Título: Peroxidação de lipídios em membranas e tecidos de dois cultivares de sorgo (Sorghum bicolor (L.) Moench) com tolerância diferencial ao alumínio
Lipids peroxidation in membranes and tissues of two sorghum cultivars (Sorghum bicolor (L.) Moench) with differential tolerance to aluminum

Abstract: Neste trabalho, estudaram-se os efeitos do alumínio (Al) em nível tóxico sobre os sistemas metabólicos que controlam a produção e a eliminação de radicais livres e, consequentemente, a peroxidação dos lipídios em plantas de dois cultivares de sorgo, um sensível (BR007A) e outro tolerante (BR006R) ao Al, cultivadas durante 10 dias em solução nutritiva, pH 4,0, na presença (0,185 mM) e na ausência do Al. Na presença do Al, a atividade da H + -ATPase reduziu no sistema radicular das plantas, mas, principalmente nas membranas plasmáticas provenientes dos ápices das raízes do cultivar tolerante. Dentre as principais modificações verificadas sobre a composição de ácidos graxos das membranas na presença do Al, as mais importantes foram observadas nos ápices das raízes, onde a relação ácidos graxos insaturados/ácidos graxos saturados e, principalmente, o índice de ligações duplas para ácidos graxos com dezoito carbonos foram bastante reduzidos no cultivar sensível. Na presença do Al, a concentração do ácido linolênico reduziu nas membranas provenientes dos ápices das raízes do cultivar sensível, mas, de modo contrário, aumentou muito no tolerante. A atividade enzimática nos tecidos das plantas foi bastante alterada na presença do Al. A atividade das peroxidases aumentou nas raízes e nas folhas dos dois cultivares, principalmente no sensível. De modo contrário, a atividade das peroxidases do ascorbato e das catalases reduziu nesses tecidos, exceto nas raízes do cultivar sensível, onde o aumento da atividade das peroxidases do ascorbato foi observado. A atividade das dismutases do superóxido e das oxidases dos polifenóis aumentou, respectivamente, apenas nas folhas do cultivar tolerante e nas raízes do sensível. A atividade das lipoxigenases aumentou nas folhas em maior intensidade no cultivar tolerante, mas, de modo contrário, reduziu nas raízes em maior intensidade no cultivar sensível. Nas raízes das plantas, a atividade das amônia liases da fenilalanina reduziu nos dois cultivares, mas em maior intensidade no tolerante. A atividade das desidrogenases dos álcoois cinamílicos aumentou nas raízes do cultivar tolerante, mas reduziu no sensível. Na presença do Al, o teor de lignina aumentou nas raízes de ambos os cultivares, porém em maior intensidade no tolerante. O teor de compostos fenólicos solúveis aumentou na presença do Al nas raízes e nas folhas dos dois cultivares, principalmente no sensível. Todos os resultados sugerem que, na presença do Al, o cultivar tolerante produza menores quantidades de radicais de oxigênio livres ou, então, possua mecanismos enzimáticos de remoção, de imobilização e, ou, de neutralização desses radicais mais eficientes que no cultivar sensível, o que, aparentemente, resultou na menor intensidade de peroxidação dos lipídios nos tecidos das raízes e da parte aérea e, especificamente, nas membranas provenientes dos ápices das raízes das plantas do cultivar tolerante.
In this work, the effects of aluminum (Al) at toxic level upon the metabolic systems that control the formation and scavenging of free radicals and, consequently, the lipid peroxidation in plants were examined. The plants were grown for 10 days in nutrient solution, pH 4.0, in the presence (0.185 mM) and absence of Al. The H + -ATPase activity was reduced in the root system of both cultivars, specially in the membranes from the root tips of the tolerant cultivar. Among the main changes observed in the composition of membrane fatty acids in the presence of Al, the more important ones occurred in the root tips. In this case, the ratio of unsaturated fatty acids to saturated fatty acids, and specially the double bond index for 18 carbon fatty acids were quite reduced in the sensitive cultivar. The concentration of linolenic acid was reduced in root tip membranes of the sensitive cultivar in the presence of Al, while a great increase was obtained in the tolerant one. The enzymatic activity in plant tissues was greatly altered by the presence of Al. Peroxidases activity was increased in roots and leaves of both cultivars, but mostly in the sensitive one. On the other hand, ascorbate peroxidases and catalases activities were reduced in these tissues, except in the roots of the sensitive cultivar in which an increase in ascorbate peroxidases was observed. Superoxide dismutases and polyphenol oxidases activities were increased only in the leaves of the tolerant cultivar, and in the roots of the sensitive one. Lipoxygenases activities were increased in the leaves, but were reduced in the roots of both cultivars. The increase and reduction of lipoxygenases activities was greater in the leaves of tolerant and in the roots of the sensitive cultivar, respectively. In the roots, the phenylalanine ammonia-lyase activity was reduced in both cultivars, but mostly in the tolerant one. The activity of cinnamyl alcohol dehydrogenases was increased in the roots of the tolerant cultivar, and was reduced in those of the sensitive one. Lignin content raised in the roots of both cultivars in the presence of Al, but the increase was greater in the tolerant one. Aluminum increased the content of soluble phenolic compounds in the root and leaves of both cultivars, but mostly in the sensitive one. All the results suggest that in the presence of Al, the tolerant cultivar produces less oxygen free radicals or that it has more efficient enzymatic mechanisms of removing, neutralizing and/or scavenging them than those of the sensitive cultivar. This apparently led to a lower lipid peroxidation in the tissues of roots and leaves, and specifically, in the root tip membranes from the tolerant cultivar.
