Título: Produção de ácidos orgânicos e tolerância de sorgo à toxicidade do alumínio
Production of organic acids and sorghum tolerance to aluminum toxicity

Abstract: Este estudo objetivou analisar os efeitos do alumínio (Al), em nível tóxico, sobre o crescimento de plantas de sorgo, os teores e a exsudação de ácidos orgânicos, e a atividade de enzimas ligadas à síntese e à degradação destes ácidos, em plantas de dois cultivares de sorgo, um sensível (BR007A) e outro tolerante (BR006R) ao Al. Os pesos da matéria seca das duas partes das plantas dos dois cultivares e o comprimento da maior raiz diminuíram com o aumento das concentrações de Al na solução, tendo o cultivar sensível sido mais severamente afetado que o tolerante. Os teores de Al, tanto nas raízes quanto na parte aérea, aumentaram com os aumentos nas concentrações de Al na solução nutritiva e o cultivar sensível acumulou mais Al nas duas partes da planta. O Al acumulou-se principalmente no ápice radicular (0-5 mm). Este segmento da raiz, além de apresentar maior teor de Al que os demais segmentos analisados, no cultivar sensível, apresentou cerca de 25,0% mais Al que o cultivar tolerante. Os teores de P, K, Ca e Mg diminuíram com o aumento das concentrações de Al na solução nutritiva, nas duas partes da planta, mas os cultivares não diferiram entre si, exceto quanto ao teor de Ca na parte aérea. A aplicação de ácido málico resultou em amenização do efeito inibitório do Al sobre o alongamento radicular, mas não houve eliminação completa da toxicidade do Al, mesmo na concentração mais alta. Os teores dos principais ácidos orgânicos, encontrados nas raízes e na parte aérea, aumentaram com a exposição das plantas ao Al. Os dois ácidos orgânicos mais abundantes em sorgo e, provavelmente, os mais importantes do ponto de vista da tolerância ao Al foram os ácidos málico e t-aconítico. Dentre todos os ácidos orgânicos, o ácido málico foi aquele que, na presença de Al, sofreu o maior aumento absoluto no seu teor, sempre mais pronunciado no cultivar tolerante, sugerindo um papel importante deste ácido no mecanismo de tolerância das plantas à toxicidade de Al. O Al promoveu aumento no teor do ácido cítrico transportado na seiva xilemática, principalmente no cultivar tolerante, no qual foi cerca de 26,0% maior que no cultivar sensível. O Al, de modo geral, influenciou a atividade das enzimas estudadas, no sentido de estimular a síntese e, ou, inibir a degradação do ácido málico. Nos casos dos ácidos cítrico e t-aconítico, isto nem sempre aconteceu. No cultivar tolerante, as enzimas de síntese dos ácidos orgânicos mostraram-se mais sensíveis ao estímulo, enquanto as enzimas de degradação foram mais sensíveis ao efeito inibitório do Al tóxico. As enzimas importantes no controle da produção de ácidos orgânicos e determinantes do comportamento diferencial dos cultivares de sorgo frente ao estresse de Al parecem ser a carboxilase do fosfoenolpiruvato e, principalmente, a fumarase.
This work aimed to study effects of aluminum (Al) at a toxic level, on sorghum growth, the contents and exudation of the organic acids as well as the activities of the enzymes related to synthesis and degradation of those acids in sorghum cultivars, one of them being sensible (BR007A) and another tolerant (BR006R). The dry matter weight in both parts of the plants and the longest root length reduced with the Al concentration increases, being the sensible cultivar more severely affected than the tolerant one. The Al contents either in the roots and the aerial part increased with the increasing Al concentrations in the nutritive solution while the sensible cultivar cumulated more Al in both plant parts; Al cumulated mainly at root apex (0-5 mm). Besides presenting a higher Al content than the other segments in the sensible cultivar, the root apex showed about 25% more Al than in the tolerant cultivar. The P, K, Ca and Mg contents diminished with the increase in Al concentrations in both plant parts, although the cultivars did not differ from each other, except for Ca content in the aerial part. Application of the malic acid caused the Al inhibitory effects on the elongation of root to soften, xibut Al toxicity was not completely eliminated even at a higher concentration. The contents of the main organic acids found in the roots and the aerial part increased with plant exposure to Al. The malic and t-aconitic acids showed to be mostly abundant and probably the most important under the tolerance point of view. In Al presence the malic acid suffered the greatest absolute increase always more accentuated in the tolerant cultivar suggesting its important role on plant tolerance mechanism to Al toxicity. Al promoted an increase in the content of the citric acid transported in the xylem sap mainly in the tolerant cultivar which was 26.0% higher than the sensible cultivar. In general the Al influenced the enzyme activities by stimulating the synthesis and, or, inhibiting the malic acid degradation, which did not always occur for the citric and t-aconitic acids. In the tolerant cultivar the enzymes related to the organic acid synthesis showed to be more sensible to stimulus, whereas the ones of degradation were more sensible to the inhibitory effects from toxic Al. Those enzymes which are important in controlling the production of the organic acids and in determining the differential behavior of sorghum cultivars under Al stress appear to be a phosphoenolpyruvate carboxylase and mainly the fumarase.
