Obtaining of a soybean line without Kunitz trypsin inhibitor and lectin and its biochemical-nutricional evaluation

Abstract: O presente trabalho está inserido no Programa de Melhoramento da Qualidade e do Sabor da Soja do Bioagro/UFV e tem como objetivo principal a obtenção e caracterização bioquímico-nutricional de uma linhagem de soja com sementes livres de KTI (inibidor de tripsina Kunitz) e lectina. Foram construídos primers específicos para identificar os alelos recessivos dos genes que codificam KTI e lectina. Após a obtenção de uma linhagem contendo elevado teor de proteína e ausência de KTI e lectina, a eficiência de recuperação das características do genitor recorrente com elevado teor protéico foi avaliada. Para isso, utilizou-se a técnica de RAPD para o cálculo da distância genética entre os genótipos doadores do elevado teor de proteína (Monarca HyPro), ausência de KTI (BRM925297), ausência de lectina (Columbia) e a linhagem obtida. A similaridade genética média entre a linhagem final e a isolinha Monarca HyPro foi de 98%. Este resultado revelou um bom índice de recuperação do genoma do recorrente. Os valores de atividade de inibição de tripsina nos genótipos com presença e ausência de KTI foram, respectivamente, 390,33 e 134,40 mg de tripsina inibida/g de proteína, diferindo estatisticamente entre si. A composição química centesimal da isolinha com ausência de KTI e lectina mostrou-se de acordo com a literatura. A avaliação da qualidade protéica das sementes de soja livres de KTI e lectina foi conduzida por meio de ensaios biológicos com ratos Wistar, durante 14 dias. Os índices avaliados foram digestibilidade verdadeira, PER (coeficiente de eficiência protéica), NPR (razão protéica líquida) e NPU (utilização protéica líquida). Os ratos alimentados com a soja livre de KTI e lectina mostraram uma melhor digestibilidade em relação à variedade comercial, com valores próximos aos da caseína, considerada uma proteína padrão. A integridade das microvilosidades do epitélio intestinal de ratos alimentados com dietas à base de soja convencional ou soja livre de KTI e lectina foi avaliada. Os animais alimentados com soja convencional apresentaram um maior nível de alterações na morfologia das microvilosidades intestinais, quando comparados àqueles alimentados com a soja livre de KTI e lectina. Concluiu-se que a retirada genética do KTI e da lectina melhorou significativamente a digestibilidade das proteínas da soja, eliminando a necessidade de se empregar elevadas temperaturas no processamento da soja.
This study is inserted into the “Programa de Melhoramento da Qualidade e do Sabor da Soja”, Bioagro/UFV. It is mainly targeted to the obtainment and nutritional-biochemical characterization of a soybean line, which the seeds are free from KTI (Kunitz trypsin inhibitor) and lectin. Specific primers were obtained in order to identify the recessive alleles of the genes codifying the KTI and lectin. After the obtainment of a line with high protein content and absence of KTI and lectin, an evaluation was performed for the efficiency in recovering the characteristics of the recurrent genitor with high protein content. So, the RAPD technique was used to calculate the genetic distance among the genotypes giving the high protein content (Monarca HyPro), absence of KTI (BRM925297), absence of lectin (Columbia), and the obtained line. The average genetic similarity between the final line and the isoline Monarca HyPro reached 98%. This result revealed a good recovery index of the recurrent’s genome. The values of the trypsin inhibition activity in genotypes with presence and absence of KTI were 390.33 and 134.40 mg of inhibited trypsin/g protein respectively, therefore statistically differing from each others. The centesimal chemical composition of the isoline with absence of both KTI and lectin showed to be according to the available literature. The evaluation of the protein quality in the soy seeds KTI- and lectin-free was accomplished by biological 14-day assays with Wistar rats. The following indices were evaluated: true digestibility, PER (protein efficiency rate), NPR (net protein rate) and NPU (net protein utilization). The rats fed soybean without both KTI and lectin showed a better digestibility for the commercial variety, as the obtained values were near the casein’s that is considered as a standard protein. The integrity of the brushborder microvillous in the intestinal epithelium of the rats fed diets based on either the conventional soybean or KTI- and lectin-free soybean were evaluated. The animals fed conventional soybean showed a higher level of changes in the morphology of their intestinal microvillous, compared with those fed the soybean without both KTI and lectin. According to the results, it is concluded that the genetic removal of both KTI and lectin significantly improved the digestibility of the soybean proteins, therefore eliminating the need for high temperatures in the soybean processing.
