Título: Propriedades bioquímicas e cinético-enzimáticas de serino e cisteíno proteases produzidas por bactérias isoladas do trato intestinal de Anticarsia gemmatalis

Abstract: Inibidores de proteases atuam no mecanismo de defesa de plantas contra infestação de insetos e patógenos. A utilização de genes que codificam inibidores de enzimas digestivas para a obtenção de plantas resistentes ao ataque de insetos é uma estratégia promissora. Porém, para que isso ocorra é importante que a planta expresse uma combinação de inibidores que cubra o espectro total de proteases intestinais. Dessa forma, deve ser considerada a fisiologia do inseto, a bioquímica de sua digestão e a microbiota local. Recentemente foram isoladas diversas bactérias proteolíticas do intestino da lagarta-da-soja, Anticarsia gemmatalis. Todas foram capazes de produzir quantidades significativas de proteases quando cultivadas em meio de cultura adequado. Neste contexto o presente trabalho fundamentou-se em caracterizar as serino e cisteíno proteases provenientes de Bacillus cereus, Staphylococcus xylosus, Enterococcus mundtii e Enterococcus gallinarum, isoladas do trato intestinal de A. gemmatalis. As culturas bacterianas foram inoculadas em meio de cultura infusão cerébro coração(BHI) acrescidos de 0,1% de soro albumina bovina (BSA) mantidas a 37°C, a 200 rpm em agitador. Os extratos enzimáticos foram obtidos através de centrifugação das culturas a 10.000 rpm por 20 minutos a 4°C. Todas as serino e cisteíno proteases foram capazes de hidrolisar a caseína, e os substratos sintéticos L- BApNA e L-TAME. Verificou-se pronunciada atividade das serino proteases em pH 8,5 para B. cereus, S. xylosus, E. gallinarum, e em pH 7,5 para E. mundtii e temperaturas de maior atividade a 250C para B. cereus, a 300C para S.xylosus e E. mundtii e 350C para E.gallinarum sobre os substratos L- BApNA e L-TAME. Para as cisteíno proteases de todas as bactérias verificou-se maior atividade em pH 7,5 frente os substratos L-BApNA e L-TAME. O efeito da temperatura para as cisteíno proteases bacterianas sobre os substratos LBApNA e L-TAME mostraram maior atividade a 35ºC para B. cereus e S. xylosus, 25ºC para E. mundtii e 300C para E. gallinarum. Os valores de KMapp para serino proteases utilizando L-BApNA foram de 0,15 mM, 0,12 mM, 0,14 mM, e 0,26 mM respectivamente, para B. cereus, S. xylosus, E. mundtii e E. gallinarum. Os valores de KMapp determinados nos ensaios com o substrato L-TAME foram de 12,4 µM, 48,4 µM, 18,2 µM e 45,2 µM, respectivamente, para B. cereus, S. xylosus, E. mundtii e E. gallinarum. Para cisteíno proteases utilizando L- BApNA os valores de KMapp obtidos foram de 0,67 mM, 0,63 mM, 0,67 mM, e 0,270 mM, respectivamente para B. cereus, S. xylosus, E. mundtii e E. gallinarum, e com substrato L- TAME foram de 0,11 mM, 0,13 mM, 0,12 mM e 0,16 mM respectivamente, para B. cereus, S. xylosus, E. mundtii e E. gallinarum. Na análise do efeito de íons cálcio na atividade proteolítica frente os LBApNA e L-TAME, observou-se que as atividades das serino e cisteíno proteases presentes nos extratos bacterianos aumentaram em presença de CaCl2. O efeito de vários inibidores de proteases também foi avaliado, usando L-BApNA e L-TAME como substratos. Com relação ao efeito da concentração de EDTA, inibidor de metalo proteases e quelante de íons Ca+2, verificou-se uma diminuição na atividade amidásica e esterásica com o aumento da concentração de EDTA na mistura de reação para as serino e cisteíno proteases bacterianas. Os inibidores de serino proteases TLCK (irreversível) e Aprotinina (competitivo) diminuíram significativamente a atividade das serino proteases bacterianas. O inibidor de cisteíno proteases E-64 não influenciou as serino proteases bacterinas, o contrário foi observado para as cisteíno proteases produzidas pelas bactérias, onde ocorreu queda na atividade amidásica e esterásica frente o inibidor E-64. Um aumento de TLCK no meio fez com que a atividade das cisteíno proteases bacterianas diminuísse gradativamente, fato atribuído à reação desse inibidor com resíduo de aminoácido histidina na tríade catalítica de cisteíno proteases. A Aprotinina, não influenciou as cisteíno proteases bacterianas. Pespstatina A, inibidor de aspartil proteases não influenciou as serino nem as cisteíno proteases bacterianas. Assim, os resultados de caracterização cinética e de efeito de inibidores de proteases sobre a atividade das proteases produzidas por B. cereus, S. xylosus, E. mundtii e E. gallinarum concluem que essas bactérias sintetizam e excretam no lúmen intestinal de A. gemmatalis, enzimas da família das serino e cisteíno proteases.
Protease inhibitors are involved in the plant defense mechanism against infestation by insects and pathogens. Use of genes encoding inhibitors of digestive enzymes offer a promising strategy to produce plants resistant to insect attack. In order for this to occur, however, the plant will need to express a combination of inhibitors that cover the full spectrum of intestinal proteases. Insect physiology, biochemistry of digestion and local microbiota should therefore be considered. A number of intestinal proteolytic bacteria have been recently isolated from the velvetbean caterpillar (Anticarsia gemmatalis). All of them were able to produce significant quantities of proteases when grown in appropriate culture medium. The present study aimed at characterizing serine and cysteine proteases from Bacillus cereus, Staphylococcus xylosus, Enterococcus mundtii and Enterococcus gallinarum isolated from the intestinal tract of A. gemmatalis. Bacteria were inoculated in brain heart infusion broth (BHI), added of 0.1% bovine serum albumin (BSA) and incubated at 37°C in a rotary shaker (200 rpm). Enzymatic extracts were obtained by centrifuging bacterial cultures at 10000 rpm for 20 minutes at 4°C. All serine and cysteine proteases were able to hydrolyze casein, and synthetic substrates L-BApNA and L-TAME. There was high serine protease activity at pH 8.5 for B. cereus, S. xylosus, E. gallinarum, and at pH 7.5 for E. mundtii and high activity at the temperatures 25°C for B. cereus, 30°C for S.xylosus and E. mundtii and 35°C for E.gallinarum on the substrates L-BApNA and L-TAME. Cysteine proteases of all bacteria increased activity at pH 7.5 in the substrates LBApNA and L-TAME. The activity of bacterial cysteine proteases on the substrates LBApNA and L-TAME increased at 35ºC for B. cereus and S. xylosus, 25ºC for E. mundtii and 30ºC for E. gallinarum. KMapp values for serine proteases using L-BApNA were 0.15 mM, 0.12 mM, 0.14 mM, and 0.26 mM for B. cereus, S. xylosus, E. mundtii and E. gallinarum respectively. KMapp values determined in tests of serine proteases with L-TAME substrate were 12.4 μM, 48.4 μM, 18.2 μM and 45.2 μM for B. cereus, S. xylosus, E. mundtii and E. gallinarum respectively. Cysteine proteases with L-BApNA produced KMapp of 0.67 mM, 0.63 mM, 0.67 mM, and 0.270 mM for B. cereus, S. xylosus, E. mundtii and E. gallinarum respectively, and with L-TAME were 0.11 mM, 0.13 mM, 0.12 and 0.16 mM mM for B. cereus, S. xylosus, E. mundtii and E. gallinarum respectively. The analysis of the effect of calcium ions on the proteolytic activity using L-BApNA and L-TAME showed that the activity of serine and cysteine proteases in bacterial extracts increased in presence of CaCl2. The effect of several protease inhibitors was also evaluated using L-BApNA and L-TAME as substrates. EDTA, as inhibitor of metal mediated proteases and chelator of Ca+2 ions, caused decrease in amidasic and esterasic activities with increase of EDTA concentration in the reaction mixture of serine and bacterial cysteine proteases. Serine protease inhibitors TLCK (irreversible) and aprotinine (competitive) significantly decreased bacterial serine protease activity. The cysteine protease inhibitor E-64 did not affect bacterial serine proteases, but decreased the amidasic and esterasic activity of bacterial cysteine proteases. Increase in TLCK concentration in the medium has gradually decreased the bacterial cysteine protease activity, which was attributed to the reaction of the inhibitor with the histidine residue in the catalytic triad of cysteine proteases. Aprotinine did not affect bacterial cysteine proteases. Pespstatine A, an aspartyl protease inhibitor, did not affect serine nor cysteine proteases. Thus, the kinetic characterization and effects of protease inhibitors on the activity of proteases produced by B. cereus, S. xylosus, E. mundtii and E. gallinarum led to the conclusion that these bacteria synthesize and release enzymes of the family serine and cysteine proteases into the intestinal lumen of A. gemmatalis.
