Abstract: Porophyllum ruderale (Asteraceae), erva ruderal e aromática, conhecida popularmente como couve-cravinho. Na medicina popular é utilizada como cicatrizante e antiinflamatória, antifúngica, antibacteriana, calmante, no combate à hipertensão arterial, no tratamento de leishmaniose, no tratamento de edemas e traumatismos, no tratamento de picada de cobra, doenças reumáticas e dores em geral. A atividade cicatrizante tem sido relacionada à presença de teores variáveis de compostos fenólicos do tipo taninos. Os objetivos do trabalho foram descrever a anatomia da raiz, caule e folha de couve-cravinho e preceder a histolocalização dos compostos fenólicos e lipídicos. De cinco plantas cultivadas, em fase de pré-floração, foram coletadas e fixadas em FAA50 ou em sulfato ferroso em formalina neutra tamponada (para observação de compostos fenólicos), a raiz, o caule e as folhas. O laminário foi obtido utilizando-se metodologia tradicional. As raízes são tetrarcas, desenvolvem estrutura secundária precocemente, e possuem ductos secretores no córtex. Nos caules ductos também estavam presentes, entretanto somente nos jovens a reação de compostos fenólicos foi positiva. Nas folhas foram observados ductos delimitados por várias camadas de células epiteliais cujo conteúdo reagiu positivamente indicando a presença de compostos lipídicos e fenólicos. Conclui-se que os ductos são provavelmente as estruturas responsáveis pela secreção de taninos. As infecções micóticas de pele, de unha e de cabelo são comuns em vários países. Os fungos conhecidos como dermatófitos causam infecções crônicas e demandam tratamentos prolongados com drogas antimicóticas caras e às vezes pouco efetivas. O uso de plantas no tratamento de doenças de pele é prática antiga, entretanto são necessários estudos científicos visando a comprovação dos efeitos medicinais. Na terapêutica popular, Porophyllum ruderale é utilizada como antifúngica dentre outros usos. O óleo essencial e os taninos das folhas têm mostrado efeitos inibitórios sobre bactérias e fungos. O objetivo do trabalho foi verificar “in vitro” a atividade antifúngica do extrato das folhas de Porophyllum ruderale sobre dermatófitos (Trichophyton rubrum e T. mentagrophytes) e cândida (Cândida albicans, C. krusei e C. parapsilosis). As folhas utilizadas no preparo do extrato metanólico foram coletadas de plantas cultivadas em fase de pré-floração, secas à temperatura ambiente e trituradas. O extrato foi filtrado, pesado, concentrado, dissolvido em 5 mL de dimetilsulfóxido (DMSO), resultando nas diluições desejadas (0, 1, 2, 3, 4, 6, 8 e 10 mg/mL, e adicionado ao meio de cultura dos microrganismos. As concentrações de 8 e de 10 mg do extrato metanólico/mL do meio foram efetivas na redução do crescimento das colônias de T. rubrum e não reduziram o crescimento das espécies de cândida testadas. A concentração de 3 mg/mL reduziu significativamente as colônias de T. mentagrophytes quando comparada ao controle. Foi concluído que há evidência da couve-cravinho conter propriedades antimicóticas efetivas no tratamento de infecções causadas por T. mentagrophytes e T. rubrum, validando o uso popular da planta entre as pessoas que praticam terapêuticas tradicionais. Experiências do uso da homeopatia em vegetais vêm sendo realizadas por agricultores. São vários os benefícios da homeopatia no cultivo, destacando-se a obtenção de plantas livres de resíduos, o que é muito importante em plantas medicinais. O objetivo do trabalho foi determinar a resposta de plantas a preparados homeopáticos quanto ao teor de tanino nas folhas e nas raízes. O experimento foi instalado em blocos casualizados com três repetições e uma testemunha, sendo submetidos a análise de variância e regressão. Os preparados homeopáticos, tanto em altas quanto em baixas dinamizações, causaram efeito significativo no teor de tanino em folhas e raízes de Porophyllum ruderale, ora aumentando, ora reduzindo. Silicea (CH5) e Sulphur (C200), aplicadas, no intervalo de 10 dias, proporcionaram menor concentração de tanino na folha em relação à testemunha. A aplicação única de Sulphur, Natrum muriaticum, Kalium phosphoricum,e Calcarea carbonica, na dinamização CH4, incrementaram o teor de tanino entre 240 e 288 horas após a aplicação. Silicea e Magnésia carbonica, na mesma dinamização, incrementaram o teor de tanino entre 288 e 336 horas após a aplicação. Kalium phosphoricum e Calcarea carbonica foram os preparados homeopáticos que promoveram maior aumento no teor de tanino das folhas de Porophyllum ruderale quando comparados à testemunha. A aplicação de homeopatia nas plantas de couve-cravinho destinadas a alimentação humana reduz o teor de tanino, diminui a adstringência e aumenta a palatabilidade das folhas. Por outro lado, o uso de preparados homeopáticos que incrementam o teor de compostos fármaco-ativos, inclusive o tanino, interessa ä fitoterapia. A caracterização isoenzimática tem sido usada na abordagem inicial da diversidade de populações geneticamente pouco estudadas como Porophyllum ruderale, quanto a identificação de acessos/genótipos/variedades. O objetivo do trabalho foi caracterizar isoenzimaticamente e fitoquímicamente (quanto ao teor de óleo essencial e de tanino) seis acessos de Porophyllum ruderale. As plantas foram obtidas à partir de sementes colhidas nos municípios de Viçosa (MG), Goianá (MG), Coronel Pacheco (MG), Barra do Garça (MT), Rio Verde (GO) e Jaboticabal (SP). O óleo essencial foi extraído por hidrodestilação em aparelho Clevenger modificado e o tanino foi extraído pelo método recomendado pela AOAC. Os teores de óleo e de tanino não diferiram significativamente entre os acessos. Foram aplicados os seguintes sistemas isoenzimáticos: chiquimato desidrogenase (SKDH), isocitrato desidrogenase (IDH) e malato desidrogenase (MDH), na eletroforese em gel de amido. As amostras de tecidos radiculares e foliares foram coletadas de plantas adultas (aproximadamente 90 dias após a semeadura). Houve variação no número, na espessura e na intensidade de coloração das bandas nos três sistemas avaliados, sendo o sistema SKDH o que melhor separou os acessos. Ocorreram variações entre indivíduos do mesmo acesso, indicando que Porophyllum ruderale, como a maioria das plantas medicinais espontâneas, possui grande variabilidade genética devido ao fato de crescer em ambientes variados e ter reprodução sexuada. As espécies do gênero Porophyllum são amplamente utilizadas na medicina popular da América do Sul e Central, no entanto, poucas têm sido estudadas quimicamente, como é o caso da couve-cravinho. Sendo assim, o objetivo do trabalho foi identificar os constituintes químicos presentes nas folhas de Porophyllum ruderale por meio da triagem fitoquímica. Foi feita a cromatografia em camada delgada (CCD), os extratos foram preparados a partir da amostra vegetal seca e triturada, visando a identificação de alcalóides, polifenóis, flavonóides, taninos, antraquinonas, cardiotônicos, saponina, cumarina, antranóides e terpenos. Verificou-se nas amostras de Porophyllum ruderale analisadas a presença dos seguintes metabólitos secundários: flavonóides (rutina e provavelmente quercetina), taninos (dentre eles o ácido gálico), cumarinas, esteróides e terpenos (dentre eles o terpinol).
Porophyllum ruderale (asteraceae) is a ruderal aromatic herb also known as couve- cravinho. In popular medicine it is used in cicatrizing, anti-inflammatory, anti- fungicide, anti-bacterial and anti-stress treatments, to combat arterial hypertension and additionally in the treatment of leishimaniose, trauma, and snake bite, and for relief of rheumatic and general pain. The cicatrizing effect has been identified as being as a result of the presence of various phenolic composts, such as tannin.The objective of this study was to analyse the anatomy of the root, stem, and leaves of couve-cravinho and the location of the phenolic and lipidic composts. Five cultivated plants were picked during their pre-blossoming phase and fixed in either FAA50 or in a solution of ferrous sulphate in formalin (for observation of the phenolic composts) with respect to the root, stem, and leaves. Laminates were obtained using the usual methodology.The roots are tetrahedral and can be seen to develop an early secondary structure with cortex ducts. These were also present in the stem although only in young samples was the reaction of the phenol compost positive. In the leaves were observed ducts could be identified and were delimitated with respect to the differing layers of epithelial cells. These reacted positively, indicating the presence of lipidic and phenolic composts. As such it can be concluded that the ducts are most likely responsible for the secretion of tannin. Micotic infection of skin, nails and hair is common in many countries. Fungi such as dermatophytes may cause chronic infection and demand lengthy treatment involving expensive antimicotic drugs (and these are often ineffective). The use of plants in the treatment of skin disease is an ancient and established practice. Nevertheless scientific study is vital if medicinal effects are to be confirmed. In popular therapeutic treatment Porophyllum ruderale is utilized as an antifungal treatment. The essential oils and tannins from the leaves have an inhibitory effects on bacteria and fungi. The objective of this experiment was verify “in vitro” the antifungal effects of leaf extracts from Porophyllum ruderale on the above mentioned dermatophytes (Trichophyton mentagrophytes and Trichophyton rubrum) and candida species (Candida albicans, Candida krusei and Candida parapsilosis). The leaves utilized in the preparation of the metanolic extracts were collected from cultivated plants in their pre-blooming phase, dried at room temperatures and powdered. The extracts were filtered, weighed, concentrated, dissolved in 5 mL of dimethyl sulphoxid (DMSO), diluted (0, 1 ,2 , 3, 4, 6, 8, and 10 mg/mL) and added to a culture of the subject micro-organisms. It was found that the 8 and 10 mg/mL concentrations of metanolic extract reduced the T. rubrum colony growth but not the candida growth. The 3 mg/mL concentration reduced significatively the T. mentagrophytes colony growth when compared with control tests. It can be concluded that there is evidence that couve-cravinho plants contain effective antimicotic proprieties in the treatment of infections caused by T. mentagrophytes and T. rubrum, thus validated the popular use of this species in traditional medicine. The use of herbs and plants in homeopathic medicine was first realized in the agricultural community and has shown many medicinal benefits, emphasing that the obtaining of plants free of residue is important in the use of plants within medicine. The objective of the experiment was to determine the response of plants to homeopathic preparations irrespective of the level of tannin in the leaves and roots. The experiment was undertaken in casualized blocks with a repetition of tree subjects and controls, and submitted to different levels of analysis and regression. The homeopathies at both high and low levels of dynamization show significative effects in the yields of tannin from Porophyllum ruderale leaves and roots, with these yields both decreasing and increasing. Silicea (CH5) and sulphur (C200) applied in ten day intervals decreased the yield of tannin in leaves when compared with control tests. The unique application of sulphur, natrum muriaticum, kalium phosphoricum and calcarea carbonica, all of them in CH4 dynamization, increased tannin yields during the period between 240 and 288 hours after application while silicea and magnesia carbonica, both in CH4 dynamization, increase tannin yields during the period between 288 and 336 hours after application. Kalium phosphoricum and calcarea carbonica was the homeopathy prepared that created the greatest increase in tannin yields from Porophyllum ruderale leaves when compared with control tests. The application of homeopathy to couve- cravinho plants intented for human alimentation can decrease tannin yields, the astringence of the plants and increase the palatability of leaves. On the other hand, the use of homeopathic preparations with increased pharmaco-active compounds such as tannin is of great interest to phytotherapy. The isoenzimatic characteristics of the plant have been used in initial approaches towards identifying the diversity of lesser studied genetic populations and groupings of Porophyllum ruderale in order to identify different genotypes and varieties. The objective of this study was to identify the isoenzimatic and phytochemical (essential oil and tannin) characteristics of six varieties of Porophyllum ruderale. The plants were obtained from seeds collected in Viçosa (MG), Goiana (MG), Coronel Pacheco (MG), Barra do Garça (MT), Rio Verde (GO) and Jaboticabal (SP). The essential oil was extracted by the modified instrument Clevenger and tannin was extracted using the AOAC recommended methods. The amount of oil and tannin do not differ significantly between varieties. The following izoenzimatic systems were applied: desidrogenase shiquimate (SKDH), desidrogenase isocitrate (IDH) and desidrogenase malate (MDH) in the gel electrophoresis of starch. The vegetal tissue (root and leaf) sample was collected from adult plants (approximately 90 days after seeding). There was some variation in the number, thickness and intensity of color bands in tree tested systems and the SKDH system was the one that was found to best separate differing varieties. There was some variation between individuals of the same type indicating that Porophyllum ruderale, as the most spontaneous of the medicinal plants, has significant genetic variability due to growing in different kind of environments and methods of sexual reproduction. The species of the genus porophyllum are extensively utilized in popular medicine throughout South America and Central America however few (such as couve-cravinho) of these species have been chemically studied. Therefore the objective of this study was to identify the chemical compounds present in Porophyllum ruderal leaves through phytochemical screening. This was done by way of thin layer chromatography (CCD), the extracts were prepared from vegetal samples, were dried and powdered, in order to identify alkaloids, phenols, flavonoids, tannins, quinones, cardiac glycosides, saponins, coumarins, antranoid and terpene. It was verified that the analysed samples of Porophyllum ruderale have some secondary metabolites: flavonoids (rutine and quercetine), tannins (among them gallic acid), coumarins, steroids and terpenes (among them terpinol).
