Título: Caracterização da via das lipoxigenases de folhas de soja submetidas a ferimento
Characterization of the lipoxygenase pathway of soybean leaves submitted to wounding

Abstract: No presente trabalho foi avaliada a capacidade da planta de soja de responder ao ferimento mecânico através da via das lipoxigenases. Para isso, foi analisado o efeito do ferimento mecânico em folhas de soja sobre a atividade do “pool” de lipoxigenases (LOXs) e os possíveis produtos finais da via. Foram utilizados dois genótipos: um normal (variedade IAC-100) e outro com ausência de LOXs em suas sementes (linhagem IAC-100 TN). Folhas de plantas de soja dos dois genótipos foram submetidas a ferimento mecânico e coletadas 0, 6, 12, 24, 48 e 168 horas após o ferimento. Foram analisadas a resposta local e a resposta sistêmica. Os resultados do estudo bioquímico e cinético das LOXs de folhas de soja dos genótipos revelaram dois picos mais acentuados de atividade de LOXs a pH 4,5 e 6,0, tanto na resposta local quanto na sistêmica. O valor da temperatura ótima das isoenzimas de LOXs a pH 6,0 foi a 25 oC; valores de KM aparente na resposta local e na sistêmica aumentaram consideravelmente seis horas após o ferimento e diminuíram a partir de 12 horas, tendo em 168 horas o menor valor. O menor valor de KMapp em 168 horas sugere uma alteração do “pool” de LOXs em resposta ao ferimento nos dois genótipos analisados. Estes resultados indicam que a remoção genética das LOXs de semente de soja não está interferindo na expressão dos genes que codificam para as LOXs que atuam no mecanismo de defesa da planta de soja contra danos causados por ferimento. Os resultados do estudo dos produtos da via das lipoxigenases revelaram que nos dois genótipos ocorreu aumento de inibição tríptica após ferimento na resposta local e na sistêmica. Os valores em mg de tripsina inibida de proteína foram maiores em 6, 12, 24, 48 e 168 horas. Os resultados deste trabalho reforçam a hipótese de que em folhas de soja ocorre aumento da produção de inibidores de proteases em resposta a ferimento. Em ambos os genótipos estudados ocorreu ligeiro aumento dos níveis de hexanal em 48 e 168 horas e de aldeídos totais em 168 horas após tratamento. Neste estudo, os resultados mostraram que ocorreu aumento da atividade de lipoxigenases e de inibidores de proteases nas plantas de soja submetidas a ferimento. A remoção genética de LOXs da semente de soja no genótipo IAC 100 TN não interferiu na defesa da planta, pela via das lipoxigenases, em resposta ao ferimento mecânico.
In the present work the capacity of the soybean plant was evaluated to response to the mechanical wound through the pathway of the lipoxygenases. For that the effect of the mechanical wound was analyzed in soybean leaves about the activity of the " lipoxygenases pool " (LOXs) and the possible end products of the pathway. Was used: a normal genotype (variety IAC-100) and another with absence of LOXs in its seeds (lineage IAC-100 TN). Leaves of plants of soybean genotypes were submitted to mechanical and collected 0, 6, 12, 24, 48 and 168 hours, after the wound. The local response and response systemic were analyzed. The results of the biochemical and kinetic study of LOXs of leaves of soybean of the two genotypes revealed: two accentuated picks of activity of LOXs the pH 4,5 and 6,0, so much in the local response as in the systemic. The value of the great temperature of the enzymes of LOXs the pH 6,0 went for 25 oC, values of apparent KM in the local response and systemic increased considerably 6 hours after the wound and it decreased starting from 12 hours, tends in 168 hours the smallest value. The smallest value of KMapp in 168 hours suggests an alteration of the " pool " of LOXs in response to the wound in the two analyzed genotypes. These results indicate that the genetic removal of LOXs of soy seed is not interfering in the expression of the enzymes that act in the defense mechanism of the soybean plant to damages caused by wound. The results of the study of the products of the road of the lipoxygenases revealed that in the two genotypes happened increase of inhibition tryptic after wound in the local answer and systemic. The values in mg of inhibited trypsin of protein were larger in 6, 12, 24, 48 and 168 hours. Our results reinforce the hypothesis that in soybean leaves happens increase of the production of proteinases inhibitors in response to wound. In both studied genotypes it happened a slight increase of the levels of hexanal 48 and 168 hours and of total aldehydes in 168 hours after treatment. Our results showed that happened increase of the lipoxygenases activity and of proteinases inhibitors in the soybean plants submitted to wound. The genetic elimination of LOXs of the soybean seed didn't affect in the defense of the plant, for the pathway of the lipoxygenases, in response to the mechanical wound.
