Abstract: Os fungos são capazes de ocasionar sérios danos aos grãos durante o armazenamento, podendo comprometer as características organolépticas e nutritivas, bem como o aproveitamento industrial dos grãos e seus subprodutos. Além disso, podem produzir micotoxinas que são metabólitos secundários potencialmente danosos à saúde humana e animal. Desta forma, é fundamental o controle de fungos nas unidades armazenadoras. Dentre as tecnologias apontadas como promissoras no controle de microrganismos, destaca-se a ozonização. O ozônio é um poderoso agente oxidante que promove estresses oxidativos em células vivas e vem sendo indicado como alternativa no controle de microrganismos. No entanto, pouco se conhece a respeito do potencial desse gás como agente redutor de populações microbianas em milho armazenado. Diante do exposto, objetivou-se com este trabalho determinar a concentração e o tempo de saturação do ozônio em massa de grãos de milho, definir o tempo de ozonização eficaz para a desinfecção microbiológica dos grãos, bem como avaliar o efeito da ozonização nas características físico-químicas do produto. Utilizou-se nos ensaios grãos de milho (Zea mays L.), com teor de água de 14,4% b.u., contendo 94 e 97% de infecção natural por Penicillium spp e Aspergillus spp, respectivamente. Durante a ozonização, amostras de 2,0 kg de grãos foram acondicionadas em recipientes cilíndricos de PVC (19,3 x 23,0 cm). Para determinação da concentração e do tempo de saturação dos grãos de milho pelo ozônio, aplicou-se o gás na concentração de 2,14 mg L-1 e fluxo de 5,8 L min-1. Considerou-se como tempo de saturação o momento a partir do qual os valores de concentração residual do ozônio se mantiveram praticamente constantes. A concentração residual obtida neste ponto foi considerada como a concentração de saturação. Foram estabelecidos cinco períodos de exposição ao ozônio, com a finalidade de avaliar o efeito do gás na desinfecção microbiológica e na qualidade dos grãos de milho. Os períodos de exposição adotados foram 0, 10, 20, 30 e 50 h. No controle, grãos de milho foram submetidos ao tratamento com ar atmosférico, em condições semelhantes ao tratamento com gás ozônio. A quantificação de fungos filamentosos e leveduras em meio de cultura e a ocorrência dos gêneros Aspergillus e Penicillium nos grãos foram avaliadas com a adoção de dois diferentes métodos. O primeiro método foi o da contagem em placas para fungos filamentosos e leveduras e, o segundo, o plaqueamento direto sobre papel de filtro (Blotter test). Ao final de cada período de exposição, os grãos de milho foram avaliados quanto ao teor de água, à condutividade elétrica, ao teor de lipídeos, ao teor de proteínas e ao índice de peróxido. Observou-se que a concentração e o tempo de saturação do gás ozônio nos grãos de milho foi de 0,9874 mg L-1 e 138,56 min, respectivamente. Ocorreu redução em 2,0 ciclos log na contagem de fungos filamentosos e leveduras nas amostras de milho ozonizadas. O ozônio reduziu em 78,5 e 98,0% o índice de ocorrência de Aspergillus spp e Penicillium spp. Com relação às características qualitativas, verificou-se que o ozônio reduziu o teor de água e elevou o índice de peróxido dos grãos de milho. No entanto, a ozonização não alterou a condutividade elétrica, o teor de lipídios e o teor de proteínas. Os resultados demonstrados neste estudo confirmam o potencial efeito fungicida do gás ozônio em grãos de milho, nas condições adotadas.
Fungi are capable of causing serious damage to the grains during storage; they can compromise the organoleptic and nutritional caracteristcs, as well as the industrial use of grains and their by-products. In addition, fungi may produce mycotoxins, which are secondary metabolites potentially harmful to the human and animal health. Thus, it is essential to control the fungi in storage facilities. Among the technologies considered to be promising in the control of microorganisms, ozonation stands out. Ozone is a powerful oxidizing agent that promotes oxidative stress in live cells and has been indicated as an alternative in microorganism control. However, little is known about the potential of this gas as a reducer of microbial populations in stored maize. Given the above, the objectives of this study were to determine the concentration and time of saturation of ozone in the mass of maize grains; to define the effective ozonation time for the microbiological disinfection of the grains; and to evaluate the effect of ozonation on the physicochemical characteristics of the product. In the trials we used maize grain (Zea mays L.) with a moisture content of 14.4% wb., containing 94 and 97% of natural infection by Penicillium spp and Aspergillus spp, respectively. During ozonation, samples of 2.0 kg of grains were conditioned in cylindrical PVC containers (19.3 × 23.0 cm). To determine the concentration and time of saturation of the maize grains by ozone, we applied the gas at a concentration of 2.14 mg L-1 and flow of 5.8 L min-1. The time of saturation was considered to be the moment from which the values of residual concentration of ozone remained practically constant. The residual concentration obtained at this point was considered the concentration of saturation. Five periods of exposure to ozone were established for evaluation of the effect of the gas on the microbiological disinfection and on the quality of maize grains. The periods of exposure adopted were 0, 10, 20, 30 and 50 h. In control, the grain corns were subjected to the treatments with atmospheric air, in similar conditions to the treatment with ozone gas. The quantification of filamentous fungi and yeasts in a culture medium and the occurrence of the genera Aspergillus and Penicillium in the grains were evaluated by adopting two different methods: counting on plates for filamentous fungi and yeasts and direct plating on filter paper (Blotter test). At the end of each period of exposure, the maize grains were assessed as to the moisture content, the electric conductivity, the lipid content, the protein content and the peroxide value. The concentration and the time of saturation of ozone gas in the maize grains were 0.9874 mg L-1 and 138.56 min, respectively. There was reduction of 2.0 log cycles in the count of filamentous fungi and yeast in the ozonated samples. The ozone reduced the rate of occurrence of Aspergillus spp and Penicillium spp by 78.5 and 98.0%. Regarding the qualitative traits, ozone reduced the moisture content and elevated the peroxide value of maize grains. However, ozonation did not affect the electric conductivity, the lipid content or the protein content. The results obtained in this study confirm the potential fungicidal effect of ozone gas on maize grain under the conditions adopted.
