Effect of water déficit in the photosynthetic metabolism of Coffea canephora cv. Conilon clones with differential drought tolerance

Abstract: Os efeitos do déficit hídrico no metabolismo dos carboidratos, foram investigados em quatro clones de Coffea canephora Pierre cv. Conilon, com tolerância diferencial à seca. Quando o potencial hídrico foliar na antemanhã (Ψ w ) decresceu para valores em torno de -2 e -3 MPa, estresse hídrico moderado e severo, respectivamente, houve redução na fotossíntese líquida, provocada principalmente pelo fechamento dos estômatos, visto que, não houve fotoinibição nem alteração nos teores de clorofilas a e b. Sob estresse hídrico severo houve aumento na concentração foliar de sacarose nos clones tolerantes à seca (120 e 14). Porém, com exceção do clone 120, todos apresentaram redução na atividade da sintase da sacarose-fosfato, que ficou mais bem evidenciada no ensaio seletivo, principalmente sob estresse hídrico severo. O clone 120 foi o único também em que foi verificado estímulo da fosfatase da frutose-1,6-bisfosfato, sob estresse hídrico. Uma tendência geral de estímulo na atividade da invertase ácida foi verificada, principalmente sob estresse hídrico severo, sendo que somente no clone 14, nesse nível de estresse houve aumento na atividade da invertase alcalina. Entretanto, não se verificaram grandes alterações nos níveis de hexoses. A sintase da sacarose não teve sua atividade alterada. Tanto sob estresse hídrico moderado quanto severo, verificou-se redução no conteúdo de amido, em todos os clones. No entanto, apenas nos clones 14 e 46, houve redução na atividade da pirofosforilase da ADP-glicose. Os clones 120 e 109A também apresentaram uma tendência de aumento na concentração de aminoácidos, que ficou mais bem evidenciada sob estresse hídrico severo. A atividade da cinase da frutose-6-fosfato, não foi alterada em resposta ao déficit hídrico, sugerindo que não houve alteração no fluxo glicolítico. Apesar da complexidade apresentada com relação a atividade das enzimas de metabolismo dos carboidratos, esses resultados permitem sugerir que os clones mais tolerantes acumulam mais açúcares solúveis sob estresse hídrico, indicando maior eficiência fotossintética.
The effects of water deficit on carbohydrate metabolism were investigated in four clones of Coffea canephora Pierre cv. Conilon, with differential drought tolerance. When the leaf predawn water potential (Ψ w ) decreased for values around -2 and -3 MPa, moderate and severe water stress, respectively, there was reduction in the net photosynthesis, provoked mainly by the closing of the stomata, because, there probably were not photoinhibition nor alteration in the levels of chlorophylls a and b. Under severe water stress there was increase in the leaf concentration of sucrose in the drought tolerant clones (120 and 14). However, except for the clone 120, all presented reduction in the activity of the sucrose-phosphate sintase, that was better evidenced in the selective assay, under severe water stress. The clone 120 was also the only in that increase in the activity of fructose-1,6-bisphosphate phosphatase was verified under water stress. A general tendency of increase in the activity of acid invertase was verified, mainly under severe water stress, and only in the clone 14, in that stress level, there was increase in the activity of the alkaline invertase. However, great alterations were not verified in the hexoses levels. There was not alteration in the activity of sucrose sintase. Under moderate and severe water stress, reduction was verified in the levels of starch, in all the clones. However, only in the clones 14 and 46, there was reduction in the activity of the ADP-glucose pirophosphorilase. The clones 120 and 109A also presented a tendency to increase the concentration of amino acids, that was better evidenced under severe water stress. The activity of fructose-6-phosphate kinase was not altered in response to water déficit, suggesting that there was not alteration in the glicolitic path. In spite of the complexity presented with relationship the activity of the enzymes of carbohydrate metabolism, those results allow us to suggest that the most tolerant clones accumulate more soluble sugars under water stress, indicating better photosynthetic efficiency.
