Item Genetic variability of Ceratocystis fimbriata on Tectona grandis and Carapa guianensis in Brazil(Universidade Federal de Viçosa, 2016-10-27) Valdetaro, Denise Cristina de Oliveira Franco; Alfenas, Acelino Couto; http://lattes.cnpq.br/3842072851787460Recently, Ceratocystis fimbriata Ellis & Halsted was reported in Tectona grandis L. f (teak) and Carapa guianensis Aubl. (andiroba) in Brazil. In teak, the fungus causes death and wood discoloration, which affects the production of high valuable timber. In andiroba, the fungus infects and kills seeds and seedlings, which may affect negatively its natural regeneration and, probably, its biodiversity. The identification of C. fimbriata on these hosts was based only on morphological characteristics and analysis of ITS rDNA sequences. Furthermore, the genetic and physiological variability of C. fimbriata populations as well as their phylogenetic relationships with other isolates and species of Ceratocystis were not analyzed . Such studies are essential to support disease management strategies. Therefore, this study aimed to: 1) Assess the relationship among isolates of the fungus from both host species with isolates from other hosts of the Latin American Clade (LAC) by using mating type genes and ITS rDNA region. 2) Assess the interesterility through crossings between isolates from different host species. 3) Determine the genetic variability in populations of C. fimbriata through microsatellite markers and their physiological especialization by f cross-inoculation experiments. Out of 24 isolates of the fungus obtained from teak from of locations in the state of Mato Grosso, and of 73 isolates of C. fimbriata from andiroba in the states of Acre, Roraima and Amazonas, new ITS haplotypes and mating types identified were distributed among other isolates within the LAC and reside in a single group that includes isolates from sweet potato, from which the species was originally described. Teak and andiroba isolates were inter fertile with other strains of C. fimbriata, showing that this is a single biological species of C. fimbriata. Microsatellite analysis of the fungal population on teak showed low genetic variability. In cross-inoculations, although teak plants presented relatively small lesions, two isolates differed from the control. The eucalyptus isolates were not pathogenic on teak. The populations of C. fimbriata from andiroba of the three states (AC, AM, and RR) showed 14 genotypes and intermediary levels of genetic variability compared to other native populations of the fungus in Brazil. Cross-inoculations of andiroba, teak, eucalyptus and mango isolates suggest that the isolates of C. fimbriata from these species are host specific and that there is variability in aggressiveness among isolates of the fungus from andiroba.Item Morfologia, variabilidade genética e patogenicidade de Ceratocystis fimbriata em Hevea brasiliensis(Universidade Federal de Viçosa, 2012-07-16) Valdetaro, Denise Cristina de Oliveira Franco; Ferreira, Maria Alves; http://lattes.cnpq.br/5181939734384805; Alfenas, Acelino Couto; http://lattes.cnpq.br/2514320654462590; http://lattes.cnpq.br/3842072851787460; Pereira, Olinto Liparini; http://buscatextual.cnpq.br/buscatextual/visualizacv.do?id=K4767879D4; Harrington, Thomas CharlesEspécies do complexo Ceratocystis fimbriata sensu lato (s.l.) são importantes patógenos de um grande número de plantas arbóreas e herbáceas, dentre elas a seringueira (Hevea sp.), nativa da região amazônica e pertencente a família Euphorbiaceae. A seringueira é cultivada visando a produção do látex, a principal matéria prima para obtenção da borracha natural que é utilizada em diversos setores. Ceratocystis fimbriata infecta o painel da seringueira, causando uma doença conhecida como mofo cinzento que afeta negativamente a produção de látex. Ceratocystis fimbriata s.l. considerado, atualmente, como um complexo de espécies contendo várias espécies crípticas, muitas das quais ainda não descritas. Deste modo, objetivou-se com este trabalho, determinar a morfologia, o relacionamento filogenético e especificidade de hospedeiro do fungo causador do mofo cinzento em seringueira, de modo a confirmar se é uma nova espécie do complexo fimbriata. Para isso, foram utilizados 20 isolados de seringueira para análise de variabilidade genética. Dentre estes, quatro isolados foram selecionados para análises morfológicas e filogenéticas e destes, dois foram utilizados para testes de patogenicidade. De acordo com as sequências de ITS r-DNA analisadas, os isolados de C. fimbriata de seringueira pertencem ao clado da América Latina, com dois genótipos de ITS, de acordo com a origem geográfica, sendo eles originados do Estado do Acre e o outro do Estado da Bahia, também confirmados por análises de microssatélites. As inoculações mostraram que não há especialização, quanto ao hospedeiro, uma vez que foram patogênicos a diferentes espécies hospedeiras (manga, crotalária, acácia, eucalipto e kiwi). Portanto, de acordo com os resultados deste trabalho, os isolados de seringueira pertencem ao clado da América Latina e são confirmados como C. fimbriata e não uma espécie distinta.
