Título: Histomorfometria do epidídimo de morcegos frugívoros (Artibeus lituratus Olfers, 1818) tratados com fungicidas Mancozeb e Tebuconazol
Histomorphometry evaluation of the epididymis of fruit-eating bats (Artibeus lituratus Olfers, 1818) treated with fungicides Mancozeb and Tebuconazole

Abstract: Mancozeb e o Tebuconazol são fungicidas utilizados em lavouras brasileiras, principalmente em fruticulturas e culturas de cereais. Sabe-se que morcegos que se alimentaram de frutos contaminados com estes fungicidas apresentaram alterações metabólicas e hepáticas. Não há relatos sobre seu efeito no epidídimo, importante órgão reprodutivo responsável na maturação e na estocagem de espermatozoides. O objetivo deste trabalho foi avaliar os efeitos do mancozeb e do tebuconazol sobre parâmetros histomorfométricos epididimários. Foram utilizados morcegos da espécie Artibeus lituratus (n=25) divididos em cinco grupos (G=5/grupo). Animais do grupo G1 (controle 1) foram alimentados com mamão sem mancozeb ou espalhante, enquanto que animais do G2 se alimentaram de mamão com espalhante adesivo (1 mL/L; controle 2) e do G3 receberam mamão com calda de espalhante adesivo (1mL/L) e mancozeb (2 g/L) durante 7 dias. Já o grupo 4 recebeu por 7 dias tebuconazol (1 mL/L), com espalhante adesivo, e o grupo 5 tebuconazol (1 mL/L) por 30 dias mais o espalhante. Os epidídimos foram obtidos após eutanásia (CEUA 106/2014) dos animais, sendo os epidídimos fixados inteiros em solução de Karnovsky e incluídos em resina. Imagens microscópicas de 10 cortes histológicos de cada região epididimária, cabeça, corpo e cauda (aumento de 200X), foram analisadas no software Image Pro Plus 4.5, mensurando-se altura de epitélio (μm) e diâmetros luminal (DL) e tubular (DT) (μm). A proporção volumétrica (%) de epitélio, lúmen e interstício foi avaliada através da contagem de 266 pontos sobre 10 cortes histológicos/região em aumento de 200X. Os resultados foram submetidos a ANOVA, sendo as médias comparadas pelo teste de Student-Newman-Keuls a 5%. Os resultados obtidos mostraram que houve um aumento no percentual de epitélio da região da cabeça epididimária nos animais do G3 (61,14 ± 3,05) e G4 (58,85 ± 3,25) em relação aos animais de G1 (52,0 ± 3,37) e G2 (51,2 ± 2,51). O percentual de interstício, na mesma região, foi menor em G3 (28,10 ± 3,86) e G4 (29,61 ± 3,08) que em G1 (36,0 ± 1,21) e G2 (36,77 ± 1,66). Foi observado aumento no DT (139,83 ± 9,42 μm) e no DL em G4 (63,23 ±4,20 μm) em relação aos demais grupos. A região do corpo apresentou um aumento no diâmetro tubular médio em animais do G3 (148,75 ± 9,65 μm), G4 (147,90 ± 13,0 μm) e G5 (145,18 ± 9,29 μm) em relação aos animais do G1 (125,53 ± 10,05 μm), e um aumento no percentual médio de interstício em G2 (40,00 ± 4,57), quando comparado com G1 (31,70 ± 4,56) e G3 (28,09 ± 6,77). Não foram observadas alterações histomorfométricas na região epididimária da cauda nos animais avaliados em relação ao mancozeb (P > 0,05). Houve aumento no DT (206,05 ± 22,77 μm) em G4 na região da cauda em relação aos demais grupos, exceto o G3. O aumento no percentual de epitélio pode ter contribuído para a redução do percentual de interstício em animais tratados com mancozeb e tebuconazol, quando analisada a região da cabeça. Sabe-se que o mancozeb e o tebuconazol alteram o metabolismo de andrógenos, reduzindo sua a produção de andrógenos, que afeta a funcionalidade das células epiteliais epididimárias. Pode concluir que morcegos expostos ao mancozeb por 7 dias e ao tebuconazol por 7 e 30 dias apresentaram alterações histomorfométricas nas regiões epididimárias da cabeça, corpo e cauda.
Mancozeb and Tebuconazole are fungicides used in Brazilian crops, especially in fruit growing and cereal crops. We know that bats feeding on fruits contaminated with these fungicides showed metabolic and liver changes. There are no reports about its effect in the epididymis, important reproductive organ responsible for the maturation and sperm storage. The aim of this study was to evaluate the effects of mancozeb and tebuconazole on epididymal morphometric parameters. From bats were used Artibeus lituratus species (n = 25) divided into five groups (G = 5 / group). Animals in the G1 (control 1) were fed papaya without mancozeb or spreader, while G2 animals fed on papaya with adhesive spreader (1 mL / L; control 2) and G3 received papaya with adhesive spreader syrup (1mL / L) and mancozeb (2 g / l) for 7 days, group 4 received tebuconazole for 7 days (1 mL / L), with spreader-sticker, and 5 tebuconazole group (1 ml / l) for 30 days plus the spreader . The epididymides were obtained after euthanasia (CEUA 106/2014) of the animals on the eighth day of experiment, they set whole being in Karnovsky solution and embedded in resin. Microscopic images of 10 histological cuts of each epididymal region, caput, corpus and cauda (increase of 200X) were analyzed in the software Image Pro Plus 4.5, epithelial height is measuring them (microns) and diameters, and tubular lumen (microns). The volumetric ratio (%) of epithelium, interstitium and lumen was assessed by counting 266 points 10 on histological / region 200X magnification. The results were submitted to ANOVA and the averages were compared by the test Student-Newman-Keuls 5%. The results showed that there was an increase in the epithelium of the percentage of the epididymal caput region in G3 animals (61.14 ± 3.05) and G4 (58.85 ± 3.25) compared to G1 animals (52, 0 ± 3.37) and G2 (51.2 ± 2.51). The percentage of interstitial, in the same region, was lower in G3 (28.10 ± 3.86) and G4 (29.61 ± 3.08) than in G1 (36.0 ± 1.21) and G2 (36, 77 ± 1.66). There was an increase in DT in G4 (139.83 ± 9.42 μm), and increased DL in G4 (63.23 ± 4.20 μm) compared to the other groups. The corpus region experienced an increase in average diameter tubular in G3 animals (148.75 ± 9.65 μm), G4 (147.90 ± 13.0 μm) and G5 (145.18 ± 9.29 μm) compared to animals G1 (125.53 ± 10.05 μm) and an increase in average interstitial percentage of G2 (40.0 ± 4.57) when compared to G1 (31.70 ± 4.56) and G3 (28, 09 ± 6.77). Histomorphometric changes were observed in the epididymal region of the cauda in animals evaluated for mancozeb (P> 0.05). There was an increase in DT (206.05 ± 22.77) in G4 in the cauda region compared to the other groups except the G3. The increase in the percentage of epithelium may have contributed to the reduction of interstitial percentage in animals treated with mancozeb and tebuconazole, when analyzed to the caput. It is known that tebuconazole and mancozeb alter androgen metabolism, reducing the production of androgens, which affect the functionality of epididymal epithelial cells. Can conclude that bats exposed to mancozeb for 7 days and tebuconazole for 7 and 30 days have histomorphometric changes in epididymal the caput, corpus and cauda.
