Título: Avaliação da ativação do sistema imune de planta pelo receptor NIK (NSP- interacting kinase) e identificação de um novo parceiro de NSP (Nuclear Shuttle Protein) de geminivírus

Abstract: NIK (NSP-Interacting Kinase) é um receptor cinase envolvido na resposta de defesa contra geminivírus, e foi primeiramente identificado como alvo de virulência da proteína de transporte NSP (Nuclear Shuttle Protein) de begomovírus. Previamente, foi identificado que a ativação de NIK medeia a fosforilação indireta da proteína ribossomal L10, promovendo a translocação da proteína citosólica para o núcleo, onde interage com a proteína LIMYB (L10-interacting Myb domain-containing protein), para regular genes envolvidos na maquinaria de tradução da célula, resultando em supressão da tradução global. Foi também demonstrado que a ativação constitutiva de NIK1, através da expressão ectópica do mutante T474D, induz o sistema imune de plantas em Arabidopsis. Nesta investigação, foi analisado em maiores detalhes se a ativação do módulo de sinalização NIK-RPL10-LIMYB também transmitiria um sinal típico de defesa, resultando na indução do sistema imune de plantas. Entretanto, os resultados encontrados sugerem que a via de sinalização antiviral mediada por NIK não transmite o sinal de defesa típico que culmina na ativação de genes relacionados ao sistema imune de plantas. Em primeiro lugar, por meio de estudos de variação global de expressão gênica, induzida por expressão ecotópica do mutante constitutivo T474D ou LIMYB, foi constatado que não houve enriquecimento gênico significativo na lista dos genes regulados positivamente nas categorias de defesa, como resposta de defesa a patógenos, resposta de defesa a vírus e silenciamento gênico induzido por vírus. Além disso, RT- PCR quantitativo de uma amostra representativa dos genes de defesa confirmou que estes genes de defesa não foram regulados positivamente pela expressão de T474D ou LIMYB. Ao contrário, a expressão de LIMYB nas linhagens transgênicas promoveu a regulação negativa dos referidos genes de defesa. Nesta investigação, também se avaliou novos alvos do hospederio que interagem com NSP. Inicialmente, foi isolada, por meio de microarranjos de expressão de 12.000 proteínas de Arabidopsis, uma proteína com características de uma t-SNARE, codificada pelo locus At4g30240 com a capacidade de interagir com NSP in vitro. Esta interação NSP- proteína At4g30240 foi confirmada em ensaios de duplo hibrido em leveduras e por ensaios de complementação de fluorescência bimolecular in planta. Os resultados indicam que At4g30240 interage com NSP in vivo, e pode mediar ou mesmo facilitar o transporte dessa proteína viral entre compartimentos celulares.
NIK (NSP-Interacting Kinase) is a receptor-like kinase involved in defense against geminiviruses, which was first identified as a virulence target of the begomovirus transport protein NSP (Nuclear Shuttle Protein). We have previously identified that NIK activation mediates an indirect phosphorylation of the ribosomal protein L10 causing the translocation of the cytosolic protein to the nucleus, where it interacts with LIMYB (L10-interacting Myb domain-containing protein) to regulate genes involved in the translation machinery resulting in a global translation suppression. Constitutive activation of NIK, through ectopic expression of the mutant T474D in Arabidopsis, has been previously shown to induce the plant immune system. In this investigation, we examined further whether activation of the signaling module NIK-RPL10-LIMYB would transduce a typical defense signal that promotes the induction of the plant immune system. However, the current results suggest that the NIK-mediated antiviral signaling does not induces the basal immune system. Firstly, the global variation of gene expression by ectopic expression of the constitutively activated mutant receptor T474D or LIMYB does not cause a gene enrichment in the up-regulated list of typical defense categories, such as response to pathogens, response to virus and virus-induced gene silencing. Secondly and very importantly, quantitative RT-PCR of a representative sample of defense genes confirmed that they are not up-regulated by expression of T474D or LIMYB. In contrary, expression of LIMYB in the transgenic lines caused a down-regulation of the defense genes examined. We have also examined new host targets that interact with NSP. Initially, we used a 12000 Arabidopsis protein expression-based microarray to isolate a t-SNARE-like protein with the capacity to bind NSP in vitro. This interaction NSP-At4g30240 protein was further confirmed by two- hybrid assay in yeast and by bimolecular fluorescence complementation assay in planta. Our results indicate that At4g30240 interacts with NSP in vivo to mediate or facilitate the viral protein transport among cell compartments.
