Título: Estudo da interação intermolecular entre vermelho 40 e caseína micelar
Study of the intermolecular interaction between allura red and casein micelle

Abstract: A cor de um alimento é uma característica primária de qualidade alimentar responsável por atrair consumidores de qualquer idade ou grupo. A distribuição uniforme dos corantes nos sistemas alimentícios pode ser uma indicação de interações e formação de complexos entre os corantes e um ou mais ligantes existentes na matriz alimentícia. O presente trabalho teve como objetivo estudar a nível molecular os mecanismos envolvidos na interação entre o vermelho 40 (V40) e a caseína micelar em diferentes condições de pH (2.0, 6.6 e 9.0), temperaturas (20, 25, 35, 45 e 55 ̊C) e força iônica (0.01 a 1.0 mol.L -1 ). Técnicas sensíveis e eficazes como fluorescência e microcalorimetria de titulação isotérmicas foram utilizadas para investigar a interação entre o corante e a proteína. Resultados de fluorescência mostraram que a intensidade de fluorescência da caseína micelar diminuiu à medida que as concentrações de V40 aumentaram. Para todas as condições termodinâmicas avaliadas, o mecanismo de extinção da fluorescência foi classificado como estático, havendo, portanto, formação de complexos entre o corante e o agregado proteico. Valores negativos da variação da energia livre de Gibbs padrão de formação de complexo indicaram que houve formação de complexo sendo regida, entalpica ou entropicamente dependendo da condição termodinâmica avaliada. Ainda na técnica de fluorescência, dados da constante de ligação (K b ) e da estequiometria de reação (n) foram obtidos e não apresentam grandes variações quando avaliados sob o efeito do pH. Para todos os pHs testados, os valores de K b foram na ordem de 10 4 e reduziram com o aumento da temperatura. Em relação à estequiometria de formação de complexo (n), os resultados indicaram que houve interação entre o ligante e o agregado de proteína e que, para cada sítio de ligação da molécula de caseína micelar (no sítio do Trp ou próximo ao sítio do Trp), existe uma molécula de V40 ligada. Por outro lado, estudos sobre o efeito do pH mostraram que as cargas influenciaram nas forças de interação entre as moléculas do corante e o agregado de proteína. No pH do leite (pH = 6.6) assim como no pH = 9.0, o processo de formação de complexo foi entropicamente dirigido. Já no pH = 2.0 contatou-se que o processo de formação de complexo foi entalpicamente dirigido. Nos estudos de força iônica, os dados da constante de ligação nas concentrações de 0.5 e 1.0 mol.L -1 de NaCl sugeriram que quanto maior a força iônica, maior a interação do complexo. Medições no microcalorímetro de titulação isotérmica permitiram a determinação da variação da entalpia aparente de interação (ΔH int-apar ) bem como, da variação da entalpia padrão de formação de complexo (ΔH). Considerando a mesma condição avaliada, pH = 6.6 e temperatura de 25 ̊C, o alto valor de ΔH ̊ obtido por medições microcalorimétricas indicou que mudanças conformacionais no agregado proteico assim como, a entrada e saída de fosfato de cálcio de dentro da micela influenciam no ganho de energia e são fatores que devem ser considerados. Este trabalho fornece informações importantes, até então ausentes na literatura, a respeito da interação intermolecular entre o V40 e a caseína micelar, em diferentes condições termodinâmicas.
The color of a food is a primary food quality feature responsible for attracting consumers of any age or group. Uniform distribution of dyes in food systems may be an indication of interactions and complexs formation between this and one or more an other molecules present in the food matrix. The present work aimed to study at molecular level the mechanisms involved in the interaction between Allura Red (AR) and casein micelle in different pH, temperature and ionic strength conditions. Sensitive and effective techniques such as fluorescence and isothermal titration microcalorimetry were used to investigate the interaction between the dye and the protein. Fluorescence results showed that the fluorescence intensity of casein micelle decreased as AR concentrations increased. For all the thermodynamic conditions evaluated, the fluorescence quenching mechanism was classified as static, therefore, complexes were formed between the dye and the protein. Negative values of the standard Gibbs free energy variation of complex formation indicated that there was complex formation being ruled, enthalpic or entropically depending on the thermodynamic condition evaluated. Still in the fluorescence technique, data of the binding constant (K b ) and reaction stoichiometry (n) were obtained and did not present great variations when evaluated under the effect of pH. For all pHs tested, Kb values were on the order of 10 4 and reduced with increasing temperature. In relation to the complex formation stoichiometry (n), the results indicated that there was interaction between the ligand and the protein aggregate and that, for each binding site of the micellar casein molecule (at the Trp site or near the Trp site), There is a bound V40 molecule. On the other hand, studies on the pH effect showed that the loads influenced the interaction forces between the dye molecules and the protein aggregate. At the pH of the milk (pH = 6.6) as well as at pH = 9.0, the complex formation process was entropically driven. Already at pH = 2.0 it was contacted that the process of complex formation was entalpicamente directed. In the ion-strength studies, the binding constant data at concentrations of 0.5 and 1.0 mol.L -1 of NaCl suggested that the higher the ionic strength, the greater the interaction of the complex. Measurements in the isothermal titration microcalorimeter allowed the determination of the variation of the apparent enthalpy of interaction (ΔH int-apar) as well as of the standard enthalpy change of complex formation (ΔH). Considering the same evaluated condition, pH = 6.6 and temperature of 25 ̊C, the high ΔH value obtained by microcalorimetric measurements indicated that conformational changes in the protein aggregate as well as, the entry and exit of calcium phosphate from within the micelle influence the Gain and are factors that should be considered. This work provides important information, hitherto lacking in the literature, on the intermolecular interaction between V40 and micellar casein under different thermodynamic conditions.
