Título: Fluxo gênico recíproco entre cultivares de soja sob a teoria de genética de populações

Abstract: Conforme a literatura, não é comum que a taxa de hibridação natural em Glycine max ultrapasse o valor de 1%. Com a chegada das cultivares transgênicas, novas pesquisas foram realizadas no intuito de investigar a alogamia dessas cultivares. Para os produtores de sementes para comercialização é importante conhecer este aspecto das cultivares, pois pureza genética é a garantia de qualidade do seu produto. Pesquisas demonstram que o fato da cultivar ser modificada geneticamente não altera a expectativa existente de fecundação cruzada. Na consciência de que pureza genética é requerida para qualquer variedade, o objetivo deste trabalho foi avaliar a taxa de fecundação cruzada recíproca envolvendo duas cultivares de soja, uma convencional e outra GM. Anteriormente, buscou-se ajustar metodologias de bioensaios para a detecção de sementes de soja de cultivar GM, e refletir o fluxo gênico em soja sob a perspectiva de genética de populações. A partir do ajuste das metodologias dos bioensaios: i) germinação de sementes em substrato umedecido com o glifosato, e, ii) pré-embebição de sementes em solução do herbicida, seguida de teste de germinação, obteve-se protocolos eficientes para a detecção e quantificação de sementes de soja GM entre sementes de soja convencional. O método do umedecimento do substrato de germinação com solução do herbicida, a 0,03% do seu equivalente ácido, na proporção de 3 vezes o peso do papel seco foi escolhida para os testes de detecção de hibridação natural. Diante às perspectivas de genética de populações, concluiu-se que a chance de fixação de um alelo no pool gênico de uma população após o fluxo gênico é, entre outros fatores, função de sua relevância para a aptidão de seus portadores. Além disso, que a seleção contra um alelo dominante precisa de apenas uma geração para eliminá-lo do pool gênico da população e que o efeito diluição deve ser utilizado para a divulgação das taxas de hibridação natural encontradas em estudos desta natureza. No estudo de fecundação cruzada recíproca, os diferentes métodos para a verificação da hibridação natural, na verificação da cor da flor em campo, na verificação da cor do hipocótilo em casa de vegetação, na pulverização de plântulas com solução do herbicida em casas de vegetação, e, na germinação de sementes em contato com o herbicida em laboratório de análise de sementes, demonstraram que as taxas de fecundação cruzada populacionais foram 0,05, 0,10, 0,39 e 0,22 %, respectivamente. Com base nestes resultados, verificou-se que a taxa de hibridação natural recíproca não é a mesma em ambos os sentidos e que a aplicação do efeito diluição é indispensável para apresentar os resultados de estudos sobre o fluxo gênico, já que os valores populacionais são inferiores àqueles encontrados para as fileiras estudadas.
According to the literature, it isn't common that the natural hybridization rate in Glycine max exceeds the value 1%. With the arrival of transgenic cultivars, new researches has been done in order to investigate the allogamy of these cultivars. For seed producers for marketing is important to know this aspect of the cultivars, because genetic purity is the assurance of your product. Researches show that the fact of growing genetically modified to not change the existing expectation of cross-fertilization. In the knowledge that genetic purity is required for any variety, the objetctive was to assess the rate of reciprocal cross-fertilization involving two soybean cultivars, one conventional and one GM. Previously, we attempted to adjust methods of bioassay for the detection of soybean seeds of GM crops, and reflect the gene flow of soybean from the perspective of population genetics. From the adjustment of the methods of bioassays: i) seed germination in a substrate containing glyphosate, and ii) presoaking of seeds with the herbicide, followed by germination test, we obtained efficient protocols for detecting and quantification of GM soybean seeds of the soybean seeds. The method of wetting the substrate with germination herbicide solution, 0.03% of its acid equivalent at a ratio of 3 times the weight of dry paper was selected for the testing of natural hybridization. Faced with the prospect of population genetics, it was concluded that the chance of fixation of an allele in the gene pool of a population after gene flow is, among other factors, because of its relevance to the fitness of their bearers. In addition, the selection against a dominant allele needs only one generation to eliminate it from the gene pool of the population and that the dilution effect should be used for the dissemination of natural hybridization rates found in studies of this type. In the study of reciprocal cross-fertilization, different methods for the verification of natural hybridization, examining the color of the flower field, examining the color of the hypocotyls in a greenhouse, the spraying of seedlings with the herbicide in greenhouses, and the germination of seeds in contact with the herbicide in the laboratory seed analysis showed that the rates of outcrossing population were 0.05, 0.10, 0.39 and 0.22% respectively. Based on these results, it was found that the rate of natural hybridization reverse is not the same in both directions and that the application of the dilution effect is indispensable to present the results of studies on gene flow, since the population values are lower than those found in the rows studied.
