Item Biological control of eucalyptus bacterial wilt with rhizobacteria(Biological Control, 2015-01) Santiago, Thais R.; Rossato, Mauricio; Romeiro, Reginaldo S.; Mizubuti, Eduardo S. G.; Grabowski, CristhianThe antagonistic potential of 298 rhizobacteria obtained from the rhizosphere and rhizoplane of tomato and eucalyptus plants was assessed for the control of bacterial wilt of eucalyptus caused by Ralstonia solanacearum. Several tests were performed using tomato plants as a screening system to select efficient rhizobacteria. Different methods for antagonist delivery and pathogen inoculation were evaluated: (1) seeds were microbiolized (soaked for 12 h in a suspension of the antagonist propagules) and germinated seedlings had their roots immersed in the pathogen inoculum suspension; (2) seedlings originated from microbiolized seeds were transplanted to soil infested with R. solanacearum and (3) roots of seedlings were immersed in a suspension of propagules of the antagonist and subsequently in a suspension of R. solanacearum. Nine isolates (UFV-11, 32, 40, 56, 62, 101, 170, 229, and 270) were selected as potential antagonists to R. solanacearum as they suppressed bacterial wilt in at least one of the methods assessed. The selected antagonists were evaluated against two isolates of R. solanacearum using in vitro and in vivo (inoculated eucalyptus) tests. Isolates UFV-56 (Bacillus thuringiensis), UFV-62 (Bacillus cereus) and a commercial formulation of several rhizobacteria (Rizolyptus®) suppressed bacterial wilt in eucalyptus protecting the plants during the early stages of development.Item Efficiency of phylloplane bacteria in controlling aerial tomato diseases under field conditions(Summa Phytopathologica, 2008-01) Halfeld-Vieira, Bernardo A.; Romeiro, Reginaldo S.; Mounteer, Ann; Mizubuti, Eduardo S.G.The capacity of two bacteria isolated from the tomato phylloplane to control late blight (Phytophthora infestans) was investigated in the field, and compared against the effectiveness of spraying with the fungicide chlorothalonil (1.5 g a.i. L-1 ) or water (control). A 55% reduction in late blight intensity was observed in the leaves of the middle of the plant and 62% in those of the upper leaves when using the antagonist UFV-STB 6 ( Novosphingobium capsulatum ) as compared to the control. Isolate UFV-IEA 6 (Bacillus cereus) was able to reduce disease intensity by 55%, but only in the upper leaves of the tomato plants. Treatment with isolate UFV-STB 6 also led to a significant reduction in the percentage of fruits with late blight symptoms. The results demonstrate the potential of these two bacteria in controlling this disease.Item Evidence that the biocontrol agent Bacillus cereus synthesizes protein that can elicit increased resistance of tomato leaves to Corynespora cassiicola(Tropical Plant Pathology, 2010-01) Romeiro, Reginaldo S.; Lanna Filho, Roberto; Macagnan, Dirceu; Garcia, Flávio A. O.; Silva, Harllen S. A.Isolate UFV-101 of Bacillus cereus was selected in previous studies for promoting growth inducing resistance in plants. In a previous study, supernatant from cultures of the microorganism in a liquid medium was found to induce resistance in tomato foliage against the pathogens Pseudomonas syringae pv. tomato, Xanthomonas vesicatoria, Alternaria solani and Corynespora cassiicola. In the present work the microorganism was grown in a minimal medium for 48 h and the cells precipitated for centrifugation. The supernatant was concentrated by lyophilization, dialyzed in a 12 kDa cut-off point membrane and fractioned in column containing Sephadex G25 balanced in PBS buffer. The fractions corresponding to a protein peak were applied to tomato seedlings. After four days leaflets were collected and inoculated with the pathogen. C. cassiicola. The numbers of lesions produced by the pathogen on leaflets exposed to the bacterial supernatant were similar to those exposed to acibenzolar-S-methyl but fewer than in those treated with water. It is concluded that the supernatant contained protein which induced resistance in the tomato leaves against C. cassiicola.Item Isolation and selection of Hemileia vastatrix antagonists(European Journal of Plant Pathology, 2014-04-21) Haddad, Fernando; Saraiva, Rodrigo M.; Mizubuti, Eduardo S. G.; Romeiro, Reginaldo S.; Maffia, Luiz A.Organic coffee growing is rapidly increasing in Brazil, and many diseases, especially coffee leaf rust (Hemileia vastatrix), are threatening its production. This study is a first step towards a biocontrol program for coffee rust on organically grown plants. In three organic coffee farms in the state of Minas Gerais, 393 microbial strains including 154 bacterial and 239 fungal strains were isolated from leaves, leaf debris, and soil samples, and in 6 month-old coffee plants, 17 of these isolates reduced both the infection frequency (IF) and the number of H. vastatrix urediniospores produced per leaf (UPL) by more than 70 %. The isolates were identified as eight bacteria isolates, seven Bacillus spp. and one Pseudomonas sp., and nine fungal isolates, four Fusarium spp., two Penicillium spp., one Aspergillus sp., one Acremonium sp. and one Cladosporium sp. Each fungal and bacterial isolate was applied 0, 4, 8, 12 or 16 days before and 0, 4, 8, 12 or 16 days after H. vastatrix inoculation, and the efficiency in reducing both IF and UPL was evaluated. The efficiency was higher and lasted longer when the bacterial isolates were applied before H. vastatrix inoculation. Six Bacillus (B10, B25, B143, B157, B171, B175), two Fusarium (F205, F281), and one Pseudomonas (B286) isolates are potentially efficient as biocontrol agents of H. vastatrix and will be tested using field experiments.Item Métodos de isolamento de bactérias do filoplano de tomateiro visando populações específicas e implicações como agentes de biocontrole(Fitopatologia Brasileira, 2004-11) Halfeld-Vieira, Bernardo A.; Romeiro, Reginaldo S.; Mizubuti, Eduardo S. G.Em um processo de seleção de bactérias do filoplano de tomateiro (Lycopersicon esculentum) com potencial para o controle de doenças da parte aérea da cultura, diferentes métodos de isolamento foram utilizados visando obter isolados da população total, da população da superfície foliar e isolados que habitam locais protegidos do filoplano e/ou que toleram fatores de estresse. Foi testada a capacidade de 300 isolados em controlar in vivo, as doenças causadas por Alternaria solani, Pseudomonas syringae pv. tomato e Phytophthora infestans. Os testes foram repetidos para cada um dos antagonistas selecionados, estudando-se, também, a capacidade de controlar a mancha-bacteriana, causada por Xanthomonas vesicatoria. Os resultados demonstraram haver predomínio de antagonistas provenientes de folíolos do terço superior da planta de população total ou da superfície. Entretanto, o único antagonista selecionado, isolado de folíolos do terço inferior, foi obtido de locais protegidos do filoplano e/ou capaz de tolerar fatores de estresse.
