Item Caracterização das populações de Phytophthora infestans das regiões Sul e Sudeste do Brasil(Universidade Federal de Viçosa, 2001-07-10) Reis, Ailton; Mizubuti, Eduardo Seiti Gomide; http://lattes.cnpq.br/9332397384594058Os eventos de migração de Phytophthora infestans, agente etiológico da requeima ou mela do tomateiro e da batateira, trouxeram implicações importantes para o manejo da doença em vários países. Entretanto, pouco se sabe sobre a atual estrutura da população deste patógeno no Brasil. Assim, este trabalho teve como objetivo a caracterização das populações de P. infestans das regiões Sul e Sudeste do Brasil, principais regiões produtoras de batata e tomate. Isolados foram coletados e caracterizados para os seguintes marcadores: grupo de compatibilidade, aloenzimas Gpi e Pep, RFLP do DNA genômico (com a sonda RG 57 ) e haplotipos mitocondriais. Uma parte dos isolados (258) foi submetida a testes de sensibilidade ao fungicida metalaxyl (empregando-se três métodos) e outra parte a testes de virulência em discos de folha de diferenciadoras de raças de P. infestans em batata (genes R) e tomate (genes Ph). Todos os isolados de tomate foram do grupo A1 e a maioria (94%) apresentou genótipos 86/100 e 892/100 para Gpi e Pep, respectivamente; e 81% dos isolados apresentaram fingerprinting e haplotipos mitocondriais típicos da linhagem clonal antiga US-1. Os isolados de batata, na sua maioria (87%), eram do grupo A2, apresentavam genótipos 100/100 para Gpi e Pep, e fingerprinting e haplotipos mitocondriais típicos de BR-1. Foram encontrados isolados resistentes, intermediários e sensíveis ao fungicida metalaxyl, tanto na linhagem clonal US-1, quanto em BR-1, em todos os estados das regiões Sul e Sudeste do Brasil. Foi também encontrada grande diversidade de patótipos nas duas linhagens clonais e nas duas regiões amostradas. Muitos destes patótipos apresentaram alta complexidade. Os isolados da linhagem BR-1 parecem ser adaptados a batateira e os da linhagem US-1 ao tomateiro. Os resultados sugerem que não há ocorrência de reprodução sexuada na população de P. infestans amostrada, há grande variação quanto a resistência ao fungicida metalaxyl e quanto a virulência dos isolados.Item Caracterização de isolados de Phytophthora infestans do Distrito Federal e de Goiás(Fitopatologia Brasileira, 2006-05) Ribeiro, Fabiana H. G.; Reis, AiltonForam caracterizados 123 isolados de Phytophthora infestans obtidos de 21 lavouras de tomateiro e oito de batateira, em municípios do Estado de Goiás e Cidades Satélites de Brasília, no período de abril de 2001 a setembro de 2003. Os isolados foram caracterizados para os marcadores grupo de compatibilidade (123 isolados); isoenzima glucose 6-fosfato-isomerase (Gpi) (34 isolados) e resistência aos fungicidas mefenoxam (77 isolados) e metalaxyl (32 isolados de batateira), usando o método de disco de folhas. Todos os 78 isolados de tomateiro foram classificados no grupo de compatibilidade A1, enquanto os 45 de batateira foram do grupo A2. Os fenótipos para Gpi dos isolados de tomateiro (19) e de batateira (15) foram 86/100, típico da linhagem clonal US-1, e 100/100, típico da linhagen clonal BR-1, respectivamente. Quanto à resistência a mefenoxam, constataram-se isolados de tomateiro resistentes (36%), intermediários (48%) e sensíveis (16%). A maioria dos isolados de batateira foi classificada como sensível (82%) e apenas 9% de intermediários e resistentes. Dos isolados de batateira avaliados para resistência ao metalaxyl, 25% foram resistentes, 62% intermediários e 13% sensíveis. A população de P. infestans no Distrito Federal e no Estado de Goiás é constituída de duas linhagens clonais, com especificidade por hospedeiro.Item Characterization of isolates of Phytophthora infestans from southern and southeastern Brazil from 1998 to 2000(Plant Disease, 2003-08) Reis, Ailton; Smart, Christine D.; Fry, William E.; Maffia, Luiz A.; Mizubuti, Eduardo S. G.The population of Phytophthora infestans in Brazil was first characterized 12 years ago. In this research, isolates of P. infestans from potato (n = 184) and tomato (n = 267) collected in southern and southeastern Brazil were characterized to provide more detailed analysis of the current structure of the population. All 451 isolates were analyzed for mating type, and subsets of the isolates were analyzed for allozymes, restriction fragment length polymorphism fingerprint, mtDNA haplotypes, and metalaxyl resistance. Tomato isolates were all of A1 mating type, mtDNA Ib, and US-1 genotype or some variant within this clonal lineage. Of the potato isolates, 82% were A2 mating type, mtDNA IIa, BR-1 genotype, which is a new lineage of P. infestans. All A2 isolates were found on potato, whereas 91% of the A1 isolates were from tomato. A1 and A2 isolates were never found in the same field. The frequency of resistance to metalaxyl was higher in isolates from tomato (55%) than in isolates from potato (38%). After more than a decade of coexistence of isolates of the A1 and A2 mating types, the population was highly clonal, dominated by the BR-1 and US-1 clonal lineages.Item Fusarium udum revisited: a common, but poorly understood member of the Fusarium fujikuroi species complex(Mycological Progress, 2019-02) Abreu, Lucas M.; Pfenning, Ludwig H.; Melo, Maruzanete Pereira de; Costa, Marileide Moreira; Reis, Ailton; Cabral, Cleia Santos; Lima, Cristiano S.Fusarium udum is the causal agent of a wilt disease on pigeon pea (Cajanus cajan) in tropical regions. This species shares morphological characteristics with F. oxysporum, leading to misidentification when the diagnosis is solely based on morphological markers. The sexual stage of this fungus was observed on stems of Cajanus in India and was formally described as Gibberella indica. In Brazil, a wilt disease is reported on Crotalaria, but the etiological agent has not been identified correctly so far. In this study, we tested the hypothesis that the causal agent of a wilt on Crotalaria belongs to the same species pathogenic to C. cajan. Strains obtained from diseased Crotalaria spp. were characterized through molecular phylogeny of TEF, TUB and RPB2, laboratory crosses, morphological markers, and pathogenicity tests. Strains from Crotalaria from Brazil formed a well-supported clade with F. udum strains from Crotalaria and Cajanus from other countries. Strains from Brazil intercrossed among themselves and also with others from all geographic regions, and formed fertile perithecia, defining a distinct mating population inside the Fusarium fujikuroi species complex. One strain obtained from Cajanus cajan in India is indicated as epitype, and female-fertile tester strains of both mating types were selected for the identification of field isolates through sexual crosses. These results confirm that the species associated with wilt disease on Crotalaria and Cajanus is F. udum. Wilt symptoms caused by F. udum in Cr. ochroleuca are described and illustrated.Item Genetic structure of the population of Alternaria solani in Brazil(Journal of Phytopathology, 2011-04) Lourenço Jr, Valdir; Rodrigues, Tatiana T. G.Understanding the genetic structure of the population of Alternaria solani (AS) is an important component of epidemiological studies of early blight, a severe disease that affects potato (Po) and tomato (To) worldwide. Up to 150 isolates obtained from both hosts were analysed with RAPD and AFLP markers to estimate the amount and distribution of genetic variability of AS in Brazil. Using RAPD, gene diversity (h=0.20) and scaled indices of diversity of Shannon (H′=0.66) and Stoddart and Taylor’s (G=0.31) for the Po population were higher than those of the To (h=0.07, H′=0.34, G=0.17). For AFLP, the statistics for the Po (h=0.17, H′=0.86, G=0.49) and To (h= 0.17, H′=0.85, G=0.36) populations were similar. For each RAPD and AFLP locus, the allele frequency for the overall population ranged from 0.006 to 0.988, and 0.007 to 0.993, respectively. Genetic differentiation was high (GST=0.41 and θ=0.59) and moderately high (GST=0.23 and θ=0.37) when estimated with RAPD and AFLP, respectively. Based on cluster analyses, there was strong evidence of association of pathogen haplotypes with host species. The null hypothesis of random association of alleles was rejected in the analysis of both RAPD (IA=13.1, P<0.001) and AFLP (IA=2.2, P<0.001) markers. The average number of migrants was estimated to be around one and two individuals per generation, using RAPD and AFLP, respectively. There was no correlation between genetic distance and geographical origin of AS haplotypes for RAPD (r=−0.07, P=0.84) and AFLP (r=−0.03, P=0.70). The AS population is clonal with high genetic variability, and there is genetic differentiation between the populations that affect To and Po.
