O objetivo deste trabalho foi avaliar in vitro a ação do extrato bruto enzimático de Pochonia chlamydosporia (VC4) sobre ovos de Ancylostoma sp, em meio ágar-água 2% e em cultura de fezes. Observou-se um percentual de redução na eclosão dos ovos de Ancylostoma sp, de 76,8% na placas de Petri do grupo tratado em relação ao grupo controle. O extrato bruto enzimático de Pochonia chlamydosporia foi eficiente na redução da eclosão dos ovos de Ancylostoma sp, podendo ser utilizado como controlador biológico desse nematoide.Item Atividade larvicida do extrato bruto enzimático do fungo Duddingtonia flagras sobre larvas de primeiro estádio de Angiostrongylus vasorum(Revista da Sociedade Brasileira de Medicina Tropical, 2011-01-05) Braga, Fabio Ribeiro; Araujo, Juliana Milani; Tavela, Alexandre de Oliveira; Araújo, Jackson Victor de; Soares, Filippe Elias de Freitas; Geniêr, Hugo Leonardo André; Lima, Walter dos Santos; Mozzer, Lanuze Rose; Queiroz, José Humberto deAngiostrongylus vasorum é um nematóide que parasita cães domésticos e eventualmente o homem. O objetivo deste trabalho foi observar a atividade predatória in vitro do extrato bruto enzimático do fungo Duddingtonia flagrans sobre larvas de primeiro estádio A. vasorum em condições laboratoriais no meio ágar-água 2%. Ao final do experimento, os percentuais de redução das L1 de A. vasorum observados foram de: 53,5% (24h) e 71,3% (48h) O extrato bruto enzimático do fungo D. flagrans destruiu in vitro as L1, podendo ser utilizado como controle biológico desse nematóide.Item Efeitos da inclusão de farelo do resíduo de manga no desempenho de frangos de corte de 1 a 42 dias(Revista Brasileira de Zootecnia, 2008-05-30) Vieira, Patrícia Aparecida Fontes; Queiroz, José Humberto de; Albino, Luiz Fernando Teixeira; Moraes, George Henrique Kling de; Barbosa, Anderson de Almeida; Müller, Elisa Sialino; Viana, Maurício Tárcio dos SantosUm experimento foi conduzido com o objetivo de avaliar a utilização de farelo do resíduo de manga (FRM) (Mangifera indica L., var. Ubá) em rações para frangos de corte de 1 a 42 dias de idade. Utilizaram-se 600 pintos machos, Ross, distribuídos em delineamento inteiramente casualizado, com cinco tratamentos, cada um com seis repetições de 20 aves por unidade experimental. Os tratamentos consistiram da inclusão de 0,0; 2,5; 5,0; 7,5 ou 10,0% de farelo do resíduo de manga em dietas à base de milho e farelo de soja. Avaliaram-se o consumo de ração (CR), o ganho de peso (GP), a conversão alimentar (CA) e o índice de eficiência produtiva (IEP). No período de 1 a 42 dias de idade, não houve efeito dos níveis de farelo do resíduo de manga no consumo de ração. O ganho de peso e a conversão alimentar também não foram afetados pela inclusão de 2,5 e 5,0% de farelo do resíduo de manga na ração. A inclusão de até 5% de farelo do resíduo de manga na ração não afeta o desempenho de frangos no período de 1 a 42 dias de idade.Item Effect of dry coffee residues fermented with Monascus ruber on the metabolism of Apo E mice(Arquivos Brasileiros de Cardiologia, 2012-03-23) Brito, Larissa Froede; Queirós, Lívia Dias de; Peluzio, Maria do Carmo Gouveia; Ribeiro, Sônia Machado Rocha; Matta, Sérgio Luis Pinto da; Queiroz, José Humberto deAtherosclerosis is a chronic inflammatory disease of multifactorial origin, which occurs in response to endothelial injury. The fungus Monascus ruber has hypocholesterolemic activity, and the polyphenols present in coffee residue have an antioxidant activity and can help prevent cardiovascular diseases. Coffee residue has a significant amount of fermentable sugars, being an adequate substrate for growing fungi. The objective of this study was to assess the effect of dry coffee residue fermented with Monascus ruber on the lipid metabolism of Apo E knockout mice. The biological assay was performed with 30 Apo E knockout mice, divided into five groups and undergoing different treatments. The phytochemical prospection and quantification of phenolic compounds of the fermented and non-fermented coffee residues were performed. The sera of the animals were analyzed by using enzyme kits, and the aortic tissue was embedded in paraffin and stained with hematoxylin and eosin to undergo histopathological analysis. Comparing with the control group, the group receiving 2% non-fermented coffee residue showed a reduction of 42% in the serum levels of triacylglycerols and of approximately 41% in VLDL-c. The groups receiving 10% non-fermented coffee residue and 2% fermented coffee residue showed reductions in the lesion areas of 10.5% and 15.4%, respectively, as compared with the control group. The fermented coffee residue showed a higher content of phenolic compounds as compared with the non-fermented coffee residue. The present study showed that coffee residue fermentation has a potentially beneficial effect on cardiovascular diseases, especially atherosclerosis.Item Elaboration and characterization of Japanese Raisin Tree (Hovenia dulcis Thumb.) pseudofruits fermented alcoholic beverage(Food Science and Technology, 2017-12) Pinto, Juliana Tensol; Oliveira, Tânia Toledo de; Queiroz, José Humberto de; Dias, Disney Ribeiro; Schwan, Rosane Freitas; Oliveira, Diego Pinto de; Alvarenga, Luana FarahHovenia dulcis pseudofruits have underexplored properties for food purposes, despite their pleasant sensory characteristics and therapeutic benefits. The aim of this study was the elaboration and chemical characterization of the alcoholic fermented beverage of H. dulcis, using selected strain of Saccharomyces cerevisiae (CCMA 0200). The resulting fermented beverage presented high content of phenolic compounds and antioxidant activity when compared to other fruits and beverages (DPPH and ABTS assay). The alcohol content was 12.9 oGL and total sugars 3.57g/L. By the GC-MS analysis, 39 compounds were identified including metabolites with therapeutic potential such as eugenol, trans-farnesol salicylates. The flavonoid dihidromyricetin was identified and quantified (75.17 mg/L) by HPLC-DAD and UPLC-MS/MS. The results reinforce the interest on nutraceutical and functional properties of this beverage and opens perspectives for new studies that value this underexplored pseudofruit.Item Hydrolysis of galacto-oligosaccharides in soy molasses by α -galactosidases and invertase from Aspergillus terreus(Brazilian Archives of Biology and Technology, 2010-05) Reis, Angélica Pataro; Guimarães, Valéria Monteze; Ferreira, Joana Gasperazzo; Queiroz, José Humberto de; Oliveira, Maria Goreti Almeida; Falkoski, Daniel Luciano; Almeida, Maíra Nicolau de; Rezende, Sebastião Tavares deTwo α -galactosidase (P1 and P2) and one invertase present in the culture of Aspergillus terreus grown on wheat straw for 168 h at 28ºC were partially purified by gel filtration and hydrophobic interaction chromatographies. Optimum pH and temperatures for P1, P2 and invertase preparations were 4.5-5.0, 5.5 and 4.0 and 60, 55 and 65ºC, respectively. The KM app for ρ -nitrophenyl-α -D-galactopyranoside were 1.32 mM and 0.72 mM for P1 and P2, respectively, while the KM app value for invertase, using sacarose as a substrate was 15.66 mM. Enzyme preparations P1 and P2 maintained their activities after pre-incubation for 3 h at 50ºC and invertase maintained about 90% after 6 h at 55 ºC. P1 and P2 presented different inhibition sensitivities by Ag+, D-galactose, and SDS. All enzyme preparations hydrolyzed galacto-ologosaccharides present in soymolasses.Item In vitro and in silico characterization of a novel dextranase from Pochonia chlamydosporia(3 Biotech, 2018-03) Sufiate, Bruna; Araújo, Jackson Victor de; Queiroz, José Humberto de; Gouveia, Angélica S; Cardoso, Evandro; Moreira, Samara Silveira; Soares, Filippe Elias de Freitas; Braga, Fabio RibeiroThe objective of this study was to purify, characterize, and phylogenetically and structurally analyze the dextranase produced by the fungus Pochonia chlamydosporia. Dextranase produced by the fungus P. chlamydosporia was purified to homogeneity in two steps, with a yield of 152%, purification factor of 6.84 and specific activity of 358.63 U/mg. Its molecular weight was estimated by SDS-PAGE at 64 kDa. The enzyme presented higher activity at 50 °C and pH 5.0, using 100 mM citrate–phosphate buffer, was inhibited by Ag1+, Hg2+, Cu2+, Mg2+, and presented KM of 23.60 µM. Mature dextranase is composed of 585 amino acids residues, with a predicted molecular weight of 64.38 kDa and pI 5.96. This dextranase showed a strong phylogenetic similarity when compared to Trichoderma harzianum dextranase. Its structure consists of two domains: the first composed by 15 β strands, and the second composed by a right-handed parallel β-helix.Item Low-temperature clean-up method for the determination of pyrethroids in milk using gas chromatography with electron capture detection(Talanta, 2008-02-08) Goulart, Simone Machado; Queiroz, Maria Eliana L.R. de; Neves, Antônio Augusto; Queiroz, José Humberto deThis paper describes a new gas-chrormatography with electron capture detection (GC-ECD) method for determination of some pyrethroids in milk samples. The extraction of the pyrethroids was carried out by liquid–liquid extraction with clean-up by precipitation at low temperature, without additional stages for removal of fat interferences. The method was efficient with recoveries of 93.0 ± 0.1% for cipermethrin and 84.0 ± 0.3% for deltamethrin. The quantification limits were 0.75 μg L−1 for both pyrethroids. The method was simple, of easy execution, and used only small quantities of organic solvent. After optimization and validation, the method was used for the determination of residues of the pyrethroids cipermethrin and deltamethrin in milk and in lactea drink commercialized in Viçosa (MG, Brazil). Some samples presented contamination with deltamethrin at levels below the maximum contamination limits established by the FAO.Item Minimum cocktail of cellulolytic multi-enzyme complexes obtained from white rot fungi via solid-state fermentation(3 Biotech, 2018-01) Soares, Filippe Elias de Freitas; Queiroz, Paula Viana; Tavares, Gabriella Peterlini; Sufiate, Bruna Leite; Queiroz, José Humberto de; Cardoso, Wilton Soares; Santos, Fernando Almeida; Kasuya, Maria Catarina MegumiThe objective of this work was to optimize the total cellulase activity of the crude extract cocktails from five white rot fungi produced by solid-state fermentation, by means of the central composite design. The white rot fungi Pleurotus ostreatus PLO 06, Pleurotus eryngii PLE 04, Trametes versicolor TRAM 01, Pycnosporus sanguineus PYC 02 and Phanerochaete chrysosporium PC were tested. For optimization process aiming at the maximum value of total cellulase activity (FPAse), the multi-enzyme cellulase complexes (crude extracts) of each fungus were mixed simultaneously in different proportions. There was increase in FPAse activity for the cocktails formed by the extracts of the five fungi together, compared to the extracts of each fungus alone. The model presented the minimum cocktail of enzymes for maximum total cellulase activity, with 100.00 μL PYC; 100.00 μL PC; 100.00 μL PLO06; 100.00 μL PLE04 and 200 μL TRAM01. The maximum value found was of 304.86 U/L. The result of the cocktails was very relevant, showing that there is an enzymatic complementation in the extracts that should be further studied. Most fungi complexes had better hydrolysis rates compared with purified commercial enzyme.Item Nematicidal action of Pleurotus eryngii metabolites(Biocatalysis and Agricultural Biotechnology, 2017-10) Sufiate, Bruna Leite; Soares, Filippe Elias de Freitas; Moreira, Samara Silveira; Gouveia, Angélica de Souza; Monteiro, Thalita Suelen Avelar; Freitas, Leandro Grassi de Freita; Queiroz, José Humberto dePleurotus eryngii is one of the most cultivated and consumed mushroom species in North Africa, Europe and Asia. Fungi from Pleurotus genus have demonstrated nematophagous activities, however most of the literature reports are focused on Pleurotus ostreatus. The aim of this work was to evaluate P. eryngii action on Panagrellus sp. and to evaluate the effect of this fungus culture extract on Meloidogyne javanica eggs. P. eryngii fungus and its extract significantly reduced (p < 0.01) the number of intact Panagrellus sp. larvae after 24 h treatment in 60% and 90%, respectively. This effect is not related to enzymatic activity, but to the presence of other metabolites. M. javanica eggs, when treated with P. eryngii extract, showed a 53% reduction (p < 0.01) in the number of intact eggs. The M. javanica intact eggs reduction is attributed to enzymatic activity, once the extract showed proteolytic and chitinolytic activities of, respectively, 32.74 and 3.57 U/mL. P. eryngii fungus has predatory activity against Panagrellus sp. larvae due to toxins production and negatively affects M. javanica eggs and juveniles development due to chitinases and proteases production, evidencing this fungus potential to be used in biological control.Item Nematicidal activity of Paecilomyces marquandii proteases on infective larvae of Ancylostoma spp(Brazilian Archives of Biology and Technology, 2016-01) Genier, Hugo Leonardo André; Queiroz, José Humberto de; Braga, Fabio Ribeiro; Soares, Filippe Elias de Freitas; Araújo, Jackson Victor de; Pinheiro, Iara RebouçasThe present study aimed to evaluate the action of Paecilomyces marquandii proteases on Ancylostoma spp L3. White halos in the zymogram confirmed the proteolytic action. Results suggest that proteases present in E. milii latex concentrate samples are milin and miliin. Difference (p < 0.01) between larvae number in the different groups was found, with 65.59% and 96.46% Panagrellus redivivus larvae reduction after 24 and 48 h, respectively.Item The nematophagous fungus Monacrosporium thaumasium and its nematicidal activity on Angiostrongylus vasorum(Revista Iberoamericana de Micología, 2013-09-24) Soares, Filippe Elias de Freitas; Braga, Fabio Ribeiro; Araújo, Jackson Victor de; Lima, Walter dos Santos; Queiroz, José Humberto deThe dog acts as a reservoir and environmental disseminator of potentially zoonotic parasites. The objective of this work was to study the fungus Monacrosporium thaumasium regarding its nematicidal potential in laboratory trials and its proteolytic profile. The in vitro test was carried out through two assays (A and B). In assay A, conidia of the fungus N34a were added in positive coprocultures for Angiostrongylus vasorum. In assay B, crude extract (treated group) and distilled water (control group) were added to coprocultures. Next, the proteolytic profile of crude extract of the nematophagous fungus M. thaumasium (NF34a) was revealed by performing a zymogram. There was a reduction (p < 0.01) in the averages of larvae recovered from the treated groups (conidia and crude extract) in relation to control groups. The zymogram suggested that the nematophagous fungus M. thaumasium produces a protease of approximately 40 kDa. The results of this work confirm that the conidia as well as the crude extract of the fungus M. thaumasium may be used to control A. vasorum L1. The proteolytic profile suggested the presence of one protease (Mt1) of approximately 40 kDa that in the future may be used in biological control of L1 of this nematode.Item Optimization and validation of liquid–liquid extraction with low temperature partitioning for determination of carbamates in water(Analytica Chimica Acta, 2010-05-10) Goulart, Simone Machado; Alves, Renata Domingos; Neves, Antônio Augusto; Queiroz, José Humberto de; Assis, Tamires Condé de; Queiroz, Maria Eliana L.R. deUsing a 23 experimental design, liquid–liquid extraction with low temperature partitioning (LLE-LTP) was optimized and validated for analysis of three carbamates (aldicarb, carbofuran and carbaryl) in water samples. In this method, 2.0 mL of sample is placed in contact with 4.0 mL of acetonitrile. After agitation, the sample is placed in a freezer for 3 h for phase separation. The organic extract is analyzed by high performance liquid chromatography with ultraviolet detection (HPLC-UV). For validation of the technique, the following figures of merit were evaluated: accuracy, precision, detection and quantification limits, linearity, sensibility and selectivity. Extraction recovery percentages of the carbamates aldicarb, carbofuran and carbaryl were 90%, 95% and 96%, respectively. Even though extremely low volumes of sample and solvent were used, the extraction method was selective and the detection and quantification limits were between 5.0 and 10.0 μg L−1, and 17.0 and 33.0 μg L−1, respectively.Item Optimization of protease production by the fungus Monacrosporium thaumasium and its action againstAngiostrongylus vasorum larvae(Revista Brasileira de Parasitologia Veterinária, 2012-09-20) Soares, Filippe Elias de Freitas; Queiroz, José Humberto de; Braga, Fabio Ribeiro; Araújo, Jackson Victor de; Lima, Walter dos Santos; Mozzer, Lanuze RoseThe objectives of this study were to optimize protease production from the nematophagous fungus Monacrosporium thaumasium (NF34a) and evaluate its larvicidal activity and biological stability. An isolate of the nematophagous fungus Monacrosporium thaumasium (NF34a) was used to produce the enzyme. The Plackett-Burman design was used in order to scan which components of the culture medium could have a significant influence on protease production by the fungus NF34a. An in vitro assay was also performed to evaluate the larvicidal activity of NF34a. It was observed that only one component of the culture medium (yeast extract), at the levels studied, had any significant effect (p < 0.05) on protease production. There was a reduction (p < 0.01) in the mean number of larvae recovered from the treated groups, compared with the control groups. The results confirm previous reports on the efficiency of nematophagous fungi for controlling nematode larvae that are potentially zoonotic. Thus, given the importance of biological control, we suggest that further studies should be conducted on the protease produced by the fungus Monacrosporium thaumasium.Item Perfil biométrico, histomorfométrico e bioquímico no tratamento com atorvastatina cálcica de ratas com osteoporose induzida com dexametasona(Revista Brasileira de Ortopedia (English Edition), 2018-09) Ferreira Junior, Davilson Bragine; Pizziolo, Virgínia Ramos; Oliveira, Tânia Toledo de; Matta, Sérgio Luis Pinto da; Píccolo, Mayra Soares; Queiroz, José Humberto deAvaliar os efeitos da atorvastatina cálcica no tratamento da osteoporose induzida com dexametasona. A indução da osteoporose consistiu na administração de dexametasona na dose de 7,5 mg/kg de peso corporal, por via intramuscular, uma vez por semana durante quatro semanas, à exceção dos animais do grupo controle (G1). Os animais foram distribuídos nos seguintes grupos: G1 (grupo controle sem osteoporose), G2 (grupo controle com osteoporose sem tratamento), G3 (grupo controle com osteoporose tratado com alendronato de sódio 0,2 mg/kg) e G4 (grupo com osteoporose tratado com atorvastatina cálcica 1,2 mg/kg). Após 30 e 60 dias do início do tratamento dos animais, foram feitas as dosagens dos níveis séricos de fosfatase alcalina, fosfatase alcalina óssea, avaliação biométrica e histomorfométrica óssea. Em relação às análises biométricas e histomorfométricas, aos 60 dias de tratamento o G4 apresentou densidade óssea (índice Seedor), densidade trabecular óssea e espessura da cortical de 0,222 ± 0,004 g/cm, 59,167 ± 2,401% e 387,501 ± 8,573 μm, respectivamente, com diferença positiva, estatisticamente significativa (p < 0,05), em relação ao grupo G2. Aos 30 e 60 dias de tratamento, o G4 apresentou níveis séricos de fosfatase alcalina óssea estatisticamente significativos (p < 0,05) e superiores a todos os grupos (7,451 ± 0,173 μg/L e 7,473 ± 0,529 μg/L, respectivamente). O tratamento com atorvastatina cálcica demonstrou a capacidade desse fármaco de aumentar a atividade osteoblástica e a atividade reparadora tecidual óssea, atuar de forma diferente do alendronato de sódio, que demonstrou atividade preponderantemente antirreabsortiva.Item Proteolytic activity of the nematophagous fungus Arthrobotrys sinensis on Angiostrongylus vasorum larvae(BMC Research Notes, 2014-11-18) Soares, Filippe Elias de Freitas; Queiroz, José Humberto de; Braga, Fabio Ribeiro; Araújo, Jackson Victor de; Lima, Walter dos Santos; Zamprogno, Tatiana ToniniThe predatory nematophagous fungus Arthrobotrys sinensis (SF53) produces three proteases with nematicidal activity when grown on solid media culture. However, the proteolytic profile produced by this fungus, when grown in liquid culture medium remains unknown. Thus, the objective of this work was to evaluate the production of proteases from nematophagous fungus Arthrobotrys sinensis in liquid medium and its nematicidal activity on first stage larvae of A. vasorum. Proteases were obtained in its crude form, using Whatman no.1 filter paper, followed by centrifugation for 5 min at 10 × g and 4°C. A zymogram was performed with co-polymerized casein in an acrylamide gel as substrate. An in vitro assay to evaluate the nematicidal action of the proteases of A. sinensis (SF53) produced in liquid medium on A. vasorum L1 was conducted. By the analysis of the zymogram, it was observed a single halo at the beginning of digestion of the gel, suggesting that the three proteases of SF53 are produced in an enzymatic complex of large molecular weight. Regarding nematicidal activity, within 24 hours, the proteases produced in liquid medium of A. sinensis (SF53) showed a percentage reduction of 64% on the number of L1 of A. vasorum. In the present work, it is suggested that the three proteases of SF53 are produced in an enzymatic complex and was also demonstrated that these enzymes were effective in destroying A. vasorum L1.Item Serum lipid profile of broilers fed diets with inclusion of mango waste meal(Semina: Ciências Agrárias, 2016-06-16) Souza, Christiane Silva; Vieira, Patrícia Aparecida Fontes; Barbosa, Anderson de Almeida; Lima, Heder José D’Ávila; Fontes, Edimar Aparecida Filomeno; Vieira, Breno César; Oliveira, Maria Goreti de Almeida; Moraes, George Henrique Kling de; Queiroz, José Humberto de; Albino, Luiz Fernando TeixeiraWe evaluated the effect of including 2.5, 5.0, 7.5 and 10.0% mango waste meal (MWM) variety UBA in corn and soybean meal-based diet on the serum lipid profile of broilers. The experimental design was completely randomized, with five treatments and six replicates with 20 birds per experimental unit. Concentrations of creatinine, albumin, total protein, total cholesterol, triacylglycerols (TAG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and very-low-density lipoprotein cholesterol (VLDL-C), were evaluated at the ages of 14, 28 and 42 days, with 30 birds evaluated per age. At 14 days, there was no difference serum creatinine, total cholesterol, HDL, triacylglycerols, total proteins or VLDL-C concentrations as compared with control. Albumin concentration was the highest at the MWM inclusion levels of 5.0, 7.5 and 10.0%. At 28 days, triacylglycerols, VLDL-C and LDL-C were the lowestat 7.5% inclusion whereas at 42 days, these same variables were the lowest with addition of 10.0% meal. The variables studied in the Plackett–Burman design were the proteases and chitinases of AC001 or VC4 as destructive agents of S. westeri and trichostrongylides larvae, and P. fastosum eggs. All tested enzymes had a significant effect (P < 0.05) on the destruction of S. westeri larvae. Furthermore, only VC4 and AC001 proteases showed a significant effect (P < 0.05) on the destruction of trichostrongylides larvae. On the other hand, chitinases of VC4 showed the highest significance (P < 0.05) on the destruction of P. fastosum eggs. It is proposed that statistical planning for the use of enzymes derived from nematophagous fungi is a viable way to elucidate some questions about their mechanism of action.
