Item Avaliação de progênies de macaúba na fase juvenil e estimativas de parâmetros genéticos e diversidade genética(Pesquisa Florestal Brasileira, 2012-03-30) Manfio, Candida Elisa; Motoike, Sérgio Yoshimitsu; Resende, Marcos Deon Vilela de; Santos, Carlos Eduardo Magalhães dos; Sato, Aurora YoshikoA macaúba (Acrocomia aculeata) é uma palmeira oleaginosa e o extrativismo de seus frutos é de grande importância socioeconômica para os pequenos produtores rurais. O objetivo deste trabalho foi avaliar a variabilidade genética e obter informações sobre o controle genético de características morfológicas relacionadas ao crescimento e vigor da macaúba. Foram coletadas 51 progênies em Minas Gerais e São Paulo, as quais são integrantes do banco ativo de germoplasma da Universidade Federal de Viçosa. Doze meses após o plantio, essas progênies foram avaliadas quanto ao crescimento da planta (cm) e número de folhas emitidas. Estas características apresentaram grande variabilidade entre as progênies. As estimativas dos coeficientes de herdabilidade foram de 87% e 48%, para crescimento de planta e número de folhas, respectivamente, revelando controle genético e boas perspectivas para o melhoramento. O método de Tocher mostrou-se eficiente em alocar as progênies do Estado de São Paulo em um mesmo grupo. O método de seleção pelo Índice de rank médio permitiu selecionar indivíduos e progênies mais equilibrados em termos dos dois caracteres.Item Casca e coco de macaúba adicionados ao concentrado para vacas mestiças lactantes em dietas à base de silagem de milho(Revista Brasileir a de Agro pecuár ia Sustentável, 2012-07) Sobreira, Hugo Freitas; Lana, Rogério de Paula; Mancio, Antônio Bento; Fonseca, Dilermando Miranda da; Motoike, Sérgio Yoshimitsu; Silva, José Carlos Peixoto Modesto da; Guimarães, GeicimaraA possibilidade de uso dos resíduos gerados na produção de biodiesel pode ser uma ajuda valiosa na alimentação dos ruminantes, já que a quantidade de resíduos é grande; deste modo, a destinação final deve ocorrer de forma apropriada. Foi testada a viabilidade do uso da casca do coco e do coco triturado na alimentação animal, substituindo a ração constituída de milho e farelo de soja em 0, 0,4; 0,8 e 1,2 kg de um total de 2,5 kg/vaca/dia. O concentrado foi fornecido às vacas leiteiras no momento da ordenha, realizada duas vezes ao dia, e a silagem, uma vez, pela manhã. Oito vacas mestiças Holandês-Gir foram utilizadas, sendo distribuídas em dois quadrados latinos 4 x 4, com média de 11 kg de leite/dia. As análises químicas do leite foram feitas na Embrapa Gado de Leite em Juiz de Fora - MG. Foram realizadas pesagens da produção de leite e dos animais a cada término de período de 10 dias. Foi feita análise das variáveis em delineamento quadrado latino. O modelo estatístico incluiu efeitos de tratamento (níveis de substituição), quadrado latino (QL), animal/QL e período/ QL à 5% de probabilidade. Não foram verificadas diferenças na produção de leite. Os teores de lactose, extrato seco e extrato seco desengordurado apresentaram-se significativamente diferentes, porém a produção diária em kg/dia não. Assim a casca do coco e o coco de macaúba triturados podem ser utilizados em até 1,2 kg, de um total de 2,5 kg de concentrado/vaca/dia, sem afetar o desempenho de vacas mestiças produzindo aproximadamente 11 kg de leite/dia e recebendo silagem de milho como volumoso.Item Cloreto de clorocolina (CCC) influenciando a fertilidade das gemas da videira (Vitis vinifera) cv. Italia(Revista Ceres, 1996-01) Motoike, Sérgio Yoshimitsu; Cardoso, Antonio Américo; Casali, Vicente Wagner Dias;; Bruckner, Cláudio HorstVideiras do cultivar Itália (Piróvano 65), em experimento fatorial 2 x 5, foram tratadas com cinco pulverizações consecutivas, em intervalos de 15 dias, com duas doses de adubo foliar (0 e 4 g.l-1) e cinco níveis de CCC (0 , 50, 100, 150, 200 mg.l-1), a fim de se averiguar o efeito destes tratamentos no número de gemas férteis e na percentagem de brotação. O efeito do CCC na ausência do adubo foliar não foi significativo para o número de gemas férteis, apesar da tendência de aumento linear nas médias. Na presença do adubo foliar, o CCC apresentou efeito quadrático. Quanto à percentagem de brotação, o CCC, tanto na presença quanto na ausência do adubo foliar, não apresentou efeito significativo.Item Cross-species amplification and characterization of new microsatellite markers for the macaw palm, Acrocomia aculeata (Arecaceae)(Plant Genetic Resources, 2016-09) Mengistu, Fekadu Gebretensay; Motoike, Sérgio Yoshimitsu; Caixeta, Eveline Teixeira; Cruz, Cosme Damião; Kuki, Kacilda NaomiMicrosatellites or simple sequence repeats (SSRs) are useful molecular markers allowing for efficient conservation and sustainable use of genetic resources of plant species. Development of SSR marker system for new species is a very expensive task and time consuming. Crossspecies amplification of microsatellite loci is considered as a cost-effective approach for developing microsatellite markers for new species. The aim of this work was to examine the transferability of some SSR markers of two Arecaceae species (Astrocaryum aculeatum and Elaeis oleifera), in Acrocomia aculeata. Out of the total markers analysed 44% of the markers successfully amplified the genomic DNA in A. aculeata, of which 26% were polymorphic detecting a range of three to eight alleles with an average of 4.5 per locus. High average percentage of polymorphic loci (P 1⁄4 71.2%) per provenance was obtained within a range of 57–100% detecting genetic variation in A. aculeata germplasm collections. The polymorphic markers detected a positive inbreeding coefficient (F . 0) per locus revealing heterozygote deficiency in the accessions that were analysed. As the cross-amplification was at family level, in which the taxonomic distance is relatively wider between the sources (A. aculeatum and E. oleifera) and the target (A. aculeata) species, the amplification success was relatively low. However, the results are promising and implicated that high cross-amplification success could be achieved at species or genus level in A. aculeata. The markers will contribute towards the domestication of the potential macaw palm through realizing various studies such as population genetics, germplasm characterization, genetic improvement and conservation.Item Diversity and genetic relatedness among genotypes of Vitis spp. using microsatellite molecular markers(Revista Brasileira de Fruticultura, 2013-06-18) Leão, Patrícia Coelho de Souza; Cruz, Cosme Damião; Motoike, Sérgio YoshimitsuThe purpose of this research was to study the genetic diversity and genetic relatedness of 60 genotypes of grapevines derived from the Germplasm Bank of Embrapa Semiárido, Juazeiro, BA, Brazil. Seven previously characterized microsatellite markers were used: VVS2, VVMD5, VVMD7, VVMD27, VVMD3, ssrVrZAG79 and ssrVrZAG62. The expected heterozygosity (He) and polymorphic information content (PIC) were calculated, and the cluster analysis were processed to generate a dendrogram using the algorithm UPGMA. The He ranged from 81.8% to 88.1%, with a mean of 84.8%. The loci VrZAG79 and VVMD7 were the most informative, with a PIC of 87 and 86%, respectively, while VrZAG62 was the least informative, with a PIC value of 80%. Cluster analysis by UPGMA method allowed separation of the genotypes according to their genealogy and identification of possible parentage for the cultivars 'Dominga', 'Isaura', 'CG 26916', 'CG28467' and 'Roni Redi'.Item Early selection of elite clones of an ornamental bromeliad in vitro(Ciência Rural, 2010-07) Manfio, Candida Elisa; Motoike, Sérgio Yoshimitsu; Paula, Cláudio Coelho de; Valente, Magno Sávio; Melo, Cristiane GamaranoOrthophytum grossiorum is a typical bromeliad from Atlantic forestry threatened of extinction. The objectives of this research were to select O. grossiorum clones with ornamental values easy to propagate in vitro, and establish in vitro propagation protocols for these clones. The project was developed in three steps: germination and in vitro selection of seedlings responsive to BAP (6-benzylaminopurine), selection of clones with ornamental values, and establishment of protocol for in vitro propagation of the selected clones. In the first step only 18.33% of plantlets germinated in vitro were responsive to BAP. These plantlets were selected and replicated in vitro several times, each replicated plantlet constituting a clone. In the second step these clones were established ex vitro and surveyed for ornamental attributes. Five out of 11 clones were selected in this step. These clones presented distinct phenotypic traits and were considered of high ornamental quality. In the third step a protocol for in vitro propagation was developed for each selected clone.Item Ecophysiological traits of the macaw palm: A contribution towards the domestication of a novel oil crop Author links open overlay panel(Industrial Crops and Products, 2013-01) Pires, Thiago Pereira; Souza, Elma dos Santos; Kuki, Kacilda Naomi; Motoike, Sérgio YoshimitsuAcrocomia aculeata, or macaw palm, is a highly productive oleaginous palm tree with the potential to be a new oil crop. Plant productivity is influenced by the exchange of gases between the leaf and the atmosphere. The assimilation of CO2 is dependent on both environmental factors and the intrinsic characteristics of the leaf, such as age, position in the canopy, and nutrient and pigment content. This study was conducted to characterize some ecophysiological aspects of macaw palm cultivated under field conditions. Foliar content and gas exchange parameters were analyzed in relation to diel variation, light intensity, and the position within the canopy and rachis. The evolution of photosynthesis and related variables during the daytime followed the standard patterns for most species with C3 metabolism, reaching peak levels during the morning hours. The light curve response also displayed a C3 pattern; however, this species demonstrated a high photosynthetic capacity with a maximum net photosynthesis and apparent quantum efficiency of approximately 23 μmol m−2 s−1 and 0.07 mol/mol, respectively. Gas exchange parameters of the fronds along the canopy varied according to the leaf position and the leaflet's insertion location within the rachis. Overall values indicate that the second and third leaves and the leaflets in the middle of the rachis have the best physiological capacity. Foliar pigment and mineral content were relatively stable among the leaves, which is a pre-requirement for long lasting leaves.Item Efeito dos reguladores de crescimento dicamba e picloram na embriogênese somática em Eucalyptus grandis(Revista Árvore, 2007-02-04) Titon, Miranda; Xavier, Aloisio; Otoni, Wagner Campos; Motoike, Sérgio YoshimitsuEste trabalho objetivou avaliar o efeito das auxinas dicamba e picloram na indução de embriogênese somática em embriões zigóticos maduros, cotilédones e hipocótilos de Eucalyptus grandis. Os calos foram induzidos em meio de cultura MS contendo dicamba (0,25; 0,5; 1,0; e 2,0 mg L-1) ou picloram (0,5; 1,5; 5,0; e 10,0 mg L-1). Nos tratamentos com 0,5 mg L-1 de dicamba ou 5,0 e 10,0 mg L-1 de picloram, em explantes cotiledonares, foram observadas estruturas semelhantes a embriões somáticos em diferentes estádios de desenvolvimento. A análise histológica confirmou tratar-se de estruturas independentes, com um sistema vascular fechado, evidenciando se tratar de embriões somáticos.Item Effect of nitrogen and potassium rates on early development of macaw palm(Revista Brasileira de Ciência do Solo, 2015-06-29) Pimentel, Leonardo Duarte; Bruckner, Claudio Horst; Martinez, Hermínia Emília Prieto; Motoike, Sérgio Yoshimitsu; Manfio, Candida Elisa; Santos, Rafael Carlos dosThe economic exploitation of macaw palm [Acrocomia aculeate(Jacq.) Lodd. ex Mart.] is currently in transition, from extractivism to agricultural cultivation, thus requiring studies on the fertilization of the crop. This study evaluated the response of three genotypes of macaw palm to increasing rates of nitrogen and potassium, grown in the field until the 2nd year and to establish reference contents of mineral nutrients in the leaf. The experiment was a split-plot randomized block design with five main treatments (N and K rates) and three secondary treatments (genotypes), with three replications, each plot containing three plants. Plant height, leaf number, vigor, and nutrient contents in leaf tissues were evaluated at the end of 2nd year of cultivation. Differential responses were observed among genotypes, indicating that some genotypes are more efficient in the use of mineral inputs. There was a differentiated and positive response to increasing side-dressed N and K rates in the vegetative development of macaw genotypes until the 2nd year of field cultivation, indicating variability in the species in terms of nutrient use efficiency. The N and K fertilization rate corresponding to 360 g N + 480 g K2O per plant, in four split applications over the two years of cultivation, was insufficient to induce maximum vegetative development in the three macaw genotypes. There was no variation in macro- and micronutrient contents in leaf dry matter of the three macaw genotypes.Item Enzyme activity and reserve mobilization during Macaw palm ( Acrocomia aculeata ) seed germination(Acta Botanica Brasilica, 2016-07) Motoike, Sérgio Yoshimitsu; Borges, Eduardo Euclydes de Lima e; Guimarães, Valéria Monteze; Ataíde, Glauciana da Mata; Bicalho, Elisa MontezeReserve mobilization in seeds occurs after visible germination, which is marked by the protrusion of the radicle or cotyledonary petiole, as in species of Arecaceae. Acrocomia aculeata (macaw palm), usually produces hard seeds whose endosperm has mannan-rich cell walls. We investigated the composition of storage compounds in macaw palm seed and the roles of two enzymes (endo-β-mannanase, α-galactosidase) during and after germination. The seeds were firstly submitted to pre-established protocol to overcome dormancy and promote germination. Enzyme activity in both embryo and endosperm were assayed from the initiation of germinative activities until leaf sheath appearance, and the status of seed structures and reserve compounds were evaluated. Protein content of the embryo decreased with the initiation of imbibition while the lipid content began decreasing six days after removal of the operculum. Increases in enzyme activity and starch content were both observed after visible germination. We suggest that endo-β-mannanase and α-galactosidase become active immediately at germination, facilitating haustorium expansion and providing carbohydrates for initial seedling development. Protein is the first storage compound mobilized during early imbibition, and the observed increase in the starch content of the haustorium was related to lipid degradation in that organ and mannan degradation in the adjacent endosperm.Item Estimation of genetic parameters for in vitro oil palm characteristics (Elaeis guineensis Jacq.) and selection of genotypes for cloning capacity and oil yield(Industrial Crops and Products, 2015-12-23) Corrêa, Thais Roseli; Motoike, Sérgio Yoshimitsu; Coser, Sara Morra; Silveira, Gustavo da; Resende, Marcos Deon Vilela de; Chia, Gilson SanchezOil palm has stood out among the promising species for biofuel production and plant improvement through cloning superior individuals is the technology that contributes most to increase oil production. However, there are no studies on the genotypes behavior regarding both in vitro performance and yield potential simultaneously. By this mean, the objective of this paper was to study the genetic control of the related characteristics to the oil palm cloning process, and to identify responsive genotypes within a collection of 32 elite materials of a commercial planting in order to select the superior genotypes for the formation of a clonal garden. Callogenesis and production of embryogenic lines, which are main characteristics related to the cloning process, presented genetic control, verified through estimates of genetic parameters: heritability, coefficient of relative variation, and selective accuracy; also indicating efficiency in the selection of superior genotypes within the evaluated set. Genotypes A-13, A-14, A-18, A-20 and A-21 were selected as superior genotypes for both characteristics, such as oil yield and in vitro performance, due to formation of embryogenic lines. This has been the first study investigating the genetic control of the cloning capacity aiming to the selection of genotypes for a clonal garden formation.Item Fruit development, growth, and stored reserves in macauba palm (Acrocomia aculeata), an alternative bioenergy crop(Planta, 2016-06-16) Montoya, Sebastián Giraldo; Motoike, Sérgio Yoshimitsu; Kuki, Kacilda Naomi; Couto, Adriano DonatoMain conclusion Macauba palm fruiting is supra-annual, and the fruit growth follows a double sigmoidal trend. The prevailing compound in the mesocarp differs as the fruit ages, oil being the major storage compound. Acrocomia aculeata, macauba palm, is a conspicuous species in the tropical Americas. Because the species is highly productive in oil-rich fruits, it is the subject of domestication as an alternative vegetable oil crop, especially as a bioenergy feedstock. This detailed study first presents the macauba fruit growth and development patterns, morphological changes and accumulation of organic compounds. Fruits were monitored weekly in a natural population. The fruiting was supra-annual, and the fruit growth curve followed a double sigmoidal trend with four stages (S): SI—slow growth and negligible differentiation of the fruit inner parts; SII—first growth spurt and visible, but not complete, differentiation of the inner parts; SIII—growth slowed down and all structures attained differentiation; and SIV—second growth spurt and fruit maturation. In SII, the exocarp and endocarp were the main contributors to fruit growth, whereas the mesocarp and endosperm were responsible for most of the weight gain during SIV. In comparison with starch and oil, soluble sugars did not accumulate in the mesocarp. However, starch was transitory and fueled the oil synthesis. The protective layers, the exocarp and endocarp, fulfilling their ecological roles, were the first to reach maturity, followed by the storage tissues, the mesocarp, and endosperm. The amount and nature of organic compounds in the mesocarp varied with the fruit development and growth stages, and oil was the main and final storage material. The description of macauba fruit’s transformations and their temporal order may be of importance for future ecological and agronomical references.Item Genetic diversity in table grapes based on RAPD and microsatellite markers(Pesquisa Agropecuária Brasileira, 2011-08-05) Leão, Patrícia Coelho de Souza; Motoike, Sérgio YoshimitsuThe objective of this work was to analyze the genetic diversity of 47 table grape accessions, from the grapevine germplasm bank of Embrapa Semiárido, using 20 RAPD and seven microsatellite markers. Genetic distances between pairs of accessions were obtained based on Jaccard's similarity index for RAPD data and on the arithmetic complement of the weighted index for microsatellite data. The groups were formed according to the Tocher's cluster analysis and to the unweighted pair‑group method with arithmetic mean (UPGMA). The microsatellite markers were more efficient than the RAPD ones in the identification of genetic relationships. Information on the genetic distance, based on molecular characteristics and coupled with the cultivar agronomic performance, allowed for the recommendation of parents for crossings, in order to obtain superior hybrids in segregating populations for the table grape breeding program of Embrapa Semiárido.Item Genetic parameters and selection of macaw palm ( Acrocomia aculeata) accessions: an alternative crop for biofuels(Crop Breeding and Applied Biotechnology, 2018-07) Costa, Annanda Mendes; Motoike, Sérgio Yoshimitsu; Corrêa, Thais Roseli; Silva, Thaís Cristina; Coser, Sara Morra; Resende, Marcos Deon Vilela de; Teófilo, Reinaldo FranciscoThis study aimed to estimate the parameters related to the genetic control of the physical characteristics of the fruits, oil content, and oil yield, and to proceed with the selection of macaw palm accessions. Forty-four macaw palm accessions of the active germplasm collection of the Federal University of Viçosa were collected for the evaluation of epicarp dry matter, pulp dry matter, endocarp dry matter, kernel dry matter, oil content, and oil yield. Narrow-sense individual heritability estimates were considered as of intermediate magnitude. The coefficient of repeatability and the accuracy in family selection was of high magnitude. Oil yield per plant presented the highest coefficient of individual genetic variation. Five different accessions contributed to the ten first individuals selected by the individual BLUP. The Mulamba and Mock’s rank index allowed classifying the accessions in a sequence favorable to selection.Item Germinação in vitro de sementes de citrus afetada por tratamentos mecânicos(Revista Ceres, 2008-07) Mendes, Rita de Cássia; Oliveira, Márcio Antônio Rocha de; Motoike, Sérgio Yoshimitsu; Alexandre, Rodrigo Sobreira; Siqueira, Dalmo LopesO presente estudo trabalho objetivou mostrar o efeito de tratamentos mecânicos aplicado a sementes de quatro espécies cítricas, Citrus sunki Hort. Ex Tan., C. limonia Osb., C. reshni Hort. Ex Tan. e Poncirus trifoliata, e do híbrido C. paradisi Macf. x P. trifoliata, para melhorar a germinação in vitro. Os tratamentos mecânicos consistiram em remoção do tegumento da semente e divisão do endosperma ao meio. Os parâmetros avaliados foram a porcentagem de germinação, o índice de velocidade de germinação, o comprimento de plântulas e o número de plântulas por semente. Foi concluído que tratamentos mecânicos aplicados às sementes são benéficos para germinação in vitro de C. paradisi x P. trifoliata, C. reshni e P. trifoliata. Os tratamentos melhoraram a porcentagem de germinação de semente, bem como o índice de velocidade de germinação e o comprimento de plântulas destas espécies. A divisão do endosperma provou ser o melhor tratamento aplicado às sementes e, portanto, o indicado por melhorar a germinação in vitro destas espécies. Porém, C. sunki e C. limonia apresentaram bom desempenho de germinação in vitro sem qualquer tratamento, mostrando que estas espécies não requerem tratamento para germinação.Item Manejo da irrigação e fertirrigação com nitrogênio sobre as características químicas da videira ‘Niágara Rosada’(Ciência Rural, 2011-07) Busato, Cristiani Campos Martins; Soares, Antonio Alves; Sediyama, Gilberto Chohaku; Motoike, Sérgio Yoshimitsu; Reis, Edvaldo Fialho dosA qualidade da uva pode responder ao manejo da irrigação e ao ajuste da adubação nitrogenada. Assim, este trabalho teve como objetivo estudar o efeito do manejo de irrigação e doses de nitrogênio sobre a qualidade da uva ‘Niagara Rosada’ no município de Colatina-ES. Foram avaliadas três doses de nitrogênio, aplicadas via fertirrigação: 35, 70 e 140g planta -1 e quatro estratégias de manejo de irrigação. No início do experimento até a fase de maturação das bagas, todos os tratamentos receberam a mesma lâmina de irrigação, elevando-se a umidade do solo à capacidade de campo (M1=100%). Após este período, foram avaliados mais três estratégias de manejo de irrigação: M2=67%, M3=33% e M4=0% da lâmina correspondente ao tratamento M1. Foram avaliados os teores de sólidos solúveis (SS), pH e acidez titulável (AT). Os resultados indicaram que os tratamentos sob déficit hídrico (M3 e M4), nas doses de 70 e 140g planta - 1 apresentaram maiores teores de SS e pH das bagas e diminuição dos valores de AT.Item Organogênese in vitro a partir de gemas apicais e axilares de plantas adultas de orquídeas do grupo Cattleya(Revista Ceres, 2002-11) Ventura, Gizella Machado; Dias, José Maria Moreira; Teixeira, Sílvio Lopes; Carvalho, Virgínia Silva; Motoike, Sérgio Yoshimitsu; Novais, Roberto Ferreira de; Cecon, Paulo RobertoO cultivo in vitro de ápices caulinares e gemas axilares é importante para uma rápida e eficiente propagação clonal. O objetivo deste trabalho foi determinar a capacidade morfogênica de ápices caulinares e de gemas axilares de Laelia lobato 'Jenni' x L labota 'Alba', identificando o melhor tipo de explante para este híbrido. Ápices caulinares e gemas axilares foram retirados de brotações e bases de pseudobulbos de plantas adultas, após desinfestação. Estes explantes foram inoculados em meio de cultivo liquido com a formulação de Murashige e Skoog (1962), contendo 0,2 mg L^-1 de cinetina e 1,0 mg L^-1 de ANA. Após 50 dias e já com formação de estruturas, os explantes foram transferidos para meio de mesma composição, porém sólido, com carvão ativado e sem fitorreguladores. Os tratamentos foram três tipos de explantes: 24 ápices caulinares (AC), 19 gemas axilares de brotações (GB) e 10 gemas axilares de pseudobulbos (GP). O experimento foi montado no delineamento inteiramente casualizado. Avaliaram-se a porcentagem de estruturas formadas, o aspecto fisiológico e a área final dos explantes. Os explantes GB foram os que apresentaram maior porcentagem de estruturas formadas (47,4%), seguidos dos GP (20%) eAC (12,5%). O grau de oxidação dos explantes foi avaliado como totalmente oxidado (71,7%), oxidado com ápice verde (7,5%), verde com extremidade oxidada (7,5%) e verde expandido (13,2%). Os explantes GB apresentaram maior área final aos 50 dias de cultivo. Embora os três tipos de explantes apresentem alta porcentagem de oxidação, todos mostram boa capacidade morfogênica, principalmente as gemas axilares de brotações, potencializando uma técnica para propagação em escala comercial dessa espécie.Item Quebra de dormência de sementes da videira cv. niágara rosada sem estratificação(Revista Ceres, 2010-03) Val, Aurinete Daienn Borges do; Motoike, Sérgio Yoshimitsu; Alvarenga, Eveline Mantovani; Cecon, Paulo RobertoO objetivo deste trabalho foi verificar o efeito de três fatores na germinação de sementes de uva cv. niágara rosada: tratamentos físicos, tratamentos químicos e ambientes de germinação. Os tratamentos físicos consistiram de sementes com corte transversal na região mediana e de sementes com corte na micrópila. Os tratamentos químicos foram aplicados mediante imersão das sementes inteiras em solução aquosa de ácido giberélico (GA3) nas concentrações de 0 e 4000 mg L-1. Os ambientes avaliados foram in vitro e ex vitro. O experimento foi um fatorial do tipo 2 x 2 x 2 no delineamento em blocos ao acaso, com quatro repetições, sendo a parcela experimental constituída de um Gerbox®, contendo 25 sementes ou uma grade com 25 tubos de ensaio. Os parâmetros avaliados foram porcentagem e velocidade de germinação das sementes. Os resultados indicaram que o corte das sementes é imprescindível à germinação de sementes não estratificadas. A combinação entre cortes na micrópila, 4000 mg L-1 de GA3 e ambiente in vitro, proporcionou a germinação de 77% das sementes, sendo o ambiente in vitro significativamente superior ao ex vitro.Item Repetibilidade em características biométricas do fruto de macaúba(Ciência Rural, 2010-10-25) Manfio, Candida Elisa; Motoike, Sérgio Yoshimitsu; Santos, Carlos Eduardo Magalhães dos; Pimentel, Leonardo Duarte; Queiroz, Vanessa de; Sato, Aurora YoshikoEstimativas do coeficiente de repetibilidade têm sido utilizadas no estudo de caracteres de várias espécies de palmeiras, auxiliando na definição do número e período adequado de avaliações dos genótipos para maior eficiência dos programas de melhoramento. Para macaúba, não há relatos de estudos dessa natureza. Nesse contexto, faz-se importante a determinação do coeficiente de repetibilidade das seguintes características biométricas dos frutos: peso do fruto, volume do fruto, diâmetro equatorial do fruto, peso da amêndoa, volume da amêndoa, diâmetro da amêndoa e espessura do endocarpo; e a determinação do número mínimo de avaliações para um eficiente processo de seleção e avaliação de subamostras de macaúba. O trabalho foi desenvolvido no Departamento de Fitotecnia da Universidade Federal de Viçosa, analisando-se 145 subamostras de macaúba, provenientes de coletas realizadas nos estados de Minas Gerais, São Paulo, Mato Grosso do Sul, Pará, Maranhão e Pernambuco. Houve diferença significativa para todas as características analisadas e observou-se concordância nas magnitudes dos coeficientes de repetibilidade de cada característica, obtidos pelos diferentes métodos. No entanto, os coeficientes de repetibilidade variam entre as oito características, de 0,68 a 0,99, observados para a característica espessura do endocarpo e volume da amêndoa, respectivamente. As estimativas dos coeficientes de determinação obtidas a partir das 10 repetições para as oito características e pelos quatro métodos foram superiores a 95%. A avaliação de quatro frutos é suficiente para compreensão das características estudadas das subamostras de macaúba com 90% de acurácia.Item Somatic embryogenesis in macaw palm (Acrocomia aculeata) from zygotic embryos(Scientia Horticulturae, 2009-02-17) Moura, Elisa Ferreira; Motoike, Sérgio Yoshimitsu; Ventrella, Marı́lia Contin; Sá Júnior, Adauto Quirino de; Carvalho, MychelleMacaw palm (Acrocomia aculeata) is an oleaginous palm tree that is highly productive and adapted to semiarid ecosystems, which oil can be used to produce biodiesel. Such characteristics make macaw palm a potential crop to be used by farmers from semi-arid regions, but its propagation is still problematic. This paper reports the first description of somatic embryogenesis for macaw palm from zygotic embryos. The explants were cultured on Y3 medium and different combinations of plant growth regulators. After 60 days of culture, embryogenic callus were induced with 9 μM of 4-amino-3,5,6-trichloropicolonic acid (picloram) or 2,4-dichlorophenoxyacetic acid (2,4-D), combined or not with 1 μM N-phenyl N′1,2,3 thidiazol-5-yl urea (TDZ). Naphthoxy acetic acid (NOA) and 4-chlorophenoxyacetic acid (CPA) did not generate embryogenic callus. Somatic embryos were only obtained when embryogenic callus were induced with 9 μM picloram and then subcultured for 120 days on the same medium with 3.0 g l^−1 activated charcoal. Anatomical sections showed that somatic embryos had a typical protoderm, procambial strands and an apical meristem. When transferred to Y3 medium +3.0 g l^−1 activated charcoal, without growth regulators, half of the somatic embryos germinated, but only a few completed the germination.
