Item Soybean seed galactinol synthase activity as determined by a novel colorimetric assay(Revista Brasileira de Fisiologia Vegetal, 2000) Ribeiro, Marluci; Felix, Carlos R.; Lozzi, Silene de PaulinoGalactinol synthase (GS) is a key enzyme for the biosynthesis of raffinose oligosaccharides (RO) which are the flatulence factors present in soybean seeds and several other legumes. Understanding of soybean seed GS properties is, therefore, of biotechnological interest. The GS enzyme catalyses formation of galactinol and UDP from UDP-gal and myo-inositol. This enzyme is currently assayed by an isotopic method. We have then idealized a more convenient method for GS assay based on the indirect colorimetric determination of the UDP formed which is then hydrolyzed by exogenous apyrase and the resulting Pi quantified by a modification of the colorimetric method of Fiske & SubbaRow. The color developed is stable, and the method is suitable for detection of very low GS activity. The GS activity profiles of developing soybean seeds determined by the isotopic and the colorimetric methods are closely related. The GS enzyme was partially purified (46-fold) by treatment of seed extract with MnCl2, sequential chromatographies on DEAE-Sepharose, Phenyl-Sepharose CL-4B and Q-Sepharose columns. The crude and the partially purified enzyme showed maximum activity at pH 7.0 and 50 ºC. Dithiothreitol and MnCl2 enhanced considerably the activity of the partially purified enzyme. While UDP-glc could be hydrolyzed by the enzyme at a reative activity corresponding to 49% of that calculated for UDP-gal, UDP-man and sucrose were completely ineffective as alternative substrates.Item Variação do tamanho corporal de machos de Eulaema nigrita Lepeletier (Hymenoptera, Apidae, Euglossini). Resposta materna à flutuação de recursos?(Revista Brasileira de Zoologia, 2003-06) Peruquetti, Rui CarlosIt was compared body size (measured as intertegular span) variance of trapped-males of Eulaema nigrita Lepeletier, 1841 sampled in dry (July, August and September) and wet (December, January and February) seasons of the years 1988/89 and 1994/95 in Viçosa (Minas Gerais, Brazil). It was also compared the body size variance between males and females sampled in three nests found in São Carlos (São Paulo, Brazil) and between these males and trapped ones. The smaller male (6.4 mm) was sampled in June and the bigger (8.9 mm) in July 1994, but the majority (32.3%) showed an intertegular span ranging from 7.8 to 8.0mm. The observed variance in body size was similar between males sampled in nests and trapped-males and the variance found inside a nest was similar between the sexes. However, males sampled in 1988/89 were bigger than 1994/95 males and males sampled in nests were smaller than trapped-males. The variance of the body size of males did differ between 1988/89 and 1994/95. In the first period differences between males sampled in dry or wet season was not observed, but in 1994/95 period the males sampled in dry season showed significantly greater variance in body size than males sampled in wet season. The body size variance did not differ between the wet seasons of 1988/89 and 1994/95. The body size variation, measured as the coefficient of variation in intertegular span, did not differ between males of E. nigrita (CV = 4.3%) and ground-nesting bees. These results show that the variation in body size of males of E. nigrita reflects that one found inside the nests of this bee, being similar among males and females. This variation is expected as result of ecological factors influences the nesting females. El Niño climatic events alone or in association with the lack of local food resources due to deforestation and presence of monocultures might play a role in observed body size variation. However this hypothesis is not sufficient to explain the observed body size variation inside a single nest. The absence of pressures of selection related to the females' advantages of produce large offspring perhaps contributes to the maintenance of the observed size variation. Studies regarding maternal allocation in E. nigrita may be useful to explain either the variation in body size or sex ratio found in this Neotropical bee.Item Efeito do alumínio sobre a absorção, o acúmulo e o fracionamento do fósforo em sorgo(Bragantia, 2008-10) Pereira, Jamil Morais; Cambraia, José; Fonseca Júnior, Élcio Meira da; Ribeiro, ClebersonO trabalho teve como objetivo estudar o efeito do Al sobre a absorção, o acúmulo e o fracionamento do P em duas cultivares de sorgo. As plantas foram expostas a níveis tóxicos de Al durante dez dias e, então, colhidas e determinados o crescimento em tamanho e produção de massa seca, os teores de Al e de P total e as diversas formas de P nas duas partes das plantas. Avaliou-se, também, o efeito do Al sobre a absorção de P pelas raízes de plantas intactas. O Al reduziu o crescimento da raiz seminal e a produção de matéria seca de raízes e parte aérea nas duas cultivares, especialmente na sensível. Os teores de Al e de P total aumentaram nas raízes, mas não foram modificados na parte aérea nas duas cultivares. A absorção de P, entretanto, decresceu na presença de Al nas duas cultivares, principalmente na sensível. O Al, de modo geral, modificou as concentrações das várias formas de P solúvel (Pi e Porg) e insolúvel (PRNA e Presidual), exceto a da forma PLIP. Algumas dessas modificações parecem ser importantes e podem estar relacionadas com o mecanismo de tolerância ao Al em sorgo.Item Absorção e acúmulo de cádmio e seus efeitos sobre o crescimento relativo de plantas de aguapé e de salvínia(Revista Brasileira de Fisiologia Vegetal, 2001) Oliveira, Juraci Alves de; Cambraia, José; Cano, Marco Antônio Oliva; Jordão, Cláudio PereiraA absorção e acúmulo de Cd e seus efeitos sobre o crescimento relativo foram estudados em aguapé (Eichhornia crassipes (Mart.) Solms) e salvínia (Salvinia auriculata Aubl.), em solução nutritiva. O aguapé absorveu quantidade significativamente maior de Cd do que a salvínia, tendo a diferença aumentado com o tempo de exposição ao Cd. O Km da absorção de Cd, estatisticamente igual nas duas espécies aquáticas quando as folhas contatavam a solução nutritiva, tornou-se maior em salvínia quando se impediu fisicamente que as folhas contatassem a solução de absorção. O Vmax de absorção de Cd, por outro lado, foi sempre maior em salvínia, independentemente do contato ou não das folhas com a solução nutritiva. A quantidade de Cd adsorvido e absorvido aumentou com a elevação da concentração de Cd na solução de cultivo, nas duas espécies estudadas, tendo sido maior em salvínia, possivelmente em razão da participação da parte aérea no processo de absorção. O acúmulo de Cd nas plantas, elevado nas primeiras 12 h de exposição, decresceu rapidamente com o tempo de exposição, estabilizando após três dias de exposição. Salvínia apresentou maior acúmulo diário de Cd por unidade de peso mas aguapé maior acúmulo total por unidade experimental. Os teores de Cd aumentaram com o aumento do tempo de exposição e da concentração de Cd na solução nutritiva, nas duas partes da planta das duas espécies, especialmente nas raízes. Cerca de 80% de todo o Cd absorvido acumulou-se nas raízes de aguapé, e a distribuição não foi afetada pelo contato das folhas com a solução nutritiva. Em salvínia, quando se impediu o contato das folhas com a solução nutritiva observou-se uma distribuição do Cd aproximadamente igual à de aguapé. Quando, porém, se permitiu o contato das folhas com a solução nutritiva a parte aérea passou a contribuir com quase 50% do Cd absorvido. As taxas de crescimento relativo decresceram intensamente com o aumento da concentração de Cd na solução nutritiva nas duas espécies aquáticas, especialmente em salvínia. As concentrações de Cd para promover redução de 25% no crescimento relativo foram, em média, doze vezes menores em salvínia do que em aguapé, em razão de sua maior sensibilidade a este metal pesado.Item Reproductive biology of Oligosarcus argenteus (Gunther, 1864) adult males and description of the gonadal maturation stages(Brazilian Archives of Biology and Technology, 2009-01) Gomes, Marcos de Lucca Moreira; Andrade, Dálcio Ricardo de; Matta, Sérgio Luis Pinto daOligosarcus argenteus belongs to the Acestrorhynchinae subfamily, being restricted to South America, and found in several Brazilian hydrographic basins, in lotic and lentic environments, where they are able to reproduce. With the purpose of studying the reproductive biology of the males from this species, many morphological parameters were analyzed during a 24 month period, as well as characterizing the different testicular maturation stages. A maturity scale, with three stages (I – Initial Maturing, II – Intermediate Maturing, III – Final Maturing) was proposed for the adult males of Oligosarcus argenteus. The reproductive period was established by the bimonthly frequency of spermatogenesis and by the gonadal maturation stages.Item A comparative study of the ovaries in some Brazilian bees (Hymenoptera; Apoidea)(Papéis Avulsos de Zoologia, 2004) Martins, Gustavo Ferreira; Serrão, José EduardoThe present paper concerns the morphological features of ovaries in 33 species of bees with different social behavior patterns. The ovaries of bees were examined under light microscope. They are polytrophic-meroistic ovaries formed for an anterior germarium and a basal vitellarium. The germarium houses the germ cells and in the vitellarium there are follicles arranged linearly. In general the follicle is constituted by a nutritive chamber (a cluster of nurse cells) and an oocyte chamber, both covered by a single epithelial layer of follicular cells. The number of ovarioles per ovary and the number of mature oocyte per ovary were analyzed. Measurements of ovariole length, oocyte size, oocyte width, follicular epithelial height and the intertegular distance were made to support the comparative study. Statistical analysis showed that representatives of Meliponini and Apini have the largest ovaries. On the other hand, in solitary bees were found the bigger oocytes. Furthermore, our results suggest that there is a tendency for increase in ovary size and ovariole number, with increasing level of sociality.Item Endosperm genotyping as a strategy to differentiate the allele source in maize heterozygous progeny(Pesquisa Agropecuária Brasileira, 2009-10) Martins, Francielle Alline; Carneiro, Pedro Crescêncio Souza; Cruz, Cosme Damião; Carneiro, José Eustáquio de Souza; Guimarães, Claudia TeixeiraThe objective of this work was to distinguish the parental source of alleles in heterozygous progeny using semiquantitative polymerase chain reaction (PCR) in maize endosperm. Endosperms derived from direct and reciprocal single-cross hybrids between maize inbred lines L3 and L1113-01 were genotyped by semiquantitative PCR methodology (SQ-PCR) using fluorescent microsatellite primers. The amplification products were evaluated by the ratios of fluorescence intensity (RFI), calculated between the peaks corresponding to the alleles derived from each parental line. Based on the statistically significant contrast between RFI mean values of direct and reciprocal single-cross hybrids, it was possible to distinguish the number of alleles received from each parental line and, ultimately, to determine the origin of the alleles of each cross. Thus, endosperm genotyping using SQ-PCR is a promising strategy to map QTL in maize outbred populations.Item Genetic analysis of Melipona quadrifasciata Lep. (Hymenoptera: Apidae, Meliponinae) with rapd markers(Brazilian Journal of Biology, 2002-11) Waldschmid, A. M.; Marco-Junior, P.; Barros, E. G.; Campos, L. A. O.Melipona quadrifasciata ("mandaçaia") can be subdivided into two subspecies: M. q. anthidioides and M. q. quadrifasciata. In the present study we used RAPD markers to estimate intercolonial genetic variation among 69 colonies of Melipona quadrifasciata. Ten workers per colony were analyzed. The intercolony genetic distances based on RAPD markers ranged from 29.5% (colonies collected in the State of São Paulo vs colonies from the State of Minas Gerais) to 34.2% (São Paulo vs Santa Catarina). These results indicate a high genetic similarity among the colonies analyzed.According to the genetic distances two different groups could be distinguished. The first containing the samples from Santa Catarina region and the second, samples from Paraná, São Paulo, Minas Gerais, and Espírito Santo. Based on the molecular analysis, bees belonging to the different subspecies M. q. quadrifasciata (from Santa Catarina) and M. q. anthidioides (from the other regions) were distinguished.Item QTL mapping for protein content in soybean cultivated in two tropical environments(Pesquisa Agropecuária Brasileira, 2008-11) Good-God, Pedro Ivo Vieira; Soares, Taís Cristina Bastos; Miranda, Fábio Demolinari de; Soares, Yaska Janaína Bastos; Schuster, Ivan; Piovesan, Newton Deniz; Barros, Everaldo Gonçalves de; Moreira, Maurilio AlvesThe objectives of this study were to detect quantitative trait loci (QTL) for protein content in soybean grown in two distinct tropical environments and to build a genetic map for protein content. One hundred eighteen soybean recombinant inbred lines (RIL), obtained from a cross between cultivars BARC 8 and Garimpo, were used. The RIL were cultivated in two distinct Brazilian tropical environments: Cascavel county, in Paraná, and Viçosa county, in Minas Gerais (24º57'S, 53º27'W and 20º45'S, 42º52'W, respectively). Sixty-six SSR primer pairs and 65 RAPD primers were polymorphic and segregated at a 1:1 proportion. Thirty poorly saturated linkage groups were obtained, with 90 markers and 41 nonlinked markers. For the lines cultivated in Cascavel, three QTL were mapped in C2, E and N linkage groups, which explained 14.37, 10.31 and 7.34% of the phenotypic variation of protein content, respectively. For the lines cultivated in Viçosa, two QTL were mapped in linkage groups G and #1, which explained 9.51 and 7.34% of the phenotypic variation of protein content. Based on the mean of the two environments, two QTL were identified: one in the linkage group E (9.90%) and other in the group L (7.11%). In order for future studies to consistently detect QTL effects of different environments, genotypes with greater stability should be used.Item Ganhos genéticos preditos por diferentes métodos de seleção em progênies de Eucalyptus urophylla(Pesquisa Agropecuária Brasileira, 2009-12) Rosado, Tatiana Barbosa; Resende Júnior, Márcio Fernando Ribeiro; Cruz, Cosme Damião; Bhering, Leonardo Lopes; Rosado, Antônio MarcosO objetivo deste trabalho foi avaliar parâmetros genéticos e comparar os ganhos preditos por meio de diferentes métodos de seleção em famílias de meios-irmãos de Eucalyptus urophylla. Foi utilizada seleção entre e dentro, seleção combinada e seleção com base em modelos mistos (REML/BLUP) para os caracteres diâmetro à altura do peito, altura total e volume total com casca. Foi utilizado o teste de progênie constituído de 100 famílias de meios-irmãos com 55 meses de idade, em espaçamento de 3x2 m, em delineamento de blocos ao acaso, com cinco repetições. As progênies apresentaram variabilidade genética significativa e elevada magnitude de herdabilidade para os caracteres estudados, o que evidencia alto controle genético e condições favoráveis para seleção. Todos os métodos avaliados foram eficientes para aplicação no melhoramento de eucalipto. No entanto, a seleção combinada e a seleção por modelos mistos (BLUP) proporcionam estimativas de ganhos significativamente maiores às obtidas com a seleção entre e dentro, e maior eficiência na escolha dos melhores indivíduos dentro da população.
