Avaliação da atividade biológica de produtos naturais e sintéticos visando o tratamento de feridas ocasionadas por leishmaniose tegumentar: estudo in vitro e in silico

Resumo
A Leishmania. V. braziliensis é o agente etiológico da Leishmaniose Tegumentar (LT) doença que afeta milhares de pessoas, principalmente aquelas em estado de vulnerabilidade social. O tratamento normalmente que é utilizado para tratar essa doença é baseado em fármacos que possuem toxicidade e custos elevados. Sendo assim, a busca de compostos químicos é essencial para o seu controle e tratamento. A busca de compostos oriundos de produtos naturais, advindos da flora ou fauna ou sintéticos através da química medicinal é de grande importância. Neste contexto, o presente trabalho tem como objetivo avaliar o potencial de alcaloides indólicos e derivados de dibenzoilmetanos como cicatrizante e leishmanicida.tal, foram feitos testes in vitro para avaliar a citotoxicidade em células de macrófagos RAW264. Os compostos que obtiveram uma taxa de viabilidade maior que 80% foram selecionados para os demais testes como atividade anti-inflamatória, atividade antioxidante para avaliar a capacidade de proteção contra danos oxidativos e pela técnica inibição do radical livre DPPH, e posteriormente atividade leishmanicida por infecção em macrófagos. Em seguida os compostos que tiveram como destaques para serem utilizados como tratamento de leishmaniose tegumentar foram levados para análise in silico para avaliar o potencial dos compostos dentro das regras de lipinski e ADMET. Os resultados obtidos apontaram três alcalóides e dois derivados de dibenzoilmetanos como potenciais agentes leishmanicida, cicatrizantes e anti- inflamatório.

Leishmania V braziliensis is the etiologic agent of Cutaneous Leishmaniasis, a disease that affects thousands of people, especially those in social vulnerability. The treatment is based on drugs that have high toxicity and cost, therefore, the search for new chemical compounds is essential for the control and treatment of this disease. For this reason, the search for natural products or synthetic compounds is very important. The present work aims to test the capacity of Indole alkaloids and dibenzoylmethane derivatives, as active principles with healing and leishmanicidal potential. For this purpose, in vitro tests were carried out to evaluate the cytotoxicity in RAW 264.7 macrophage cells and to evaluate the proliferation capacity and toxicity of the compounds. Compounds that obtained a viability rate greater than 80% were selected for the other tests such as anti-inflammatory activity, antioxidant activity to evaluate the ability to protect against oxidative damage and DPPH free radical inhibition capacity, and subsequently leishmanicidal activity by infection in macrophages. The results indicated three alkaloids and two derivatives of dibenzoylmethanes as potential leishmanicidal, healing and anti-inflammatory agents. Keywords: Healing. Cutaneous leishmaniasis.
