Comparative studies of animal models submitted to dysfunctions lipidics (hyperlipidemia) for the action of natural compositions (flavonoids and coloring)

Abstract: Realizaram-se 4 ensaios biológicos e 1 in vitro com a finalidade de se avaliar as ações farmacológicas de flavonóides e corantes naturais sobre o metabolismo lipídico. Inicialmente, avaliou-se qual espécie animal, entre coelhos da variedade Nova Zelândia, ratos machos da variedade Wistar e Cobaias (Cavia porcellus), seriam melhores para a indução da hiperlipidemia, através da administração de ácido cólico e colesterol juntamente com a ração específica para cada espécie e água ad libitun. O sangue foi coletado aos 30 e 60 dias após o início do tratamento e analisados os teores de colesterol, colesterol-HDL, triacilgliceróis e glicose no soro desses animais. Após o término dos experimentos, foram coletadas amostras de fígado e arco aórtico dos animais, numa repetição de quatro amostras para cada grupo, escolhidos ao acaso, a fim de proceder à análise histopatológica. Neste ensaio, a indução mais eficiente em provocar a hiperlipidemia foi conseguida nos coelhos, mostrando níveis séricos extremamente altos em um curto intervalo de tempo, o que o faz ser um dos animais mais procurado atualmente para trabalhos com esta finalidade. Em posse dos resultados, montou-se o segundo ensaio biológico, optando-se então por utilizar coelhos da raça Nova Zelândia na determinação da melhor via de administração dos flavonóides e corantes naturais, que são utilizados atualmente no estudo de seus efeitos farmacológicos como agentes hipolipidêmicos. De acordo com o que foi observado e pela praticidade na administração, optou-se por administrar as substâncias a serem testadas, pela via oral, nos outros experimentos. Um terceiro ensaio biológico foi realizado no intuito de se determinar a melhor concentração na qual os flavonóides e corantes atuam como fármacos hipolipidêmicos. Os tratamentos foram constituídos por dois grupos controles, sendo um grupo alimentado só com ração e o outro com ração + colesterol + ácido cólico (RCA), e por outros 16 grupos que receberam as seguintes substâncias: quercetina, rutina, morina e naringenina, nas doses e 5 mg, 10 mg, 15 mg e 30 mg. A administração destas substâncias foi pela via oral, sendo ministradas em cápsulas tendo o talco farmacêutico como veículo. As melhores reduções conseguidas na diminuição do nível do colesterol total em relação ao grupo hiperlipidêmico foram observadas nos tratamentos com quercetina e rutina na dose de 5 mg e naringenina na dose de 10 mg, sendo estas variações de 56,42%, 55,00% e 54,04%, respectivamente, sendo significativo pelo teste de Dunnett. O melhor tratamento encontrado na redução dos níveis triglicêmicos foi o com quercetina a 10 mg, sendo que este tratamento abaixou o nível de triacilglicerol destes animais em 77,51% quando comparado com o grupo hipertriglicêmico. Em relação ao colesterol-HDL, foi considerada mais eficiente a substância que promoveu um maior aumento no nível deste constituinte no soro, que foi conseguido pelo tratamento com a morina nas dosagens de 10 e 15 mg, obtendo-se um aumento de 10,79% e 9,00%, respectivamente, em relação ao grupo hiperlipidêmico. Em um último ensaio biológico, comparou-se os efeitos dos flavonóides baicaleína, morina, naringenina, naringina, quercetina, rutina, crisina, luteolina e apigenina, os corantes naturais do extraídos do urucum norbixina e bixina e o medicamento comercial hipolipidêmico colestiramina na dosagem de 10 mg/Kg de peso corporal, por via oral, em cápsula, utilizando o talco como veículo. Sendo esta forma de mais fácil manejo, menos sacrificante para o animal e com uma maior garantia das concentrações ingeridas. A indução da hiperlipidemia foi através do ácido cólico e colesterol, colocados na ração diária dos animais. Esta forma de indução é mais próxima da real e menos sacrificante para o animal. Todas as substâncias reduziram os teores de colesterol no soro sanguíneo dos coelhos, sendo que as melhores reduções foram observadas nas substâncias naringenina, 54,04%, naringina, 49,26%, quercetina, 49,78%, rutina, 48,89%, crisina, 55,56%, luteolina, 52,58% e apigenina, 49,88. Todos os flavonóides estudados e o corante bixina reduziram os níveis de colesterol a valores inferiores ao do medicamento colestiramina e sua média foi estatisticamente diferente a médias deste medicamento, pelo teste de Tukey. Estes resultados mostram que as substâncias estudadas podem ser mais eficientes no combate a hipercolesterolemias dos que os medicamentos atualmente no mercado, com a vantagem de serem menos tóxicos que estes. Os melhores efeitos observados na redução do triacilglicerol plasmático foram conseguidos pelos flavonóides baicaleína, quercetina e crisina. A baicaleína e a rutina diminuíram os níveis de triacilglicerol a valores inferiores ao do grupo controle alimentado apenas com ração, sendo que os valores conseguidos pela baicaleína (-56,91%) foram estatisticamente diferente deste grupo controle pelo teste de Dunnett. A maior parte dos flavonóides aqui estudados teve um melhor efeito sobre o triacilglicerol do que o medicamento colestiramina. As substâncias que promoveram um maior aumento no nível de colesterol-HDL foram os flavonóides baicaleína, apigenina, morina e luteolina e o medicamento colestiramina. Estas substâncias tiveram um efeito hipercolesterolêmico para esta fração lipídica do colesterol, que foram de 122,21%, 281,63%, 10,79%, 21,14% e 37,26%, respectivamente. De uma maneira geral, pode-se observar que as substâncias estudadas apresentaram uma ação farmacológica bastante acentuada no metabolismo lipídico de coelhos hiperlipidêmicos sem, sobretudo, causar efeitos de toxicidade, aparentemente. No ensaio in vitro, procurou-se caracterizar o efeito das substâncias estudadas sobre a atividade da lipase, variando-se a concentração dos modificadores, sendo que todos eles aumentaram esta atividade. Este trabalho demonstrou os efeitos benéficos de alguns flavonóides e corantes naturais sob o metabolismo lipídico de coelhos, sendo que um dos mecanismos para explicar os níveis reduzidos de triacilgliceróis é o aumento da atividade da lipase por essas substâncias, porém, novas pesquisas devem ser realizadas para que estes compostos possam ser utilizados como medicamento e testados no homem.
Four biological assays were realized and 1 in vitro to evaluated the pharmacological action of flavonoids and natural coloring about the lipid metabolism. Initially, was evaluated which animal sort, between rabbits of the race New Zealand, male mouse of the race Wistar and guinea-pig (Cavia porcellus), would be better for the induction of the hyperglycemia, through of the administration of colic acid and cholesterol jointly with the feed specifies for each species and water ad libitun. The blood was collected 30 and 60 days after the beginning of the treatment and the levels of cholesterol, cholesterol – HDL, triacylglicerols and glucose in the serum of those animals were analyzed. After at the end of the experiments, were collected samples of liver and aortic arch of the animals, in a repetition of four samples for each group, choose at random, in order to proceed to the hystopatologic analysis. In this assays the induction more efficient in provoke to hyperlipidemia was obtained us rabbits, showing levels consecutives extremely loud in a short break of time, what is going to be one of the animals more found nowadays for works with this purpose. In possession of the results, was prepared the second biological assays, opting then by utilize rabbits of the race New Zealand in the determination of the best way of administration of the flavonoids and natural coloring, that are utilized nowadays in the study of her pharmacologicals effects how hypolipidemics agents. In agreement with what was observed in the administration, opted by administer the substances it will be tested, by the oral way, in the others experiments. In the third biological assay was realized in the intention of be determined the better concentration in the which the flavonoids and natural coloring like hypolipidemics pharmacs. The treatments were constituted by two groups of controls, being an alone group fed with feed and another with feed + cholesterol + colic acid (RCA), and by others 16 groups that received the following substances: quercetin, rutin, morin and naringenin, in the dose and 5 mg, 10 mg, 15mg and 30 mg. The administration of these substances was by oral way, being administered in capsule having the pharmaceutical talc as vehicle. The best reductions obtained in the diminution of the level of the total cholesterol regarding the group hyperlipidemic were observed us treatment with quercetin and rutin in the dose of 5 mg and naringenin in the dose of 10 mg, being this variation of 56,42%, 55,00% and 54,04%, respectively, being significant by the test of Dunnett. The best reduction found in the decrease of the levels triglycemics was with quercetin to 10 mg, being that this treatment lowered the level of triacylglicerol of these animals in 77,51% when was compared with the group hypertriglycemic. Regarding the cholesterol – HDL, was considered more efficient the substance that promoted the most greatest increase in the level of this constituent in the serum, that was obtained by thetreatment with the morin in the dosages of 10 and 15mg, obtaining itself an increase of 10,69% and 9,00%, respectively, regarding the group hyperlipidemic. In a last biological assays, compared the effects of the flavonoids baicalein, morin, naringenin, naringin, quercetin, rutin, cricin, luteolin and apigenin, the natural coloring extracted of the aurum norbixin and bixin and the commercial medicine hypolipidemic colestiramin in the dosage of 10mg/kg of corporal weight, by oral way, in capsule, utilizing the talc as vehicle. Being this forms of easier management, less sacrificial for the animal and with a bigger guarantee of the concentrations ingested. The induction of the hyperlipidemia was through of the colic acid and cholesterol, put in the daily feed of the animals. This form of induction is more near to the real one in less sacrificant for the animal. All the substances reduced the levels of cholesterol, in the blood serum of the rabbits, being that the best reductions were observed in the substances naringenin, 54,04%, naringin, 49,26%, quercetin, 49,78%, rutin, 48,89%, crisin, 55,56%, luteolin, 52,58% and apigenin, 49,88. All of the flavonoids studied and coloring bixin reduced the levels of cholesterol to lower values to the of the medicine colestiramin and his medium was statistics peculiar on average of this medicine, by test of Tukey. These results show that the substances studied can be more efficient in the combat to hypercholesterolemias than the medicines at present in the market, with the advantage of will be less toxic than these. The best effects observed in the reduction of the triacylglicerol plasmatic were obtained by the flavonoids baicalein, quercetin and crisin. The baicalein and to rutin diminished the levels of tryglicerids to lower values the of the group control feeding just with feed, being that the values obtained by the baicalein (- 56, 91%) were statistics unlike this control group by the test of Dunnett. Most of the flavonoids here studied had a better effect about the triacylglicerol than the medicine colestiramin. The substances that promoted a bigger increase in the level of cholesterol – HDL were the flavonoids baicailen, apigenin, morin and luteolin and the medicine colestiramin. These substances had the effect hypercholesterolemic for this part of the lipid of the cholesterol, that were of 122, 21%, 281,63%, 10,79%, 21,14% and 37,26% respectively. In general way, is able to be observed that the substances studied presented an pharmacological action enough accentuated in the lipid metabolism of rabbits hyperlipidemics without, above all, cause effects of toxicity, apparently. In assays in vitro, found be characterized the effect of the substances studied about the activity of the lipase, varying the concentration of the modifier, being that all they increased this activity. This presentation showed the beneficial effects of some flavonoids and natural coloring in the lipid metabolism of rabbits, being that one of the mechanisms for explain the limited levels of triacylglicerols and increase of the activity of the lipase by those substances, however, new researches should be realized for that these composed can be utilized like medicine and tested in the man.
