Item Deterioração por umidade na pré-colheita de sementes de soja: alterações físicas, fisiológicas e bioquímicas(Universidade Federal de Viçosa, 2019-08-06) Pinheiro, Daniel Teixeira; Dias, Denise Cunha Fernandes dos Santos; http://lattes.cnpq.br/7263173009787619A soja é uma das culturas mais importantes do agronegócio mundial e cresce a demanda de sementes de alta qualidade. A deterioração por umidade é um dos fatores que mais afeta a qualidade das sementes e a avaliação deste fator com base em aspectos fisiológicos, bioquímicos e físicos pode ajudar a elucidar estes processos. O objetivo do trabalho foi avaliar as alterações fisiológicas, bioquímicas e físicas em sementes de soja submetidas à deterioração por umidade na fase pré-colheita. Seis cultivares de soja (BMX APOLO, BMX 6563, NS 5959, NA 5909, BMX POTÊNCIA e TMG 1175) foram produzidas em casa de vegetação e submetidas à deterioração por umidade por meio de um sistema de precipitação, aplicando-se 0; 60 (30+30); 120 (60+60) e 180 (90+90) mm de precipitação quando as plantas atingiram o estádio R8. Após a colheita, as sementes foram avaliadas quanto à germinação, vigor, estresse oxidativo, metabolismo proteico e teste de raios X. A deterioração causou reduções de até 30 pontos percentuais na germinação, além de reduções no vigor, atividade de enzimas antioxidativas e teor de proteínas, principalmente nos maiores níveis de precipitação. Observou-se também a deterioração por umidade na fase de pré-colheita ocasionou acúmulos significativos de peróxido de hidrogênio, malonaldeído e atividade da protease, caracterizando de maneira geral, o estresse oxidativo. Com exceção dos danos por umidade que foram altamente correlacionados com a qualidade fisiológica, a densidade foi pouco alterada pela deterioração. Em geral, as cultivares BMX 6563 e BMX POTÊNCIA foram mais suscetíveis e NA 5909 e TMG 1175 mais tolerantes à deterioração por umidade na fase de pré-colheita. Palavras-chave: Germinação. This study aimed to investigate the strategies used by rice plants under different divalent and trivalent sources of iron excess. Rice plants from a lowland and upland cultivar were grown in nutrient solution with toxic concentrations of ferrous or ferric iron. A mineral nutrient quantification and anatomical analysis were performed on leaves and roots. Physiological damage was assessed by leaf photochemical parameters and lipid peroxidation. Expression levels of genes related to iron homeostasis were analyzed. More pronounced nutritional deficiencies, oxidative stress and physiological damage were observed in plants exposed to toxic levels of ferrous iron. Ferritin expression increased in leaves of both cultivars under ferrous or ferric iron excess. We showed that sulfate iron was more toxic to the two rice cultivars even though this iron source was less translocated in the plant. Trivalent iron complexed to citrate is easily translocated through rice plants, but it is less toxic than the divalent iron. Rice plants are able to cope with this iron overload by keeping photosynthetic apparatus working properly.Item Effect of cyanide by sodium nitroprusside (SNP) application on germination, antioxidative system and lipid peroxidation of Senna macranthera seeds under saline stress(Journal of Seed Science, 2019-01) Silva, Aparecida Leonir da; Pinheiro, Daniel Teixeira; Borges, Eduardo Euclydes de Lima e; Silva, Laércio Junio da; Dias, Denise Cunha Fernandes dos SantosThe effects of NO donors on germination under saline stress have been much investigated for many species, however, there are reports that the effect caused by donors are effects of cyanide present. The aim of this study was to evaluate the effects of sodium nitroprusside (SNP) on germination, antioxidative system and lipid peroxidation of Senna macranthera seeds under saline stress. The osmotic potentials of -0.4 and -0.5 MPa of NaCl were used, as well as the concentration of 100 μM of sodium nitroprusside, inactive sodium nitroprusside. Germination rate, imbibition curves, antioxidant enzyme activity, lipid peroxidation, and protein content were evaluated. Similar effects were observed for sodium nitroprusside and inactive sodium nitroprusside, indicating that these effects were related to the release of the cyanide present in sodium nitroprusside. The sodium nitroprusside (SNP) improves the germination of Senna macranthera seeds under salt stress conditions, through release of cyanide. The cyanide favored germination, reducing lipid peroxidation and increasing the activity of the antioxidant enzymes.Item Estresse salino no potencial fisiológico de sementes e no desenvolvimento vegetativo de melão (Cucumis melo L.)(Universidade Federal de Viçosa, 2015-07-17) Pinheiro, Daniel Teixeira; Dias, Denise Cunha Fernandes dos Santos; http://lattes.cnpq.br/7263173009787619A salinidade é citada como o principal fator abiótico limitante do crescimento e da produtividade de plantas. De maneira geral, causa redução do potencial osmótico do meio e efeitos tóxicos, causando distúrbios metabólicos em todos os estádios de crescimento e desenvolvimento, comprometendo eventos fisiológicos que vão desde a germinação à produção. Os mecanismos de tolerância ao estresse salino em plantas e sementes ainda não estão suficientemente compreendidos, fazendo com que o progresso no desenvolvimento de culturas tolerantes à salinidade seja lento. O objetivo do trabalho foi avaliar os efeitos do estresse salino no potencial fisiológico de sementes e no desenvolvimento vegetativo de melão (Cucumis melo L.). Para todas as avaliações, foram utilizadas sementes de melão amarelo Diplomata F1. O estresse salino reduziu a germinação de sementes e o desenvolvimento de plântulas de melão, sendo mais crítico a partir do potencial de -0,2 MPa. De maneira geral, as atividades das enzimas antioxidativas SOD, CAT e POX aumentaram com a indução do estresse. O estresse salino reduz a emergência de plântulas de melão, mesmo em porcentagens de sódio trocável (PST) consideradas mais baixas, como em 20%. O aumento da PST no solo reduz emergência, comprimento e massa seca de plântulas de melão, principalmente em valores superiores a 20%. O híbrido Diplomata F1 se mostrou mais sensível ao estresse salino na emergência de plântulas do que no desenvolvimento vegetativo ao longo do tempo.Item Germination and antioxidant action in melon seeds exposed to salt stress(Pesquisa Agropecuária Tropical, 2016-09) Pinheiro, Daniel Teixeira; Silva, Aparecida Leonir da; Silva, Laércio Junio da; Sekita, Marcelo Coelho; Dias, Denise Cunha Fernandes dos SantosSalinity influences all germination stages and may adversely affect the seedlings establishment in the field. This study aimed at assessing the effects of salt stress on the physiological potential of melon (Cucumis melo L.) seeds. Germination and activity of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POX) enzymes were assessed at the osmotic potentials of 0.0 MPa, -0.1 MPa, -0.2 MPa, -0.3 MPa, -0.4 MPa and -0.5 MPa. The enzymatic activity was assessed after 24 h, 48 h and 72 h of imbibition. Seed germination was zero at -0.5 MPa and 91 % and 85 % at -0.1 MPa and -0.2 MPa, respectively. In general, the activity of SOD, CAT and POX enzymes increased with the salt-induced stress, in the different times of imbibition, and these increases were more evident at -0.4 MPa and -0.5 MPa. The salt stress induced by NaCl diminishes the germination rate of melon seeds, particularly from the osmotic potential of -0.2 MPa. There is a significant increase in the activity of SOD, CAT and POX enzymes, mainly after 48 h of exposure.Item Salinity tolerance in Senna macranthera (DC. ex Collad.) H. S. However, there are no precise reports about which compound present in sodium nitroprusside would cause these effects in seeds under salt stress. The aim of this study was to evaluate the effects of SNP on germination of Senna macranthera seeds under salt stress. The osmotic potentials of -0.3, -0.4, and -0.5 MPa of NaCl were used, as well as the concentration of 100 μM of sodium nitroprusside, inactive sodium nitroprusside, and ferrocyanide. The sodium nitroprusside in germination was applied before, during, and after salt stress. Germination rate, germination speed index, percentage of normal seedlings, and seedling development components were evaluated. The treatments that proved to be more promising in recovery of germination under salt stress are -0.4 MPa together with SNP; -0.4 MPa together with inactive SNP; -0.5 MPa together with SNP and -0.5 MPa together with inactive SNP. Effects similar to those brought about by the sodium nitroprusside treatment were also observed in the treatments with inactive sodium nitroprusside and ferrocyanide, indicating that the effects observed were related to release of the cyanide present in the sodium nitroprusside.
