Abstract: A exposição ao ar desencadeia respostas de estresse e estresse oxidativo nos peixes. Quando a produção de espécies reativas de oxigênio (ROS) e espécies reativas de nitrogênio (RNS) supera a sua neutralização pelas defesas enzimáticas e não enzimáticas ocorrem danos em lipídeos, proteínas e ácidos nucleicos. Dentre os antioxidantes com potencial para reduzir esses danos destacam-se a vitamina C, antioxidante hidrossolúvel, e o óleo orégano, antioxidante lipossolúvel. A combinação de antioxidantes hidrossolúveis e lipossolúveis pode promover efeitos sinérgicos na proteção contra danos oxidativos por atuarem em diferentes partes das células. Assim, objetivamos avaliar a interação entre vitamina C e óleo de orégano na capacidade antioxidante de Astyanax aff. bimaculatus submetidos à exposição ao ar. Utilizamos esquema fatorial 5x2 em delineamento inteiramente casualizado, com cinco níveis de inclusão de vitamina C (0,0; 40,0; 80,0; 120,0 e 160,0 mg kg -1 ) e dois níveis de inclusão de óleo de orégano na dieta (0,0 e 1000,0 mg kg -1 ) com três repetições por tratamento. Foram avaliadas a glicose sanguínea nos peixes expostos ao ar, a atividade das enzimas antioxidantes superóxido dismutase (SOD), catalase (CAT), glutationa S-transferase (GST) e os níveis de malondialdéido (MDA) e de proteína carbonilada (PC) nas brânquias e no fígado dos peixes não expostos e expostos ao ar. Nos peixes não expostos ao ar, não houve interação significativa entre a suplementação de vitamina C e óleo de orégano sobre as variáveis de estresse oxidativo nas brânquias e no fígado. Nesses peixes, a vitamina C reduziu a atividade da SOD e o orégano reduziu os níveis de PC nas brânquias. No fígado, a vitamina C reduziu a atividade da CAT e os níveis de MDA, e o orégano reduziu os níveis de MDA e aumentou a atividade da GST. Nas brânquias dos peixes expostos ao ar, houve interação significativa entre a vitamina C e o óleo de orégano sobre os níveis de MDA. A vitamina C reduziu os níveis de MDA apenas na presença do orégano. No fígado dos peixes expostos ao ar, houve interação significativa entre a vitamina C e o óleo de orégano sobre a atividade da GST. Na ausência do óleo de orégano houve efeito quadrático da vitamina C sobre a atividade da GST, com valor estimado que maximiza essa variável de 98,6 mg kg -1 de vitamina C. Já na presença do orégano, houve efeito cúbico da vitamina C sobre a atividade da GST. No fígado dos peixes expostos ao ar, houve efeito quadrático da vitamina C sobre a atividade da SOD e os níveis de PC, com valores estimados que minimizam essas variáveis de 135,5 e 91,3 mg kg -1 de vitamina C, respectivamente. A vitamina C também reduziu a atividade da CAT e os níveis de MDA no fígado dos peixes expostos ao ar. A vitamina C reduziu os níveis de glicose sanguínea nos peixes expostos ao ar. Portanto, a vitamina C e o orégano protegeram os peixes contra o estresse oxidativo tanto antes quanto após a exposição ao ar. O efeito sinérgico observado após a exposição ao ar indica a importância da suplementação de antioxidantes hidrossolúveis e lipossolúveis em dietas para peixes em cultivo. As maiores concentrações de vitamina C avaliadas promoveram maior proteção contra os danos oxidativos nas brânquias e no fígado de A. aff bimaculatus. Assim, recomendamos a utilização de 160,0 mg kg -1 de vitamina C. O óleo de orégano apresentou efeito complementar ao da vitamina C na proteção contra os danos oxidativo e, portanto, recomendamos a utilização de 1000,0 mg kg -1 de óleo de orégano associada à suplementação de vitamina C. Em condições de desafio, a combinação dos dois antioxidantes é ainda mais importante devido ao efeito sinérgico desses antioxidantes.
Air exposure causes stress responses and oxidative stress in fish. When the production of reactive oxygen species (ROS) and reactive nitrogen species (RNS) exceeds their neutralization by enzymatic and non-enzymatic defenses, damage to lipids, proteins and nucleic acids occurs. Among the antioxidants with potential to reduce these damages stand out vitamin C, water soluble antioxidant, and oregano oil, liposoluble antioxidant. The combination of water-soluble and liposoluble antioxidants may promotes synergistic effects in protecting against oxidative damage because they act on different parts of the cells. Thus, we aimed to evaluate the interaction between vitamin C and oregano oil in the antioxidant capacity of Astyanax aff. bimaculatus exposed to air. We used factorial scheme 5x2 in a completely randomized design, with five inclusion levels of vitamin C (0.0, 40.0, 80.0, 120.0 and 160.0 mg kg -1 ) and two inclusion levels of oregano oil in the diet (0.0 and 1000.0 mg kg -1 ) with three replicates per treatment. We evaluated the blood glucose in fish air exposed air, the activity of superoxide dismutase (SOD) catalase (CAT), glutathione S-transferase (GST) and malondialdide (MDA) and carbonylated protein (PC) levels in the gills and liver of fish not air exposed and air exposed. In the fish not air exposed, there was no significant interaction between supplementation of vitamin C and oregano oil on the variables of oxidative stress in the gills and liver. In these fish, vitamin C reduced SOD activity and oregano reduced PC levels in the gills. In the liver, vitamin C reduced CAT activity and MDA levels, and oregano reduced MDA levels and increased GST activity. In the fish gills air exposed, there was significant interaction between vitamin C and oregano oil on MDA levels. Vitamin C reduced MDA levels only in presence of oregano oil. In the liver of fish air exposed, there was significant interaction between vitamin C and oregano oil on GST activity. In absence of oregano oil, there was quadratic effect of vitamin C on GST activity, with estimated values that maximize this variable equal to 98.6 mg kg -1 of vitamin C. In presence of oregano, there was cubic effect of vitamin C on GST activity. In the liver of fish air exposed, there was quadratic effect of vitamin C on SOD activity and PC levels, with estimated values that minimize this variable equal to 135,5 and 91,3 mg kg -1 of vitamin C, respectively. Vitamin C also reduced CAT activity and MDA levels in the liver of fish air exposed. Vitamin C reduced blood glucose levels in the fish air exposed. Therefore, vitamin C and oregano protected fish from oxidative stress before and after exposure to air. The synergistic effect observed after air exposure indicates the importance of supplementation of water soluble and liposoluble antioxidants in diets for farmed fish. The highest concentrations of vitamin C evaluated promoted greater protection against oxidative damage in the gills and liver of A. aff bimaculatus. Thus, we recommend the use of 160.0 mg kg -1 of vitamin C. Oregano oil showed a complementary effect to vitamin C in protection against oxidative damage, therefore we recommend the use of 1000.0 mg kg -1 of oregano oil associated with vitamin C supplementation. Under challenge situations, the combination of these two antioxidants is even more important because of the synergistic effect of these antioxidants.
