Abstract: O primeiro experimento teve como objetivo avaliar o efeito do fornecimento de concentrações crescentes de uréia nas dietas de cabras da raça Alpina sobre a dinâmica folicular ovariana e a concentração de uréia, glicose e progesterona no plasma. Foram utilizadas 29 cabras, distribuídas aleatoriamente em quatro tratamentos, que receberam 0,0% (T1 Controle, n=7); 0,73% (T2, n=7); 1,46% (T3, n=7); e 2,24% (T4, n=8) de uréia na matéria seca (MS) da dieta. A dinâmica folicular ovariana foi monitorada diariamente durante um, dois ou três períodos interovulatórios consecutivos. As concentrações de uréia e de glicose no plasma foram analisadas semanalmente. A concentração de progesterona plasmática foi analisada no dia do estro (dia 0) e nos dias três, sete, 11, 15 e 19 após o estro nas cabras que manifestaram ciclos estrais de duração normal (17 a 25 dias). Análises adicionais de progesterona plasmática no 25º dia após o estro e a intervalos semanais após o 25º dia foram realizadas nas cabras que manifestaram ciclos estrais de duração longa (>25 dias). Nas que apresentaram ciclos estrais de duração curta (<17 dias), a concentração de progesterona plasmática foi analisada no dia do estro e nos dias três e sete após o estro. Foram observados 28 (59,57%) ciclos estrais de duração normal, sete (14,89%) de duração curta e 12 (25,53%) de duração longa. Os ciclos estrais de duração curta tiveram 6,14(0,69 dias e uma única onda folicular. Os de duração longa tiveram entre 26 e 79 dias, com duas a nove ondas foliculares. Nos ciclos estrais de duração normal ocorreram duas (n=6), três (n=13), quatro (n=8) ou cinco ondas foliculares (n=1). Nas cabras com duas ondas foliculares, o dia de emergência (P<0,05) da primeira onda folicular, o comprimento (P<0,06) da primeira onda folicular e o dia de emergência do maior folículo da primeira onda folicular (P<0,05) apresentaram comportamento quadrático em função da concentração de uréia nas dietas. Nas cabras com três ondas foliculares, observou-se que o comprimento da terceira onda (P<0,01), o diâmetro máximo do maior folículo da terceira onda (P<0,01) e o número de ovulações (P<0,06) apresentaram comportamento quadrático em função da concentração de uréia nas dietas. Em relação às cabras com quatro ondas foliculares, observou-se que o dia de emergência da terceira onda (P<0,05) e o dia de emergência do maior folículo da terceira onda (P<0,06) reduziram inearmente em função da concentração de uréia nas dietas, enquanto o diâmetro do segundo maior folículo da primeira (P<0,05) e da quarta (P<0,01) ondas foliculares aumentou linearmente em função da concentração de uréia nas dietas. Os períodos interestro e interovulatório, a duração das fases lútea e folicular e as demais características da dinâmica folicular ovariana não foram influenciados pela concentração de uréia nas dietas. As concentrações de uréia e de glicose no plasma não diferiram entre os animais dos tratamentos e nem entre as semanas de coleta. Observou-se comportamento quadrático da concentração de progesterona plasmática no dia do estro e no dia 11 após, em função da concentração de uréia nas dietas. No dia 15 após o estro, a concentração deste hormônio reduziu linearmente à medida que a concentração de uréia nas dietas aumentou. No segundo experimento, avaliou-se o efeito das mesmas concentrações de uréia testadas no primeiro experimento sobre o número, a qualidade e o estádio de desenvolvimento embrionário e sobre a concentração de uréia e de glicose no plasma, no dia do estro e no da coleta dos embriões. Vinte e duas cabras da raça Alpina foram superovuladas e os embriões foram coletados pela técnica transcervical entre o sétimo e o oitavo dia após a primeira cobertura. Dezoito (81,82%) cabras manifestaram estro após a sincronização. A duração do estro e o intervalo remoção da esponja-início do estro não foram influenciados (P>0,05) pelos tratamentos. Quatorze cabras (77,77%) responderam à superovulação. Os números de estruturas e de embriões não diferiram (P>0,05) entre os tratamentos, sendo representados pelas equações ipisilon = 9,64 e ipisilon = 8,50, respectivamente. O número (P<0,10) e a percentagem (P<0,07) de embriões viáveis e o número (P<0,08) e a percentagem (P<0,05) de embriões excelentes e bons apresentaram comportamento quadrático em função da concentração de uréia nas dietas. Com exceção de um embrião de quatro células, os demais foram coletados em estádio de desenvolvimento compatível com o esperado. A concentração de uréia no plasma, antes e quatro horas após a alimentação, nos dias do estro e da coleta dos embriões, não foi influenciada (P>0,05) pelos tratamentos. No dia do estro, a concentração de glicose no plasma, antes da alimentação, aumentou linearmente com o aumento da concentração de uréia nas dietas (P<0,05). A concentração de glicose no plasma após a alimentação, no dia do estro e antes e após a alimentação no dia da coleta dos embriões não diferiu (P>0,05) entre os animais dos tratamentos. No terceiro experimento, dois blastocistos expandidos de grau I do tratamento T1, quatro blastocistos expandidos de grau I de cada um dos tratamentos T2 e T4 e três blastocistos expandidos de graus I (n=1) e II (n=2) do tratamento T3 foram submetidos à análise ultra-estrutural. Nos embriões do tratamento T1 foram observados: células trofoblásticas (CT) achatadas, pouco eletrondensas, polarizadas e com abundantes microvilosidades na sua superfície apical; citoplasma com inclusões lipídicas associadas às mitocôndrias maduras ou imaturas; complexos juncionais bem desenvolvidos entre as CTs; espaço perivitelínico (EPV) estreito e preenchido pelas microvilosidades das CTs; e baixa ocorrência de material extrudado em degeneração no EPV. Nos embriões do tratamento T1 as células da massa celular interna (MCI) não foram identificadas. A ultra-estrutura das CTs dos embriões coletados das cabras do tratamento T2 foi similar àquela observada nos embriões do tratamento T1, exceto pela presença de estruturas muito eletrondensas em contato com a zona pelúcida (ZP); poucos filamentos de actina e intermediários e maior ocorrência de fagossomos no citoplasma. As células da MCI, nos embriões do tratamento T2, apresentaram-se mais eletrondensas quando comparadas às CTs e, no seu citoplasma, foram verificadas organelas similares àquelas observadas nas CTs, vacúolos contendo material eletrondenso e corpos residuais. No interior de algumas células da MCI foram observados grandes vacúolos contendo células em processo de degeneração. O contato entre as células da MCI e as da MCI com as CTs ocorreu em pontos específicos, por aposição de suas membranas. Um dos embriões do tratamento T3 apresentou ultra-estrutura similar àquelas dos embriões dos tratamentos T1 e T2, exceto pela disposição das organelas e dos filamentos de actina e intermediários no citoplasma das CTs e pela maior ocorrência de vacúolos autofágicos e de figuras mielínicas. Os outros dois embriões do tratamento T3 apresentaram intensa modificação morfológica da célula: as CTs apresentaram aspecto globular com poucas microvilosidades, ou apresentaram-se como porções citoplasmáticas muito delgadas, desprovidas de microvilosidades; no citoplasma foram observados muitos vacúolos, fagossomos, inclusões lipídicas e poucas organelas e elementos do citoesqueleto; o EPV mostrou-se dilatado, com fragmentos celulares e blastômeros extrudados e, na blastocele, foram observados fragmentos celulares. Em um dos embriões do tratamento T3 os espaços entre as células apresentaram-se aumentados. As células em degeneração foram mais comuns no epitélio trofoblástico nos embriões do tratamento T3. Todos os embriões coletados das cabras do tratamento T4 foram perdidos durante o processamento para a microscopia. Os resultados sugerem diferenças ultraestruturais entre os embriões coletados das cabras do tratamento-controle e os das suplementadas com 0,73% e 1,46% de uréia na dieta, principalmente com relação ao aumento de estruturas morfológicas relacionadas com processos de reorganização e/ou degeneração celular.
In the first experiment the effect of different concentrations of urea in the diets of Alpine goats on the ovarian follicular dynamics and the plasma urea, glucose and progesterone profile were evaluated. Twenty nine (29) goats were randomly assigned into four treatments that received: 0.0% (T1 - Control, n=7); 0.73% (T2, n=7); 1.46% (T3, n=7) and 2.24% (T4, n=8) of urea in the dry matter (DM) of the diet. The ovarian follicular dynamics was monitored daily during one, two or three consecutive interovulatory periods. The plasma urea and glucose concentration were analyzed weekly. The plasma progesterone concentration was analyzed at estrus (day 0) and on days 3, 7, 11, 15 and 19 after estrus in the goats that showed estrous cycles of normal length (17 to 25 days). Additional blood samples were taken at the 25th day after the estrus and at weekly intervals after the 25th day in the goats which showed long length estrous cycles (>25 days). In the goats with short length estrous cycles (<17 days), the collections were done on the estrus day and in days 3 and 7 after. There were 28 goats (59.57%) with estrous cycles of normal length, seven (14.89%) with estrous cycles of short length and 12 (25.53%) with estrous cycles of long length. The short length estrous cycles had 6.14±0.69 days and one follicular wave. The long length estrous cycles had between 26 to 79 days, with 2 to 9 follicular waves. From the normal length estrous cycles, six, thirteen, eight and one were characterized by the occurrence of two, three, four and five follicular waves, respectively. In the goats with two follicular waves, the emergence day of the first follicular wave (P<0.05), the length of the first follicular wave (P<0.06), and the emergence day of the largest follicle of the first follicular wave (P<0.05) showed quadratic behavior related to the urea concentration in the diet. In the goats with three waves, the quadratic behavior of the third wave length (P<0.01), of the maximum diameter of largest follicle of third wave (P<0.01) and the ovulation number (P<0.06) were related to the urea concentration in the diets. In the goats with four waves, it was observed that the emergency day of the third follicular wave (P<0.05) and the emergency day of the largest follicle of the third wave (P<0.06) decreased linearly with the urea concentration in the diets, as long as the diameter of the second largest follicle of the first (P<0.05) and of the fourth waves increased (P<0.01) linearly with the urea concentration in the diets. The interestrus and interovulatory periods, the luteal and follicular phases lenght and the other ovarian follicular dynamic characteristics were not influenced by the urea concentration in the diets. The plasma urea and glucose concentration did not differ among the animals from the treatments and among weekly blood collections. It was observed quadratic behavior of progesterone plasma concentration in the estrus day and on the 11th day after estrus, related to the urea concentration in the diets. In the 15th day after estrus, the progesterone plasma concentration reduced linearly when the urea concentration in the diets increased. In the second experiment, the effect of the same urea levels in the diet, tested in the first experiment were evaluated on the number, quality and development state of the embryo; plasma urea and glucose concentration at estrus day and on the embryos collection day. Twenty-two Alpine goats were superovulated and the embryos were collected by the transcervical technique between the 7th and the 8th day after the first mount. Eighteen (81.82%) goats showed estrus after the synchronization. The estrus length and the interval from the removal of the sponge to the beginning of estrus were not influenced (P>0.05) by the treatments. Fourteen goats (77.77%) were responsive to the superovulation protocol. The structure and embryo numbers did not differ (P>0.05) among the animals from the treatments, and were represented by the equations ipisilon = 9,64 and ipisilon = 8.50, respectively. The number (P<0.10) and the percentage (P<0.07) of viable embryos and the number (P<0.08) and the percentage (P<0.05) of excellent and good embryos showed quadratic behavior related to the urea concentration in the diet. All embryos were collected at the development stage compatible with the expected one, but an embryo of four cells. The plasma urea concentration before and four hours after feeding at the estrus and at the embryo recovery days were not influenced (P>0.05) by the treatments. At estrus day, the plasma glucose level before feeding increased linearly with the increase of the urea concentration in the diets (P <0,05). The plasma glucose concentration after feeding, at estrus day and, before and after feeding at the embryos recovery days did not differ (P>0.05) among the animals from the treatments. In the third experiment, two grade I expanded blastocysts of the treatment T1, four grade I expanded blastocysts from each T2 and T4 treatments and three expanded blastocysts of grades I (n=1) and II (n=2) of the treatment T3 were submitted to ultra-structural analysis. The T1 embryos showed: flattened trophoblast cells (TC), minor electron-dense, polarized and with abundant microvilli in their apical surface; cytoplasm with lipid droplets associated to the mature or immature mitochondria; junctional complex well developed among TCs; perivitelline space (PVS) narrow and filled out with the microvilli of TCs; small occurrence of extruded material in the EPV. The inner cell mass (ICM) from these embryos were not identified. The ultrastructure of TCs of the collected embryos from treatment T2 goats was similar to that observed in the embryos of the treatment (T1), except by the presence of structures more electron-dense in contact with the zona pellucida (ZP) and in the cell cytoplasm; few actin and intermediate filaments and more phagosomes in the cytoplasm. The cells of IMC in these embryos were more electron-dense when compared to TCs and their cytoplasm showed similar organelles to those observed in TCs, vacuoles containing electron-dense material, residual bodies and degenerating cells. The contact between IMC cells and between IMC and TCs cells ocurred in specific points by of their membranes apposition. One treatment T3 embryo showed similar morphology to those observed in the embryos from treatments T1 and T2, except for the disposition of the organelles, cytoskeleton arquiteture (actin and intermediate filaments), occurrence of autophagyc vacuoles and mielinic figures in the cytoplasm of TC. The two other embryos of treatment T3 showed intense morphologic modifications of their cells: TCs showed globular shape with few microvilli, or with their cytoplasm portions very thin depleted of microvilli; in the cytoplasm many vacuoles, phagosomes, lipid droplets, few organelles and cytoskeleton elements were observed. The PVS were enlarged, containing cell debris and extruded blastomeres; in the blastocoele, debris and cell fragments were also observed. In one treatment T3 embryo the spaces among the TCs were enlarged. The cells in degeneration process were more common in the trophoblast epithelium of the embryos of the treatment T3. All embryos from treatment T4 were lost during the electron microscopy technique procedure. The results shown in this paper suggest the occurrence of ultra-structural differences among the embryos from goats receiving high level of urea in the diet, mainly those related to increasing morphological structures related to the cell reorganization and/or degeneration.
