Título: Efeitos do alumínio em raízes de soja: alterações morfoanatômicas, fisiológicas e metabólicas
Effects of aluminum on soybean roots: morphological, physiological and metabolic changes

Abstract: Em solos ácidos o alumínio (Al) é a principal limitação da produtividade das culturas em todo o mundo. A acidificação do solo (pH<5) solubiliza as formas tóxicas de Al e concentrações micromolares são capazes de inibir o crescimento das raízes e causar prejuízos fisiológicos e nas funções metabólicas das plantas. Aproximadamente 30% de toda a terra agricultável do mundo é constituída por solos ácidos e com a presença de Al tóxico. O Brasil é o segundo maior produtor de soja do mundo, sendo que a maior região produtora de soja do Brasil é a Centro-Oeste. Seus solos são distróficos, ácidos e com alto teor de Al disponível. O presente estudo teve como objetivos avaliar os danos causados pelo Al em raízes de soja. Para tanto, plântulas de soja foram cultivadas em sala de crescimento com solução de Clark contendo 0 e 100 μM de AlCl 3 , sob pH 4,0, e aeração constante. Os tempos experimentais foram de 24, 48 e 72 horas. Dois cultivares com resistência diferenciada ao Al foram utilizados, P98Y70 e Conquista. Foram avaliados os danos causados no alongamento da raiz, na morfologia e na anatomia radicular. O estado nutricional das plantas e o teor relativo de Al no ápice da raiz. Investigamos a presença de células em processo de morte celular, e a detecção histoquímica de Al. Também abordamos as respostas aintioxidantes da soja ao estresse por Al, bem como moléculas indicadoras de estresses, como ROS e MDA. Os ajustes metabólicos que as plantas puderam fazer frente ao estresse por Al também foram investigados. O crescimento da raiz foi intensamente prejudicado no cultivar P98Y70 após exposição ao Al. As raízes dos dois cultivares apresentaram danos morfológicos após os três tempos de exposição ao Al. O acúmulo de Al nas raízes não diferiu entre os dois cultivares. O cultivar P98Y70 apresentou as menores concentrações de K, além disso, as concentrações de Ca e Mg reduziram em ambos os cultivares. A concentração de P não foi afetada pela presença de Al. P98Y70 apresentou maior porcentagem relativa de Al na coifa e na zona meristemática, enquanto o cultivar Conquista apresentou maiores porcentagens na zona meristemática e de alongamento. Ambos os cultivares apresentaram processos de morte celular no ápice radicular. O cultivar P98Y70 também apresentou coloração mais intensa no teste de detecção de Al por hematoxilina. No resultado positivo do corante Chrome Azurol’S foi possível observar acúmulo de Al nas células da coifa e nas células da epiderme. Entretanto, no cultivar P98Y70 foi possível detectar a presença de Al nas células meristemáticas. O cultivar P98Y70 apresentou maiores concentrações de ROS, maiores níveis de peroxidação lipídica e menor eficiência no uso de enzimas antioxidantes. Diferentemente, o cultivar Conquista apresentou aumentos significativos no uso das enzimas antioxidantes, e consequentemente, menores concentrações de ROS. Além disso, o cultivar Conquista apresentou maiores teores de citrato, responsável pela quelação e anulação dos efeitos do Al na rizosfera e no meio intracelular. Com base em nossos resultados e de acordo com os mecanismos já mencionados na literatura, acerca da defesa das plantas contra o estresse por Al, sugerimos que o cultivar Conquista apresenta mecanismos de tolerância mais eficientes, pois manteve o crescimento da raiz, o equilíbrio entre a produção de ROS e a defesa antioxidante a fim de manter a integridade celular, e principalmente, manteve os níveis de ácidos orgânicos suficientes para atuarem na principal forma de defesa das plantas, já descrita atualmente, contra a toxicidade do Al. Dessa forma, esse cultivar passa a ser mais bem adaptado e recomendado para crescer em solos ácidos e com presença tóxica de Al.
In acid soils aluminium (Al) is the main limitation of crop productivity worldwide. Soil acidification (pH <5) solubilizes toxic forms of Al and micromolar concentrations are able to inhibit root growth and cause physiological damage and metabolic functions of plants. Approximately 30% of all the world's arable land consists of acid soils and the presence of Al toxic. Brazil is the second largest soybean producer in the world, with the largest soybean producing region in Brazil being the Midwest. Their soils are dystrophic, acid and with high Al content available. The present study had as objectives to evaluate the damage caused by Al in soybean roots. For this purpose, soybean seedlings were cultured in a growth room with Clark solution containing 0 and 100 μM AlCl 3, at pH 4.0, and constant aeration. Experimental times were 24, 48 and 72 hours. Two cultivars with differentiated resistance to Al were used, P98Y70 and Conquista. The damage caused by root lengthening, root morphology and anatomy were evaluated. The nutritional status of the plants and the relative content of Al at the root apex. We investigated the presence of cells in the process of cell death, and the histochemical detection of Al. We also addressed the antioxidant responses of soy to Al stress, as well as molecules that indicate stresses, such as ROS and MDA. The metabolic adjustments that plants could make in face of Al stress were also investigated. Root growth was strongly impaired in the P98Y70 cultivar after exposure to Al. The roots of the two cultivars presented morphological damage after the three exposure times to Al. The accumulation of Al in the roots did not differ between the two cultivars. The cultivar P98Y70 presented the lowest concentrations of K, in addition, the concentrations of Ca and Mg reduced in both cultivars. The concentration of P was not affected by the presence of Al. P98Y70 showed a higher relative percentage of Al in the coif and in the meristematic zone, while the cultivar Conquista had higher percentages in the meristematic zone and of elongation. Both cultivars presented cell death processes at the root apex. The cultivar P98Y70 also showed more intense staining in the hematoxylin detection test of Al. In the positive result of the Azurol'S Chrome dye, it was possible to observe accumulation of Al in the cells of the hood and in the cells of the epidermis. However, in the P98Y70 cultivar, it was possible to detect the presence of Al in the meristematic cells. The cultivar P98Y70 had higher concentrations of ROS, higher levels of lipid peroxidation and lower efficiency in the use of antioxidant enzymes. Differently, the cultivar Conquista presented significant increases in the use of antioxidant enzymes, and consequently, lower concentrations of ROS. In addition, the cultivar Conquista showed higher levels of citrate, responsible for the chelation and nullification of the effects of Al in the rhizosphere and in the intracellular environment. Based on our results and according to the mechanisms already mentioned in the literature on the defense of plants in face of Al stress, we suggest that the cultivar Conquista presents more efficient tolerance mechanisms, since it maintained the root growth, the balance between the ROS production and antioxidant defense in order to maintain the cellular integrity and, above all, maintained the levels of organic acids sufficient to act in the main form of defense of the plants, already described nowadays, in face of the toxicity of Al. Thus, this cultivar is better adapted and recommended to grow in acid soils and with a toxic presence of Al.
