Perfil oxidativo em cães submetidos a procedimentos cirúrgicos de esterilização

Abstract
O ato cirúrgico é um evento que resulta em inúmeras respostas orgânicas e está relacionado a produção de espécies reativas, que em desequilíbrio com o sistema antioxidante contribui para o estresse oxidativo. Os efeitos deletérios do estresse oxidativo, resultantes do dano oxidativo, podem comprometer funções sistêmicas assim como a recuperação pós-cirúrgica. O estudo objetiva avaliar o nível dos marcadores de estresse oxidativo e do sistema enzimático de defesa antioxidante no soro de caninos, estabelecendo um perfil oxidativo no perioperatório de esterilização. A pesquisa foi realizada com cães hígidos de raças, idades e portes variados, provenientes da rotina cirúrgica do Hospital Veterinário da Universidade Federal de Viçosa/Minas Gerais. As técnicas cirúrgicas escolhidas para realização da castração em machos foi a orquiectomia e em fêmeas a ovariohisterectomia. O experimento foi realizado após a aprovação do Comitê de Ética em Pesquisa com Animais da Universidade Federal de Viçosa (CEUA-UFV), protocolo n°15/2019 e autorização dos tutores. Todos os caninos foram submetidos a coletas sanguíneas em momentos distintos, T0 coleta realizada no momento da realização dos exames pré-cirúrgicos, T1 coleta realizada no dia da cirurgia com o animal em jejum pré-operatório antes da tricotomia e cateterização e T2 a coleta foi realizada 01 hora após o procedimento cirúrgico. A partir do soro foram realizadas avaliações espectrofotométricas para determinação da capacidade antioxidante total, atividade das enzimas Catalase (CAT), superóxido dismutase (SOD) e Glutationa-S-transferase (GST) e quantificação dos marcadores de dano proteínas carboniladas e produto de lipoperoxidação. O perfil oxidativo sérico dos cães machos e fêmeas, antes de qualquer interferência, apresentava-se em níveis similares. Cães machos apresentaram redução significativa da atividade das enzimas CAT e GST após a finalização do procedimento de castração em comparação ao tempo T1. Apenas a enzima GST apresentou redução significativa da atividade, no grupo de cães fêmeas após o procedimento de castração. Embora não tenha sido observado alteração no marcador MDA, resultante da peroxidação lipídica, nos diferentes tempos avaliados, houve redução significativa da carbonilação de proteínas em T1 e T2 no grupo de cães fêmeas. Nossos achados demonstram que os procedimentos cirúrgicos para esterilização de cães machos e fêmeas induzem alterações oxidativas. Apenas as fêmeas apresentaram redução de carbonilação de proteínas, 3 demonstrando o impacto do procedimento cirúrgico no perfil oxidativo de fêmeas durante a castração. The surgical act is an event that results in numerous organic responses and is related to the production of reactive species, which, in imbalance with the antioxidant system, contribute to oxidative stress. The deleterious effects of oxidative stress, resulting from oxidative damage, may compromise systemic functions as well as postoperative recovery. The study object to evaluate the level of oxidative stress markers and the enzymatic system of antioxidant defense in canine serum, establishing an oxidative profile in the perioperative sterilization. The research was conducted with healthy dogs of varying breeds, ages and sizes, from the surgical routine of the Veterinary Hospital of the Federal University of Viçosa/Minas Gerais. The surgical techniques chosen to perform castration were orchiectomy in males and ovariohysterectomy in females. The experiment was carried out after the approval of the Animal Research Ethics Committee at the Federal University of Viçosa (CEUA-UFV), n°15/2019 protocol and authorization of the owners. All canines were submitted to blood collections at different times, T0 collection performed at pre-surgical examinations, T1 collection performed on the day of the surgery with the animal fasting preoperatively before trichotomy and catheterization and the T2 collection was performed 01 hour after the surgical procedure. From the serum, spectrophotometric evaluations were performed to determine the total antioxidant power, activity of the enzymes Catalase (CAT), superoxide dismutase (SOD) and Glutathione-S- transferase (GST) and quantification of the markers of damage carbonyl protein and lipid peroxidation product. The serum oxidative profile of male and female dogs, before any interference, was at similar levels. Male dogs showed a significant reduction in the activity of CAT and GST enzymes after the end of the castration procedure compared to the T1 time. Only the GST enzyme showed a significant reduction in activity in the group of female dogs after the castration procedure. Although there was no alteration in the MDA marker, resulting from lipid peroxidation, at the different times evaluated, there was a significant reduction in protein carbonylation at T1 and T2 in the group of female dogs. Our findings demonstrate that surgical procedures for sterilization of male and female dogs induce oxidative changes. Only females showed reduction of protein carbonylation, demonstrating the impact of the surgical procedure on the oxidative profile of females during castration. 3 Keywords: Ovariohysterectomy.
