Item A new methodology for evaluation of nematode viability(BioMed Research International, 2015-02-19) Ferreira, Sebastião Rodrigo; Mendes, Tiago Antônio Oliveira; Bueno, Lilian Lacerda; Araújo, Jackson Victor de; Bartholomeu, Daniella Castanheira; Fujiwara, Ricardo ToshioNematodes infections are responsible for debilitating conditions and economic losses in domestic animals as well as livestock and are considered an important public health problem due to the high prevalence in humans. The nematode resistance for drugs has been reported for livestock, highlighting the importance for development of new anthelmintic compounds. The aim of the current study was to apply and compare fluorimetric techniques using Sytox and propidium iodide for evaluating the viability of C. elegans larvae after treatment with anthelmintic drugs. These fluorescent markers were efficient to stain larvae treated with ivermectin and albendazole sulfoxide. We observed that densitometric values were proportional to the concentration of dead larvae stained with both markers. Furthermore, data on motility test presented an inverse correlation with fluorimetric data when ivermectin was used. Our results showed that lower concentrations of drugs were effective to interfere in the processes of cellular transport while higher drugs concentrations were necessary in order to result in any damage to cell integrity. The methodology described in this work might be useful for studies that aim to evaluate the viability of nematodes, particularly for testing of new anthelminthic compounds using an easy, economic, reproducible, and no time-consuming technique.Item Ação ovicida do extrato bruto enzimático do fungo Pochonia chlamydosporia sobre ovos de Ancylostoma sp(Revista da Sociedade Brasileira de Medicina Tropical, 2010-09-01) Braga, Fabio Ribeiro; Araujo, Juliana Milani; Silva, André Ricardo e; Araújo, Jackson Victor de; Carvalho, Rogério Oliva; Soares, Filippe Elias de Freitas; Queiroz, José Humberto de; Gênier, Hugo Leonardo AndréAncylostoma sp é um geo-helminto potencialmente zoonótico. O objetivo deste trabalho foi avaliar in vitro a ação do extrato bruto enzimático de Pochonia chlamydosporia (VC4) sobre ovos de Ancylostoma sp, em meio ágar-água 2% e em cultura de fezes. Observou-se um percentual de redução na eclosão dos ovos de Ancylostoma sp, de 76,8% na placas de Petri do grupo tratado em relação ao grupo controle. O extrato bruto enzimático de Pochonia chlamydosporia foi eficiente na redução da eclosão dos ovos de Ancylostoma sp, podendo ser utilizado como controlador biológico desse nematoide.Item Activity of the nematophagous fungi Pochonia chlamydosporia, Duddingtonia flagrans and Monacrosporium thaumasium on egg capsules of Dipylidium caninum(Veterinary Parasitology, 2009-12-03) Araujo, Juliana Milani; Araújo, Jackson Victor de; Braga, Fabio Ribeiro; Carvalho, Rogério Oliva; Ferreira, Sebastião RodrigoNematophagous fungi are potential biological control agents of helminths. The in vitro ovicidal effect of four isolates of the nematophagous fungi Pochonia chlamydosporia (VC1 and VC4), Duddingtonia flagrans (AC001) and Monacrosporium thaumasium (NF34) was evaluated on egg capsules of Dipylidium caninum, a cestode parasite of dogs, cats and humans. One thousand egg capsules of D. caninum were plated on 2% water-agar with the grown isolates and control without fungus. The ovicidal activity of these fungi was evaluated 5, 10 and 15 days after incubation. Only P. chlamydosporia showed ovicidal activity (p < 0.05) on D. caninum egg capsules, of 19.6% (VC1) and 20% (VC4) on the 5th day; 44.2% (VC1) and 31.5% (VC4) on the 10th day; and 49.2% (VC1) and 41.9% (VC4) on the 15th day. D. flagrans and M. thaumasium caused no morphological damage to egg capsules. The results demonstrated that P. chlamydosporia was in vitro effective against capsules and eggs of D. caninum, and can be considered as a potential biological control agent for this helminth.Item Ascaris suum in pigs of the Zona da Mata, Minas Gerais State, Brazil(Revista Brasileira de Parasitologia Veterinária, 2015-04-15) Fausto, Mariana Costa; Oliveira, Isabela de Castro; Fausto, Guilherme Costa; Carvalho, Lorendane Millena de; Valente, Fabrício Luciani; Campos, Artur Kanadani; Araújo, Jackson Victor deAmong the parasites that affect pigs, Ascaris suum stands out for causing the greatest losses to livestock production systems. This parasite can be monitored during the slaughter of animals through the identification of “milk spots” or white patches on the liver caused by its larval migration. However, infection in the herd is usually subclinical, which is why the presence of this parasite in industrial pig production has been overlooked. The aim of the study was therefore to evaluate the occurrence of milk spots on the liver of animals slaughtered in the micro-region of Ponte Nova in the Zona da Mata - Minas Gerais, Brazil, and to associate these lesions with the time of year, herd size and source of origin of the animals. An evaluation was made of 1,069 lots, totaling 108,073 animals, based on data extracted from the Federal Inspection Service. The animals were slaughtered during the period of January 2011 to June 2013. Out of the total number of slaughtered animals, 10,535 (9.75%) tested positive for these lesions. Therefore, veterinarians and producers should be warned about the inefficiency of the deworming protocols that are used, and the need to develop and/or review control strategies for this parasite in production systems.Item Atividade larvicida do extrato bruto enzimático do fungo Duddingtonia flagras sobre larvas de primeiro estádio de Angiostrongylus vasorum(Revista da Sociedade Brasileira de Medicina Tropical, 2011-01-05) Braga, Fabio Ribeiro; Araujo, Juliana Milani; Tavela, Alexandre de Oliveira; Araújo, Jackson Victor de; Soares, Filippe Elias de Freitas; Geniêr, Hugo Leonardo André; Lima, Walter dos Santos; Mozzer, Lanuze Rose; Queiroz, José Humberto deAngiostrongylus vasorum é um nematóide que parasita cães domésticos e eventualmente o homem. O objetivo deste trabalho foi observar a atividade predatória in vitro do extrato bruto enzimático do fungo Duddingtonia flagrans sobre larvas de primeiro estádio A. vasorum em condições laboratoriais no meio ágar-água 2%. Ao final do experimento, os percentuais de redução das L1 de A. vasorum observados foram de: 53,5% (24h) e 71,3% (48h) O extrato bruto enzimático do fungo D. flagrans destruiu in vitro as L1, podendo ser utilizado como controle biológico desse nematóide.Item Atividade predatória de isolados de Arthrobotrys spp sobre larvas infectantes de Cooperia punctata(Brazilian Journal of Veterinary Research and Animal Science, 1997-04-16) Araújo, Jackson Victor deExperimentos laboratoriais foram realizados para investigar a capacidade de isolados de fungos predadores das espécies Arthrobotrys musiformis (isolado 3), A. conoides (isolado A) e A. robusta (isolados B e E) de predar e matar larvas infectantes de Cooperia punctata. Dois grupos foram formados para o teste de cada isolado: grupo 1, fungos e larvas infectantes e grupo 2, larvas infectantes (controle). Houve diferença estatisticamente significativa (p<0,05) entre a atividade predatória do isolado E de A. robusta, quando comparado com todos os outros isolados de Arthrobotrys spp (isolados A, B e 3). Nenhuma diferença estatística (p>;0,05) foi encontrada entre o isolado E e o grupo controle. O objetivo deste trabalho foi comparar a capacidade predatória dos fungos nematófagos Duddingtonia flagrans (AC001), Monacrosporium thaumasium (NF34) e Arthrobotrys robusta (I-31) sobre larvas infectantes (L3) de Strongyloides stercoralis em condições laboratoriais no meio ágar-água 2%. Ao final do experimento, os percentuais de redução de L3 de Strongyloides stercoralis observados foram de: 83,7% (AC001); 75,5% (NF34) e 73,2% (I-31). Os fungos nematófagos foram capazes de capturar e destruir in vitro as L3, podendo ser utilizados como controladores biológicos de Strongyloides stercoralis.Item Atividade predatória, crescimento radial e esporulação de fungos predadores de nematóides Monacrosporium spp, submetidos à criopreservação(Ciência Rural, 2004-03) Campos, Artur Kanadani; Mota, Marcelo de Andrade; Araújo, Jackson Victor de; Cecon, Paulo RobertoTestes in vitro foram realizados para avaliar o efeito da criopreservação em nitrogênio líquido com e sem adição de crioprotetores (DMSO ou glicerol), na atividade predatória sobre larvas infectantes de Cooperia sp e Haemonchus sp, crescimento radial e produção de conídios de dois isolados de fungos predadores de nematóides (Monacrosporium sinense e Monacrosporium appendiculatum). A capacidade predatória dos fungos sobre larvas infectantes de Cooperia sp e Haemonchus sp dos fungos previamente submetidos aos diferentes métodos de preservação não foi afetada. O crescimento radial dos fungos criopreservados com glicerol 10% foi mais expressivo(p<0,05), quando comparado ao apresentado pelos fungos congelados com DMSO 10 % ou água. A produção de esporos foi maior (p< 0,05) no fungo M. appendiculatum preservado a 4ºC em geladeira.Item Biological control of Ascaris suum eggs by Pochonia chlamydosporia fungus(Veterinary Research Communications, 2011-07-28) Ferreira, Sebastião Rodrigo; Araújo, Jackson Victor de; Braga, Fábio Ribeiro; Araujo, Juliana Milani; Frassy, Luiza Neme; Ferreira, Aloízio SoaresAscaris suum is a gastrointestinal nematode parasite of swines. The aim of this study was to observe Pochonia chlamydosporia fungus on biological control of A. suum eggs after fungus passage through swines gastrointestinal tract. Eighteen pigs, previously dewormed, were randomly divided into three groups: group 1, treated with the fungus isolate VC4; group 2, treated with the fungus isolate VC1 and group 3 did not receive fungus (control). In the treated groups, each animal received a 9 g single dose of mycelium mass containing P. chlamydosporia (VC1 or VC4). Thereafter, animal fecal samples were collected at the following intervals: 8, 12, 24, 36, 48, 72 and 96 h after treatment beginning and these were poured in Petri dishes containing 2% water-agar culture medium. Then, 1,000 A. suum eggs were poured into each dish and kept in an incubator at 26°C and in the dark for 30 days. After this period, approximately 100 eggs were removed from each Petri dish and morphologically analyzed under light microscopy following the ovicidal activity parameters. The higher percentage observed for isolated VC4 eggs destruction was 57.5% (36 h) after fungus administration and for isolate VC1 this percentage was 45.8% (24 h and 72 h) (p > 0.01). P. chlamydosporia remained viable after passing through the gastrointestinal tract of swines, maintaining its ability of destroying A. suum eggs.Item Biological control of goat gastrointestinal helminthiasis by Duddingtonia flagrans in a semi-arid region of the northeastern Brazil(Veterinary Parasitology, 2012-08-13) Braga, Fabio Ribeiro; Araújo, Jackson Victor de; Vilela, Vinícius Longo Ribeiro; Feitosa, Thais Ferreira; Souto, Diego Vagner de Oliveira; Santos, Herbis Eduardo da Silva; Silva, Gabriela Lucena Longo da; Athayde, Ana Célia RodriguesThe aim of this study was to test a pellet formulation in a sodium alginate matrix of Duddingtonia flagrans in the biological control of goat gastrointestinal helminths kept in a native pasture in a semi-arid region of Paraíba state, northeastern Brazil. An area of 2.4 ha was divided into three paddocks, where groups of seven goats ware formed. Each group received the following treatments during the months of March to August 2011: D. flagrans group, received 3 g of pellets containing D. flagrans (AC001) for each 10 kg/l. w., twice a week; Moxidectin 0.2% group, received 0.2 mg/kg of Moxidectin 0.2% orally, every 30 days; Control group, received 3 g of pellets without fungi per 10 kg/l. w., twice a week. Each month, a tracer goat was placed in each group for 30 days and then sacrificed and necropsied. The D. flagrans group showed a greater reduction in EPG, increased weight gain, higher rates of packed cell volume and lower parasitic load burden in the tracer goats compared to Moxidectin 0.2% and Control groups. D. flagrans was efficient in controlling goat gastrointestinal helminthiasis in a semi-arid region of northeastern Brazil.Item Coadministration of nematophagous fungi for biological control over gastrointestinal helminths in sheep in the semiarid region of northeastern Brazil(Veterinary Parasitology, 2016-03-30) Vilela, Vinícius Longo Ribeiro; Feitosa, Thais Ferreira; Braga, Fabio Ribeiro; Araújo, Jackson Victor de; Santos, Antonielson dos; Morais, Dayana Firmino de; Souto, Diego Vagner de Oliveira; Athayde, Ana Célia RodriguesThis study aimed to evaluate coadministration of Duddingtonia flagrans and Monacrosporium thaumasium in a sodium alginate matrix for controlling gastrointestinal helminths in young and adult sheep in the semiarid region of northeastern Brazil. An area of 1 ha was divided into two paddocks, in which two experimental groups (fungus and control) were formed, each consisting of six adult females and ten young males. In each group, two subgroups were formed in accordance with the animal category (adult or young). In the fungus group, each animal received 3 g of pellets containing 0.6 g of fungal mycelium, with 0.3 g of D. flagrans and 0.3 g of M. thaumasium for each 10 kg of body weight, in their feed twice a week, for six months. In the control group, each animal received 3 g of pellets without fungus for each 10 kg of body weight, in their feed twice a week, for six months, serving as a witness group. Reductions in numbers of eggs per gram of feces of 76% among the adult sheep in the fungus group and 83% among the young sheep in the fungus group were observed, in comparison with their respective control subgroups. The groups that received these fungi needed less salvage deworming and presented better packed cell volume percentages, better weight gain and lower levels of L3/kg dry matter in their paddock than the control groups. Thus, it was concluded that coadministration of D. flagrans and M. thaumasium was effective in controlling gastrointestinal helminths of adults and young sheep in the semiarid region of northeastern Brazil.Item Coadministration of sodium alginate pellets containing the fungi Duddingtonia flagrans and Monacrosporium thaumasium on cyathostomin infective larvae after passing through the gastrointestinal tract of horses(Research in Veterinary Science, 2012-11-22) Tavela, Alexandre de Oliveira; Araújo, Jackson Victor de; Braga, Fábio Ribeiro; Silveira, Wendeo Ferreira da; Silva, Vinicius Herold Dornelas e; Carretta Júnior, Moacir; Borges, Luana Alcântara; Araujo, Juliana Milani; Benjamin, Laércio dos Anjos; Carvalho, Giovanni Ribeiro; Paula, Alessandra Teixeira deThe predatory nematophagous fungi have been used as an alternative control of gastrointestinal nematodes of domestic animals in natural and laboratory conditions. However, it is unclear if the association of some of these species could bring some kind of advantage, from a biological standpoint. In this context, this study consisted of two tests in vitro: in assay A, the assessment of the viability of the association of pellets in sodium alginate matrix containing the fungus Duddingtonia flagrans (AC001) and Monacrosporium thaumasium (NF34) and its predatory activity on infective larvae (L3) of cyathostomin after passing through the gastrointestinal tract of horses and assay B, assessment of the cyathostomin L3 reduction percentage in coprocultures. Twelve crossbred horses, females, with a mean weight of 356 kg and previously dewormed were divided in three groups with four animals each: group 1, each animal received 50 g of pellets containing mycelial mass of the fungus D. flagrans and 50 g of pellets of the fungus M. thaumasium, associated and in a single oral dose; group 2, 100 g of pellets containing D. flagrans and 100 g of pellets containing M. thaumasium, associated and in a single oral dose; group 3, control. Faecal samples were collected from animals in the treated and control groups at time intervals of 12, 24, 36, 48, 60 and 72 h after the administration of treatments and placed in Petri dishes containing 2% water-agar (assay A) and cups for coprocultures (assay B). Subsequently, 1000 cyathostomin L3 were added to each Petri dish (assay A) and 1000 cyathostomin eggs were added to each coproculture (assay B) of fungi-treated and control groups. At the end of 15 days, there was observed that the two associations of pellets containing the fungi tested showed predatory activity after passing through the gastrointestinal tract of horses (assay A). In assay B, all the intervals studied showed reduction rate in the number of L3 recovered from coprocultures exceeding 80%. However, no difference (p > 0.01) was seen in recovery of not predated L3 between the fungi-treated groups in the time intervals studied. The results obtained showed that the associations of pellets (50 or 100 g of each fungal isolate) were viable after passage through the gastrointestinal tract in horses and could be used in natural conditions.Item Control of bovine gastrointestinal nematode parasites using pellets of the nematode-trapping fungus Monacrosporium thaumasium(Ciência Rural, 2004-03) Araújo, Jackson Victor de; Guimarães, Marcos Pezzi; Campos, Artur Kanadani; Sá, Nilo Chaves de; Sarti, Priscilla; Assis, Rafaela Carolina LopesThe viability of a formulation of the fungus Monacrosporium thaumasium associated with ivermectin was evaluated for the biological control of bovine gastrointestinal nematode parasites. Four groups of five calves each were placed in pastures with a stocking rate of 1.6 animal/hectare. In group 1 (control), the calves did not receive any treatment. In group 2, each animal received 20g of pellets of M. thaumasium orally twice a week during a six-month period that began with the onset of the rainy season (October 23, 2000). In group 3, each animal received 20g of pellets of M. thaumasium orally twice a week during the same period as 2, as well as two strategic treatments with ivermectin (200 mcg/kg) on May 10, 2001 and July 5, 2001. In group 4, the animals were treated with ivermectin alone as described for group 3. EPG counts for group 1 were significantly greater (P< 0.01) than those for groups 2 and 3 and the difference at the end of the study period was near 100%. The EPGs of group 4 animals remained high until the first strategic treatment with ivermectin. Values for groups 1 and 4 differed significantly (P< 0.05) from those of groups 2 and 3 from December 2000 onwards. It was concluded that the use of this dose and periodicity of application of M. thaumasium pellets makes the application of anthelminthic treatments unnecessary.Item Control of sheep gastrointestinal nematodes using the combination of Duddingtonia flagrans and Levamisole Hydrochloride 5%(Revista Brasileira de Parasitologia Veterinária, 2018-02-05) Braga, Fabio Ribeiro; Vilela, Vinícius Longo Ribeiro; Braga, Fabio Ribeiro; Vieira, Vanessa Diniz; Lucena, Samuel Cavalcante de; Araújo, Jackson Victor deThe objective was to evaluate the action of D. flagrans pellets in association with Levamisole Hydrochloride 5% for controlling sheep gastrointestinal nematodes in the northeastern Brazil. Three groups of six sheep each were formed: group 1 received 3 g of the pellets (0.6 g of D. flagrans mycelium) for each 10 kg b.w., twice a week for six months, and deworming with Levamisole Hydrochloride 5% when EPG ≥ 1500; group 2 received a dosage of Levamisole Hydrochloride 5% when EPG ≥ 1500; and group 3 received 3 g of pellets without fungi for each 10 kg b.w., twice a week for six months. EPG counts, larval cultures, packed cell volume (PCV) and weighing were performed every 15 days; monthly, samples of grass from each paddock were collected. The mean EPG of the groups began to statistically differ from day 30 (p < 0.05). Group 1 required less deworming with Levamisole Hydrochloride 5% and showed superiority of PCV values throughout the experiment (p < 0.05). There was a significant reduction (p < 0.05) in L3 recovery in the group 1 paddock from day 30 onwards. The use of D. flagrans pellets in association with Levamisole Hydrochloride 5% was effective for controlling gastrointestinal nematodes.Item Controle estratégico experimental do carrapato de bovinos Boophilus microplus (CANESTRINI, 1887) (Acarina: ixodidae) em bezerros do município de Viçosa, Zona da Mata de Minas Gerais, Brasil(Brazilian Journal of Veterinary Research and Animal Science, 1994-02-04) Araújo, Jackson Victor deNa microrregião de Viçosa-MG, foi testado um controle estratégico experimental do carrapato de bovinos Boophilus microplus em bezerros 3/4 Holandês x Zebu pela administração do Amitraz, de outubro de 1990 a fevereiro de 1991, em cinco banhos intervalados de 28 dias. O controle estratégico apresentou efeito redutor sobre o número de formas adultas de Boophilus microplus nos animais tratados, mas não foi suficiente para impedir que estes níveis se elevassem após o início de condições meteorológicas satisfatórias para o desenvolvimento de carrapato na primavera de 1991.Item Curso natural da eliminação de ovos de Eurytrema sp LOSS, 1907 nas fezes de bezerros(Brazilian Journal of Veterinary Research and Animal Science, 1993-10-19) Araújo, Jackson Victor de; Belém, Pacífico Antonio DinizTreze bezerros 3/4 holandês x 1/4 zebu, nascidos em diferentes estações do ano de 1989 e criados intensivamente, foram transferidos para pastagens de capim gordura (Melinis minutiflora) entre 7 meses e 1 ano de idade, no final do período chuvoso (março de 1990), a fim de ser acompanhado o curso natural da eliminação de ovos de Eurytrema sp por grama de fezes (OPG). De 15 em 15 dias, foram feitas as colheitas de fezes para obter as contagens de OPG durante 18 meses. Os primeiros animais que apresentaram exames positivos (a partir de agosto de 1990) foram os que tiveram as maiores contagens de OPG durante o período experimental. Os demais apresentaram os primeiros exames positivos em diferentes ocasiões e as contagens de OPG destes foram baixas e oscilantes. Três animais apresentaram exames negativos. Provavelmente, a maioria dos bovinos criados em regime de pastejo desta região deverão apresentar infecção parasitária por Eurytrema sp em alguma fase de suas vidas.Item Destruição de larvas infectantes de Strongyloides venezuelensis pelos fungos Duddingtonia flagrans, Arthrobotrys robusta e Monacrosporium sinense(Revista da Sociedade Brasileira de Medicina Tropical, 2010-08-04) Braga, Fabio Ribeiro; Araujo, Juliana Milani; Silva, André Ricardo e; Araújo, Jackson Victor de; Carvalho, Rogério Oliva; Tavela, Alexandre de Oliveira; Silva, Manoel Eduardo da; Fernandes, Fernanda Mara; Melo, Alan Lane deStrongyloides venezuelensis tem sido utilizado como um modelo para estudo da estrongiloidose humana. O objetivo deste trabalho foi comparar a capacidade predatória dos fungos nematófagos Duddingtonia flagrans (AC001), Arthrobotrys robusta (I-31) e Monacrosporium sinense (SF53) sobre larvas infectantes (L3) de Strongyloides venezuelensis em condições laboratoriais no meio ágar-água 2%. Ao final do experimento, os percentuais de redução de L3 de Strongyloides venezuelensis observados foram de: 93% (AC001); 77,2% (I-31) e 65,2% (SF53). Os fungos nematófagos foram capazes de capturar e destruir in vitro as L3, podendo ser utilizados como controladores biológicos de Strongyloides venezuelensis.Item Destruição de ovos de Toxocara canis pelo fungo nematófago Pochonia chlamydosporia(Revista da Sociedade Brasileira de Medicina Tropical, 2009-12-10) Frassy, Luiza Neme; Braga, Fabio Ribeiro; Silva, André Ricardo e; Araújo, Jackson Victor de; Ferreira, Sebastião Rodrigo; Freitas, Leandro Grassi deToxocara canis é um ascarídeo parasita do intestino delgado de cães, causador da larva migrans visceral em seres humanos. Com o objetivo de demonstrar a eficácia do fungo Pochonia chlamydosporia sobre ovos de Toxocara canis em condições laboratoriais, foi montado ensaio experimental em placas de Petri com ágar-água 2%. Houve atividade ovicida de 43,8% (p<0,01) do grupo tratado em relação ao grupo controle durante os intervalos estudados. The growth of the genus Duddingtonia present in formulation of rice bran, its predatory capability on Oesophagostomum spp. infective larvae (L3) in petri dishes (assay 1), its action in faecal cultures with eggs of that parasite (assay 2) and isolate's capability of predation after passing through gastrointestinal tract of swine (assay 3) was evaluated. At assay 3, feces were collected at time intervals of 12, 24, 36, 48, and 60 h after feed animals with the formulation. Assays 1 and 2 showed a statistical difference (p < 0.01) by the F test when comparing the treated group with the control group. At the both assays, was observed in the treated group a reduction percentage of 74.18% and 88.38%, respectively. In assay 3, there was a statistical difference between the treated group and the control group at all collection times (p < 0.01). Regarding the collection periods, there was no statistical difference over time in the treatment group (p > 0.05). The results demonstrate that the fungal isolate AC001 formulated in rice bran can prey on L3 of Oesophagostomum spp., in vitro and after passing through the gastrointestinal tract, without loss of viability. Houve atividade ovicida (p<0,05) em relação ao grupo controle no décimo dia de interação e colonização interna dos ovos de 25,5%.
