteobacteria phyla. Fish fed the PHY+XB diet showed a greater number of enriched taxa compared to the other groups. However, in general, all treatments were enriched with bacteria responsible for the manipulation and utilization of carbohydrates and other nutrients, demonstrating a beneficial modulation for improved utilization of dietary nutrients. Differential analysis of the tambaqui transcriptome (RNA-seq) revealed a total of 138 differentially expressed genes (DEGs) between CON vs. PHY, 19 genes between CON vs. XB, and 77 genes between CON vs. PH + XB. Through Gene Ontology (GO) enrichment analysis, we identified that the PHY and PHY+XB diets modulated the expression of genes related to oxygen homeostasis and redox processes. The PHY diet also affected the fish's immune system. The XB diet mainly affects carbohydrate metabolism. In summary, supplementing with phytase and xylanase-β-glucanase has shown promise in enhancing dietary utilization, body composition, positively influencing the intestinal microbiota, and regulating essential metabolic processes epigenetically. Notably, the combination of these enzymes has proven to be more effective, emphasizing the significance of the synergy between the two compounds in enhancing the observed benefits. Despite its greatest diversity being concentrated in the tropics, the Neotropical Region remains understudied concerning this subfamily. Therefore, the objective of this thesis is to fill the existing knowledge gap about these insects. To achieve this goal, collections, dissections, redescriptions, and descriptions of new species were conducted. The research has resulted in four already published articles, with a fifth in the process of publication. In the first article, a new species (Scaphisoma pandemum von Groll & Lopes-Andrade, 2021) is described for Brazil. The second article introduces a new method for collecting beetles in fungi. The third article describes five new species of the genus Cyparium Erichson, 1845, and redescribes two species. The fourth article brings a new record for Brazil: S. nigrofasciatum Pic, 1915, original from Asia was collected in Minas Gerais. The fifth article, yet to be published, presents 20 new species distributed across five genera: Cyparium, Alexidia Reitter, 1880, Baeocera Erichson, 1845, Scaphisoma Leach, 1815, and Toxidium LeConte, 1860. All taxonomic papers include, whenever possible, illustrations of the external and internal morphology of males and females, accompanied by data related to the host. In addition to taxonomic results, the collections made in Minas Gerais have significantly expanded the Entomological Collection of the Laboratory of Systematics and Biology of Coleoptera at the Federal University of Viçosa (CELC), making it the largest collection of Scaphidiinae in Brazil, with more than 1800 specimens, enabling the continuation of research on this group.
