No Brasil possui ocorrência em quase todos os estados. Aspectos epidemiológicos podem estar relacionados com a existência de um complexo de espécies do táxon A. cajennense. No presente estudo foi analisada uma porção de 340 pares de base do gene ribossomal mitocondrial 12S de carrapatos da espécie A. cajennense, utilizando a técnica de High Resolution Melt (HRM). .Foram processados 199 espécimes oriundos dos biomas Mata Atlântica, Cerrado e Pantanal. Após a identificação dos haplótipos, estes foram sequenciados a fim de obter suas sequências nucleotídicas, sendo a seguir submetidos à análises de filogenia e filogeografia. As análises filogeográficas indicaram que as populações localizadas nos biomas Mata Atlântica, Cerrado e Pantanal encontram-se em simpatria. A filogenia demonstrou que espécimes pertencentes ao Yungas argentino e da Mata Atlântica brasileira estão filogeneticamente relacionados, assim como os indivíduos do Chaco Serrano e Chaco Ocidental; Florestas montanhosas reais da Cordilheira dos Andes, florestas úmidas da Costa Rica, Guatemala, Equador, Guiana Francesa, México e florestas temperadas dos Estados Unidos da América. A existência de incompatibilidade reprodutiva e a forte estruturação genética nas Américas sugerem que o táxon A. cajennense é um de complexo de espécies. Este achado é extremamente importante uma vez que a prevalência de doenças estão diretamente ligadas aos seus vetores.Item Draft genome sequences of Staphylococcus aureus strains isolated from subclinical bovine mastitis in Brazil(Genome Announcements, 2016-02-18) Silva, Danielle Mendes; Silva, Mônica Pacheco da; Vidigal, Pedro M. Three of them were typed as sequence type 126 (ST126), a genotype with no genome sequence available. ST126 is found in several herds of southern Brazil and is described as a bovine pathogen strongly associated with milk around the world.Item Is Rickettsia amblyommii able to regulate long non-coding RNA expression in Amblyomma sculptum tick? An in silico approach(Universidade Federal de Viçosa, 2017-06-28) Barcelos, Rafael Mazioli; Siqueira, Cláudio Lísias Mafra de; http://lattes.cnpq.br/9152962499215486The Amblyomma sculptum tick belongs to Amblyomma cajennnense complex, the most important taxon in transmission and host for Rickettsia rickettsii bacteria, the main etiologic agent of Brazilian Spotted Fever in Brazil and Rocky Mountain Spotted Fever in USA. As a member of this species complex, A. sculptum tick is actually very important under human and veterinary subject acting as vector for this zoonotic agent in Brazil. Its role in biological cycle of rickettsia strains, as well Rickettsia-tick interactions, still need to be better understood since recent studies found these strains infecting A. sculptum ticks. As putative modulators of these pathogen-host interaction, the roles of long non-coding RNAs (lncRNA) of ticks are unknown and need to be discovered and characterized. The lncRNAs are involving in modulation of a plethora of cell activities as transcription, gene silencing and chromosome opening, for example. Thus, herein we present an in silico approach for analyze the modulation of lncRNAs by R. amblyommii in A. sculptum tick. Using previously published data sets of A. sculptum transcriptomes, we identified putative lncRNAs and evaluated for differential expression in midgut and ovaries in this tick specie. Two assemblers were tested, Trinity and CLC Genomics, to construct the contigs and a pipeline to select the sequences with lncRNA characteristics. We obtained 31,888 and 23,733 contigs of putative lncRNAs using Trinity and CLC Genomics, respectively. We further identified more than 500 sequences of putative lncRNA that significantly aligned to sequences of RefSeq RNA, RNA Central and NONCODE databases. The transcriptome analysis further suggests that R. amblyommii is inducing differential expression of putative lncRNAs in midgut and ovary tissues. Mitochondrial cytochrome c-oxidase subunit 2 (mtDNA cox-2), 28S rRNA and ITS1, 5.8S and ITS2 regions were amplified using the polymerase chain reaction and sequenced to evaluate the phylogenetic relationships of the larva. The genetic profile confirmed that this larva belongs to the species Pseudoterranova decipiens (sensu stricto). This is the first molecular and ultrastructural study of Pseudoterranova decipiens (sensu stricto) in imported cod sold in Brazil. The health implications of these findings are discussed.Item Occurrence of Ehrlichia canis in free-living primates of the genus Callithrix(Revista Brasileira de Parasitologia Veterinária, 2014-11-13) Claudio, Mafra; Barcelos, Rafael Mazioli; Mantovani, Cynthia; Carrizo, Juliana; Soares, Adriano Carlos; Moreira, Higo Nasser Sant'Anna; Maia, Natasha Lagos; Silva, Fernanda de Fátima Rodrigues da; Silva, Vinícius Herold Dornelas e; Boere, Vanner; Silva, Ita de Oliveira eBacteria of the genus Ehrlichia are Gram-negative and coccoid-shaped microorganisms that cause ehrlichiosis – a serious infectious disease that often leads to death. These bacteria present a strong zoonotic potential and primates may act as reservoir hosts. This study involved a molecular analysis to detect these microorganisms in blood samples collected from nineteen primates of the genus Callithrix living free in an Atlantic Forest fragment in the municipality of Viçosa, state of Minas Gerais, Brazil. One of the 19 primates was found to be infected with Ehrlichia canis. This finding points to a new wild host of E. canis with a strong potential for transmission to humans because of its increasing contact with people. This is the first report of Ehrlichia spp. in primate of the genus Callithrix.
