Título: Efeitos subletais da deltametrina no comportamento larval e nos enócitos pupais de Aedes aegypti
Sublethal effects of deltamethrin in the larval behavior and pupal oenocytes of Aedes aegypti

Abstract: O Aedes aegypti é o principal vetor dos vírus causadores da Dengue. Uma das formas de se controlar a transmissão da doença é o combate desse vetor através do uso de inseticidas. Um dos mecanismos de resistência pode estar ligado ao metabolismo de inseticidas por diferente órgãos ou células, incluindo os enócitos. Os enócitos são células de origem ectodérmica ricas em retículo endoplasmático liso e responsáveis pelos processos de metabolismo de lipídeos e desintoxicação. Nosso trabalho teve como objetivo geral avaliar os efeitos subletais da deltametrina no comportamento larval e nos enócitos pupais e do inseto em duas populações diferentes, susceptível e resistente, de A. aegypti. Para isso foram utilizados larvas de quarto instar (L4) e pupas de populações resistente e susceptível a deltametrina. L4 das duas populações foram submetidas a diferentes concentrações do inseticida: 0,001; 0,003; 0,005; 0,007 ppm e a sobrevivência foi aferida. Após a exposição ao inseticida, L4 foram submetidas a bioensaios comportamentais. Pupas com dois dias de idade após a pupação foram dissecadas e os grupos de enócitos processados para microscopia eletrônica de transmissão (MET) e reação de TUNEL de insetos expostos a 0,003 e 0,005 ppm de inseticida. A 0,007 ppm a deltametrina apresentou efeitos letais sobre as L4 susceptível, enquanto que as resistentes sobreviveram a todas as concentrações testadas. L4 susceptíveis apresentaram um comportamento letárgico e taxa de atividade e natação bem menor do que o controle. Provavelmente, isso ocorreu devido a ação neurotóxica do inseticida. Já nas L4 resistentes observou-se hiperexcitação a 0,005 ppm de deltametrina. Não foram observados efeitos subletais e morte celular significativos tanto em enócitos da população susceptível quanto da resistente expostas à deltametrina. Nossos resultados contribuem para o melhor entendimento das diferentes respostas à exposição subletal à deltametrina em populações susceptíveis e resistentes de A. aegypti e sua relação com o comportamento das formas aquáticas e a estrutura dos enócitos nessa espécie.
The mosquito Aedes aegypti is the main vector of Dengue virus. One strategy to control the transmission of these pathogens by this vector is by means insecticide use. The maj or problems related to this strategy is the emergence of resistance. The mechanisms of resistance may be linked to the metabolism of insecticides by different cells, including oenocytes. The oenocytes are ectodermal cells rich in smooth endoplasmic reticulum and responsible for lipid metabolism and detoxification. This work aimed at evaluating the sublethal effects of deltamethrin on the behavior and structure of oenocytes in immature mosquitoes, of susceptible and resistant A. aegyptistrains. Fourth instar larvae (L4) of both strains were exposed to different concentrations of deltamethrin: 0.001; 0.003; 0.005; 0.007 ppm, and the survival rate was measured. After exposition, L4 of both populations were subjected to behavioral bioassays with the aid of the program View Point Application Manager-Video Track. To study the effects of insecticide in the ultrastructure and cell integrity after insecticide exposure (0.003 and 0.005 ppm), two-days old pupae after pupation were dissected and their oenocytes were processed for transmission electron microscopy (TEM) and TUNEL. The concentration of 0.007 ppm had lethal effects on susceptible L4, while the resistant L4 survived under all the tested concentrations. Susceptible L4 are lethargic and had the swimming activity much lower than control. This probably was due to neurotoxic action of the insecticide. On the other hand, the resistant L4 were hyperexcitated under the treatment With 0.005 ppm. This might result from the inhibitory action of the insecticide on the neurotransmitter GABA. There were no sublethal effects and significant cell death in both susceptible and resistant oenocytes exposed to deltamethrin. Our results revealed the different responses of susceptible and resistant strains of A. aegypti under deltamethrin sublethal exposures, and how these two different strains differs in terms of immature stage behaviors and the structure of oenocytes under exposures.
