Item Differential physiological responses in rice upon exposure to excess distinct iron forms(Plant and Soil, 2015-06) Müller, Caroline; Kuki, Kacilda Naomi; Pinheiro, Daniel Teixeira; Souza, Laíse Rosado de; Silva, Advânio Inácio Siqueira; Loureiro, Marcelo Ehlers; Oliva, Marco Antonio; Almeida, Andréa MiyasakaRice can be cultivated in highlands, which can expose it to iron deficiency, or under irrigation, which can lead to iron toxicity and lower productivity. This study aimed to investigate the strategies used by rice plants under different divalent and trivalent sources of iron excess. Rice plants from a lowland and upland cultivar were grown in nutrient solution with toxic concentrations of ferrous or ferric iron. A mineral nutrient quantification and anatomical analysis were performed on leaves and roots. Physiological damage was assessed by leaf photochemical parameters and lipid peroxidation. Expression levels of genes related to iron homeostasis were analyzed. More pronounced nutritional deficiencies, oxidative stress and physiological damage were observed in plants exposed to toxic levels of ferrous iron. Ferritin expression increased in leaves of both cultivars under ferrous or ferric iron excess. We showed that sulfate iron was more toxic to the two rice cultivars even though this iron source was less translocated in the plant. Trivalent iron complexed to citrate is easily translocated through rice plants, but it is less toxic than the divalent iron. Esse efeito foi revertido com ácido 2-cloroetilfosfônico, composto liberador de etileno, e ácido 1-carboxílico-1-aminociclopropano (ACC), precursor imediato do etileno em sua rota biossintética. Observou-se um atraso na produção do etileno em sementes não-dormentes e uma redução significativa na produção desse hormônio em sementes dormentes tratadas com PBZ. Sementes não-dormentes tratadas com o inibidor apresentaram níveis significativamente menores de ACC livre, sendo o mesmo também observado em sementes dormentes tratadas com PBZ mais selenouréia. A atividade in vivo da oxidase do ACC foi diminuída em sementes dormentes e, em sementes não-dormentes, observou-se uma atividade decrescida nas primeiras 48 h de exposição ao inibidor. A atividade in vitro da oxidase do ACC mostrou-se diminuída a partir de 36 h de incubação em sementes não-dormentes tratadas com o PBZ, ocorrendo posteriormente à diminuição da germinação.Item Guard cell-specific upregulation of sucrose synthase 3 reveals that the role of sucrose in stomatal function is primarily energetic(New Phytologist, 2015-10-05) Daloso, Danilo M.; Williams, Thomas C. Fernie, Alisdair R.Isoform 3 of sucrose synthase (SUS3) is highly expressed in guard cells; however, the precise function of SUS3 in this cell type remains to be elucidated. Here, we characterized transgenic Nicotiana tabacum plants overexpressing SUS3 under the control of the stomatal-specific KST1 promoter, and investigated the changes in guard cell metabolism during the dark to light transition. Guard cell-specific SUS3 overexpression led to increased SUS activity, stomatal aperture, stomatal conductance, transpiration rate, net photosynthetic rate and growth. Although only minor changes were observed in the metabolite profile in whole leaves, an increased fructose level and decreased organic acid levels and sucrose to fructose ratio were observed in guard cells of transgenic lines. Furthermore, guard cell sucrose content was lower during light-induced stomatal opening. In a complementary approach, we incubated guard cell-enriched epidermal fragments in 13C-NaHCO3 and followed the redistribution of label during dark to light transitions; this revealed increased labeling in metabolites of, or associated with, the tricarboxylic acid cycle. The results suggest that sucrose breakdown is a mechanism to provide substrate for the provision of organic acids for respiration, and imply that manipulation of guard cell metabolism may represent an effective strategy for plant growth improvement.Item Nutritional efficiency of Eucalyptus clones under water stress(Revista Brasileira de Ciência do Solo, 2017-11-09) Müller, Caroline; Hodecker, Bárbara Elias Reis; Merchant, Andrew; Barros, Nairam Félix dePlant health and nutrition are strongly influenced by the availability of water. The objective of this study was to evaluate the different patterns of initial growth and nutritional efficiency of ten drought-stressed Eucalyptus clones. Water stress conditions were induced by osmotic stress, by gradual addition of polyethylene glycol 6000 over a 20-day period. Drought stress significantly reduced the dry matter of leaves, roots, and the whole plant as well as the nutritional efficiency of most clones. Clone PL040 was the least efficient in nutrient uptake and use and the most sensitive to drought stress. Interestingly, under drought stress, the drought-tolerant clones generally had high AE (absorption efficiency), but low nutrient UE (use efficiency), whereas the sensitive clones had low AE and low UE for root formation and high AE for leaf formation. No entanto, a disponibilidade deste elemento para as plantas varia com as condições do solo e sistema de cultivo. Oryza sativa pode ser cultivado em dois sistemas distintos: irrigado e sequeiro. Em solos inundados, devido as condições anaeróbicas e de baixo pH, o ferro pode ser reduzido tornando-se altamente disponível. A absorção excessiva de ferro pode gerar toxidez nas plantas de arroz ocasionando perda de produtividade. Desta forma, o presente trabalho objetivou estabelecer uma relação entre os mecanismos fotoquímicos, bioquímicos, moleculares e de dissipação de energia em diferentes cultivares de arroz que permitam realizar uma seleção de cultivares tolerantes ao ferro. A utilização de citrato férrico (Fe+3) resultou em maior translocação deste metal para as folhas das plantas de arroz. No entanto, o ferro na forma reduzida (Fe+2) apresentou maior toxidez. O excesso de Fe+2 ocasionou reduções drásticas nas taxas fotossintéticas em diferentes cultivares de arroz. A queda fotossintética foi acompanhada pela redução na taxa de transporte de elétrons e aumento na dissipação regulada de energia. Esses parâmetros apresentaram-se sensíveis ao excesso de Fe+2 na solução nutritiva de cultivo. A redução na fotossíntese deveu-se a limitações na fixação do carbono pelas plantas de arroz expostas ao excesso de ferro. Danos decorrentes do estresse por Fe+2 ocasionaram um reparo ineficiente da proteína D1 do fotossistema II, observado pela diminuição na expressão do gene OspsbA. As plantas de arroz aumentaram a expressão do gene OsFER1 em uma tentativa de minimizar o excesso de ferro nos componentes celulares. Além dos danos fisiológicos, o excesso de ferro afetou a absorção e translocação de outros minerais, reduzindo o teor de P e Mg nas folhas. Os parâmetros fisiológicos de trocas gasosas e fluorescência da clorofila a mostraram-se sensíveis aos efeitos tóxicos do ferro.
