Abstract
O objetivo deste trabalho foi avaliar a influência da idade da planta no isolamento de protoplastos de mesofilo de Passiflora suberosa L. Os protoplastos foram obtidos de plantas crescidas em condições de casa de vegetação mediante a utilização de uma mistura enzimática constituída dos sais do meio CPW, manitol a 13% ((p/v), Cellulase R-10 a 1,0% (p/v), Macerozyme R-10 a 0,2% (p/v), Driselase a 0,1% (p/v), 5,0 mM de tampão MES, Cefotaxima a 250 mgl-1e PVP-10 a 1% (p/v). A idade da planta teve influência marcante sobre o rendimento e a viabilidade dos protoplastos obtidos. Foi observado decréscimo significativo entre o rendimento e a viabilidade dos protoplastos com o aumento da idade das plantas doadoras de explantes
The aim of this work was to evaluate the influence of age of donor plants on the protoplast isolation of Passiflora suberosa L. Protoplasts were obtained from glasshouse-grown seedlings leaves using a _pectocellulolytic enzyme mixture containing CPW salts, 13% (w/v) mannitol, 1% (w/v) Cellulase R-10, 0,2% (w/v) Macerozyme R-10, Driselase 0.l% (w/v), 5.0 mM MES, 250 mgl-1 Cefotaxime and 1% (w/v) PVP-10. Age (30, 60 and 90 days after germination) of the plants, from which mesophyll protoplasts were isolated had & marked effect on protoplast yield and viability. A significant decrease in both was observed, with the increase of age.
